      • The effect of alpha lipoic acid in a porcine in-stent restenosis model

        <P><B>Summary</B></P><P><B>Background</B></P><P>The aim of this study was to investigate the effect of alpha lipoic acid (α-LA) on a porcine in-stent restenosis (ISR) model.</P><P><B>Methods</B></P><P>In protocol 1, porcine vascular smooth muscle cells (PVSMC) were stimulated by granulocyte-colony stimulating factor (G-CSF) in the presence or absence of α-LA. MTT (3-[4,5-dimethylthiazole-2-yl] 2,5-diphenyl tetrazolium bromide) assay and western blotting were used to determine the cell growth inhibitory rate and anti-inflammatory effect associated with nuclear factor-κb (NF-κb) and extracellular signal-regulated kinase (ERK). In protocol 2, 28 days after balloon overdilation injuries, 24 bare metal stents were placed in coronary artery of 12 pigs. The pigs were randomly divided to receive control diet with or without α-LA (100mg/kg). In protocol 3, 8 control stents and 8 α-LA coated stents were randomly implanted in 2 coronary arteries of 8 pigs and follow-up coronary angiogram and histopathologic assessment were performed 4 weeks after stenting.</P><P><B>Results</B></P><P>Protocol 1. The proliferation of PVSMC was inhibited and protein expression of NF-κb and ERK were attenuated by α-LA pretreatment. Protocol 2<I>.</I> On histopathologic analysis, the neointimal area (4.0±1.0mm<SUP>2</SUP> vs. 1.5±0.7mm<SUP>2</SUP>, <I>p</I><0.001) and histopathologic area of stenosis (66.7±10.7% vs. 24.2±9.7%, <I>p</I><0.001) were reduced in the α-LA feeding group compared to controls. Protocol 3. On histopathologic analysis, the neointimal area (3.9±0.8mm<SUP>2</SUP> vs. 1.0±0.4mm<SUP>2</SUP>, <I>p</I><0.001), and the histopathologic area of stenosis (67.1±8.8% vs. 17.4±10.0%, <I>p</I><0.001) were reduced in the α-LA coated stent group compared to the control stent group.</P><P><B>Conclusions</B></P><P>α-LA feeding and α-LA coated stents inhibit neointimal hyperplasia in porcine ISR, possibly through inhibiting the activation of NF-κb pathway and proliferation of PVSMC.</P>

      • VO<sub>2</sub>의 금속-절연체 전이 현상에 대한 광학적 특성 변화 측정

        VO<sub>2</sub>는 상온 근처에서 금속-절연체 전이 현상일 일어나는 것을 잘 알려진 물질이다. 금속과 절연체 전이 특성으로 인하여 전기적인 특성뿐만 아니라 광 특성에도 변화가 생긴다. 본 연구에서는 연구실의 레이저를 이용하여 간단하게 VO<sub>2</sub>의 광 특성 변화를 측정할 수 있는 실험 방법을 보여준다. 또한 인 시투 기법으로 온도를 변화시키면서 광 특성과 전기적 특성의 변화를 동시에 측정하여 비교할 수 있도록 실험 장치를 설치하고 측정을 진행하였다. 이런 측정 기법은 다른 연구에도 널리 응용될 수 있을 것이다. VO<sub>2</sub> is a well-known metal-insulator transition (MIT) material with Tc of near room temperature. MIT causes a change of electrical properties and also optical properties. We demonstrated that optical property of VO<sub>2</sub> can be simply measured using a laser beam. The experiment was setup that optical properties were simultaneously measured with electrical properties as a function of temperature at in-situ condition. Therefore, the optical properties were directly compared to the electrical properties. This measurement technique can be widely applicable to other studies.

      • 연계적 공학해석 교육을 위한 워크플로우 기반 도구 개발 및 활용 시나리오 제안

        공학현장에서 소프트웨어의 활용성 및 중요성은 날로 증대되고 있으며, 따라서 공학설계 단계에서 해석 소프트웨어의 활용 및 이해는 공학도의 핵심역량이 되었다. 본 연구에서는 다수의 해석영역 혹은 해석분야를 가진 현장의 복합적 시스템 대상 공학교육을 위한 새로운 전산 환경을 개발하고 활용 시나리오를 제시하였다. KISTI에서는 개별 해석 연구자들이 개발한 in-house 소프트웨어들을 탑재할 수 있는 계산 플랫폼의 하위모듈로서 워크플로우 실행환경을 개발하였으며, 이를 활용하여 유동, 구조, 최적설계 등의 다양한 단위 소프트웨어들을 서로 연계실행할 수 있게 되었다. 그래서 본 연구에서는 이러한 연계해석 실행환경을 공학해석 및 공학설계 융합형 교육에 활용할 수 있는 활용 시나리오를 제시하였고, 이를 적절히 적용하면 실제 현장에서의 공학해석 과정에 대한 큰 그림에 대한 이해 속에서 공학도 각자의 해석분야에 대한 전문성을 높일 수 있을 것으로 기대한다. The importance of software in the engineering field is increasing day by day, so the utilization and understanding of the simulation software in the engineering design stage has become the core competence of the engineer. This study presents a new software education method for multiple systems in the field of mechanical engineering using a workflow execution environment as a sub-module of a computational platform (called EDISON platform) that can incorporate in-house software tools-developed by many simulation tool developers. It can execute not only individual software tools such as fluid flow, structure, optimal design, but also conduct connective executions of multiple software tools. Based on this simulation environment, a methodology was proposed that can be applied to convergence types of engineering educations. By properly using this methodology, it is expected that beginning engineers could encourage their specialties understanding the big pictures of the analysis processes.

        Background: A photoaged decollete skin may cause significant cosmetic disfigurement in the middle-aged. The photoaged area shows mottled red-brown dyschromia, telangiectasia, and fine wrinkles. Objectives: To evaluate the effectiveness of a combined treatment with 1,064 nm Neodymium:Yttrium Aluminum-Garnet laser (Nd:YAG) and high intensity focused ultrasonography (HIFU) for a photoaged decollete skin. Methods: Eight subjects, all women, were enrolled. All subjects were received 2 times of HIFU treatment at 0-week and 8-week, and 6 times of 1,064 nm Nd:YAG laser treatment at 2-, 4-, 6-, 10-, 12-, and 14-week. The last follow-up was at 16-week. The clinical outcome was evaluated with digital photograph, spectrophotometry and questionnaire. Results: Most of subjects showed improvement of erythema index (14.4 to 12.4 at 2- and 16-week) and melanin index (36.4 to 33.0 at 2- and 16-week), not statistically significant (p=0.093 and p=0.069, respectively). The subjects, showing greater than moderate response (> 25% improvement) in a physician assessment, were 87.5%, 87.5%, and 62.5% in dyschromia, telangiectasia, and fine wrinkles, respectively. Also, in the survey, 75.0% and 87.5% of subjects replied that they experienced greater than moderate response in pigmentation and wrinkles. Conclusion: A combined treatment with 1,064 nm Nd:YAG and HIFU may be an effective therapeutic option for a photoaged decollete skin in Korean female adults.

        Differential Gene Expression in GPR40-Overexpressing Pancreatic β-cells Treated with Linoleic Acid

        “G protein-coupled receptor 40” (GPR40), a receptor for long-chain fatty acids, mediates the stimulation of glucose-induced insulin secretion. We examined the profiles of differential gene expression in GPR40-activated cells treated with linoleic acid, and finally predicted the integral pathways of the cellular mechanism of GPR40-mediated insulinotropic effects. After constructing a GPR40-overexpressing stable cell line (RIN-40) from the rat pancreatic β-cell line RIN-5f, we determined the gene expression profiles of RIN-5f and RIN-40. In total, 1004 genes, the expression of which was altered at least twofold, were selected in RIN-5f versus RIN-40. Moreover, the differential genetic profiles were investigated in RIN-40 cells treated with 30 μM linoleic acid, which resulted in selection of 93 genes in RIN-40 versus RIN-40 treated with linoleic acid. Based on the Kyoto Encyclopedia of Genes and Genomes Pathway (KEGG, http://www.genome.jp/kegg/), sets of genes induced differentially by treatment with linoleic acid in RIN-40 cells were found to be related to mitogen-activated protein (MAP) kinase- and neuroactive ligand-receptor interaction pathways. A gene ontology (GO) study revealed that more than 30% of the genes were associated with signal transduction and cell proliferation. Thus, this study elucidated a gene expression pattern relevant to the signal pathways that are regulated by GPR40 activation during the acute period. Together, these findings increase our mechanistic understanding of endogenous molecules associated with GPR40 function, and provide information useful for identification of a target for the management of type 2 diabetes mellitus.

        Expression Patterns of Germ Cell-specific Phosducin-like 2 during Testicular and Ovarian Development in Chickens

        Phosducin (PDC) is a photoreceptor cell-specific protein that is phosphorylated by cyclic nucleotide-dependent protein kinase. PDC and PDC-like proteins (PDCL, PDCL2, and PDCL3) are members of a conserved family of small thioredoxin-like proteins that modulate the ${\beta}$- and ${\gamma}$-subunits of G-proteins. In mammals, Pdc, Pdcl, and Pdcl3 genes show ubiquitous expression; however, Pdcl2 gene expression is limited to the testis and ovary. The aim of the present study was to examine the expression patterns of chicken Pdcl2 (cPdcl2) during testicular and ovarian development. Protein sequence comparisons performed using the CLUSTAL X program revealed that the amino acid sequences and potential phosphorylation sites of cPDCL2 and mammalian PDCL2 proteins were highly conserved. Quantitative real-time PCR analysis revealed that cPdcl2 was differentially expressed in the testis and ovary. Specifically, cPdcl2 expression was detected at low levels in the ovary at all time points. In the testis, cPdcl2 expression was detected at low levels until 5 weeks of age. At 8 weeks of age, however, cPdcl2 showed increased expression levels in the testis. Using in situ hybridization, we detected high levels of cPdcl2 expression in the testis, particularly in the spermatocytes and round spermatids. In summary, our data describe expression patterns of germ cell-specific Pdcl2 during testicular and ovarian development in chickens.

        일미찰옥수수의 유묘기 저온에 따른 생육특성 변화

        To determine the damages to waxy corn caused by low temperature weather, we investigated the relationship between the temperature and duration of low temperature treatment and the changes in growth characteristics during the recovery period after the treatments in different growth periods. Growth inhibition started in the low temperature group treated at temperatures below 5℃ for three days. The inhibition ratio (IR) was more than 22% or more in all sample groups. As the treatment duration increased and the Temperature decreased, the growth was more greatly inhibited than that in the control. The IR was the highest at 27% or more in the 2nd leaf stage group treated at below 5℃ for 5 days. The IR was in the order of 2nd leaf stage > 1st leaf stage > coleoptile. The IR during recovery was the highest in the 2nd leaf stage group treated at -3℃ degree for 7 days, nd the values were 82% and 98% for NDVI and Fv/Fm, respectively. Especially, all groups treated at -3℃ showed either no changes or decreases in the growth characteristics. As a result, growth inhibition increased as the temperature decreased, and as the duration of the low temperature increased. The degree of damage was in the order of 2nd leaf stage > 1st leaf stage > coleoptile. All early seedlings stopped growing and withered when exposed to temperatures at or below -3℃ for 3 days or more.

        Expression pattern of meiosis associated SYCP family members during germline development in chickens.

        <P>Synaptonemal complexes (SCs) are associated with synapsis of homologous chromosomes, chiasmata distribution, recombination and segregation of chromosomes during the extended prophase of meiosis I. Three isoforms of SC proteins, SYCP1, SYCP2 and SYCP3, were identified as the structural proteins of SCs, and may be involved in the assembly and disassembly of SCs. The aim of this present study is to determine the pattern of expression of chicken homologues of SYCP family members during ovarian and testicular development. Protein sequence analysis using CLUSTAL X revealed that the sequences and potential phosphorylation sites of chicken SYCP family proteins were highly conserved with mammalian homologues of SYCP family proteins. Quantitative real-time-PCR and in situ hybridisation analysis revealed that chicken SYCP family members were differentially expressed during ovarian and testicular development. During ovarian development, all chicken SYCP family members were detected in primordial germ cells (PGCs) until embryonic day (E) 8.0; the expression continued in proliferating pre-meiotic oogonia until E15.5 and was upregulated in meiotic prophase I oocytes until hatching. After hatching, all chicken SYCP family members were detected at a low level until 24-weeks-old. During testicular development, all chicken SYCP family members were detected in PGCs until E13.0; the expression continued in pro-spermatogonia and proliferating spermatogonia for up to 8 weeks, and was upregulated in meiotic prophase I spermatocytes in adults. Our data demonstrate the expression pattern of meiosis associated SYCP family members during ovarian and testicular development in chickens.</P>

      • Trichoscopic findings in androgenetic alopecia and alopecia areata

        Background: The use of trichoscopy can ease the division of various hair loss disease by magnifying the scalp and structure of hair. Objectives: Our purpose is to measure the frequency of representative trichoscopic findings in Alopecia areata(AA) and Androgenetic alopecia(AGA) patients of Korea and the frequency of various trichoscopic findings depending on the severity based on BASP classification. Methods: A retrospective cohort review of total 87 patients with AGA(n=57) and AA(n=30) between 2014-2015 at Kangbuk Samsung Hospital. Three dermatologists judged the trichoscopic findings (hair shaft, hair follicle opening, perifollicular epidermis) in 87 patients. Results: In this study, vellus hair is observed 21/30 cases (70%) in AA patients and 20/57 cases (33%) in AGA patients. In AGA patients, as severity of hair loss increases, the portion of patients with vellus hair, thickness heterogeneity, honeycomb pigmentation also increases (p < .05). Also, in AA patients, as severity increases, the portion of patients with black dot and perifollicular sign increases (p < .05). Conclusion: Trichoscopy is very useful in diagnosis between AA and AGA if it is difficult to diagnosis by naked eyes. Also hair loss disease’s severity can be assessed by trichoscopy.
