        A Way to Utilize Movies Through Exceptional Examples of English Comparatives and Imperatives

        Recent mechanical improvement in classroom facilities allows teachers to use movies as teaching material for students studying English as a foreign language. Movies help students to understand the rules of English linguistics such as grammar and sound patterns by providing them with specific real-life utterances. Unlike general textbooks, movies also provide a wide variety of interesting English examples that appear to go against the general rules. In this paper, we first divide English examples found in movies into two types: supporting examples (SptEx) that exemplify the rules of English; and exceptional examples (ExpEx) that superficially break the rules. Then, by showing a way to utilize movies via both the SptEx and the ExpEx, this paper argues that a reciprocally-related teaching method helps teachers to deepen students’ understanding of English and even broaden their linguistic views. Furthermore, it is demonstrated that this method can be applied to various fields of English linguistics such as syntax and morphology, and also to a number of English sentence types. Among others, this paper focuses on comparative and imperative sentences. In addition, several potential issues related to collecting ExpEx from movies and build up a database of examples are pointed out.

        Learning English via Cross-Cutting Observations: With Tag Questions and Movies

        This paper proposes a method to help ESL learners improve their knowledge of English grammar and broaden their understanding of the English language. One important key feature of the method is careful observation of the independently-taught rules of English grammar from one specific sentence type, which I call “cross-cutting observations”. It encourages ESL learners to see the close relation between aspects of English language learning and the application of different grammar rules, such as the imperative sentence construction, pronoun usage, adverb usage, etc., by highlighting in particular the properties of tag questions in English (e.g., Tom likes sushi, doesn’t he?). Furthermore, this paper provides suggestions as to the potential use of authentic English examples in movies (and also TV dramas) as a rich source of learning and teaching material which typical English textbooks for ESL learners may lack. The paper concludes by encouraging ESL learners desiring to widen their understanding of English forms and usage (specifically as seen from the aspect of tag questions) by tapping into authentic and natural English in movies. Overall, this paper is an attempt to bridge English education, English linguistics and movies as an effective means to engage ESL learners.

      • Expression mechanism of tryptophan hydroxylase 1 in mouse islets during pregnancy.

        <P>Serotonin signaling plays key roles in augmentation of pancreatic β-cell function during pregnancy. Increased expression of tryptophan hydroxylase 1 (Tph1), a rate-limiting enzyme for serotonin synthesis by lactogenic hormones, is involved in this phenomenon. To investigate its mechanisms, we here performed 5'-RACE and identified β-cell-specific transcription initiation sites for Tph1. Prolactin enhanced the expression of mRNA containing these exons; however, reporter gene plasmids containing the proximal 5'-flanking region of these exons did not show prolactin responsiveness in MIN6 cells. Prolactin-induced Tph1 expression was inhibited by a Jak2 inhibitor and was partially inhibited by an MEK1/2 or PI3K inhibitor. Therefore, we analyzed interferon γ-activated sequences (GAS) and found GAS-A about 9-kbp upstream of the transcription start site. The reporter gene plasmid containing the GAS-A region linked to a heterologous promoter showed increased promoter activity by prolactin, which was inhibited by the forced expression of a dominant-negative mutant form of Stat5A and a Jak2 inhibitor. Chromatin immunoprecipitation analysis showed that prolactin treatment augmented Stat5 binding to the GAS-A region in MIN6 cells, as well as in isolated mouse islets, and that Stat5 recognized the GAS-A region in pregnant mouse islets. In addition, the transactivation activity of Stat5 was enhanced by prolactin through the Erk and PI3K pathways in MIN6 cells. Finally, serotonin expression was attenuated in islets of β-cell-specific Stat5-deficient mice compared with that of control littermates during pregnancy. Our findings suggest that prolactin-induced Tph1 expression is mediated by the activation of Jak2/Stat5, Erk, and PI3K pathways in β cells.</P>

      • Identification of aryl hydrocarbon receptor signaling pathways altered in TCDD-treated red seabream embryos by transcriptome analysis

        <P>2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) induces a broad spectrum of toxic effects including craniofacial malformation and neural damage in fish embryos. These effects are mainly mediated by the aryl hydrocarbon receptor (AHR). However, the mode of action between TCDD-induced AHR activation and adverse outcomes is not yet understood. To provide a comprehensive picture of the AHR signaling pathway in fish embryos exposed to TCDD, red seabream (Pagrus major) embryos were treated with graded concentrations of TCDD (0.3-37 nM) in seawater, or with a mixture of TCDD and 500 nM CH223191, an AHR-specific antagonist. The transcriptome of red seabream embryos was analyzed using a custom-made microarray with 6000 probes specifically prepared for this species. A Jonckheere-Terpstra test was performed to screen for genes that demonstrated altered mRNA expression levels following TCDD exposure. The signals of 1217 genes (as human homologs) were significantly altered in a TCDD concentration-dependent manner (q-value < 0.2). Notably, the TCDD-induced alteration in mRNA expression was alleviated by co-exposure to CH223191, suggesting that the mRNA expression level of these genes was regulated by AHR. To identify TCDD-activated pathways, the microarray data were further subjected to gene set enrichment analysis (GSEA) and functional protein-protein interaction (PPI) network analysis. GSEA demonstrated that the effects of TCDD on sets of genes involved calcium, mitogen-activated protein kinase (MAPK), actin cytoskeleton, chemokine, T cell receptor, melanoma, vascular endothelial growth factor (VEGF), axon guidance, and renal cell carcinoma signaling pathways. These results suggest the hypotheses that TCDD induces immunosuppression via the calcium, MAPK, chemokine, and T cell receptor signaling pathways, neurotoxicity via VEGF signaling, and axon guidance alterations and teratogenicity via the dysregulation of the actin cytoskeleton and melanoma and renal cell carcinoma signaling pathways. Furthermore, the PPI network analysis indicated that the adverse outcome pathways of TCDD in the embryos might be propagated through several hub genes such as cell division control protein 42, phosphoinositide-3-kinase regulatory subunit 1, and guanine nucleotide binding proteins.         Kyphosis Progression after Balloon Kyphoplasty Compared with Conservative Treatment

        Study Design: Retrospective cohort study (level of evidence: 4). Purpose: To demonstrate the effects of balloon kyphoplasty (BKP) on prevention of kyphosis progression. Overview of Literature: Kyphoplasty can correct local kyphosis (fractured vertebra), but its efficacy is attenuated by adjacent fracture and/or disc height reduction with endplate damage. With these attenuating factors incorporated, a comparison between BKP and conservative treatment is necessary to verify the effect of BKP on kyphosis correction. Methods: This study retrospectively analyzed 129 patients treated conservatively and 95 treated with BKP in our institution. The rate of new adjacent fracture occurrence was determined using Kaplan–Meier analysis. We examined the progression of local kyphosis in patients who underwent lateral X-ray after 1 year. Local kyphosis was measured as an angle between higher end plate of the vertebra above and lower end plate of the vertebra below the fractured vertebra. The comparison included 45 patients treated conservatively and 58 treated with BKP. Results: The incidence of new adjacent fracture at 1 year was 7.3% in the conservative treatment group and 23.2% in the BKP group (p<0.001), while the progression of local kyphosis at 1 year was 5.7°±4.7° and 3.2°±4.6°, respectively (p=0.01). Relative to conservative treatment, local kyphosis did not progress after BKP, despite the higher incidence of adjacent fracture. Local kyphosis progressed in cases with adjacent fracture compared with those without adjacent fracture (6.0°±4.3° vs. 2.1°±4.3°, p=0.003), and fractured vertebral instability was found to be a risk factor for adjacent fracture in BKP. Conclusions: Compared with the conservative treatment, BKP suppressed the progression of local kyphosis. However, an adjacent fracture attenuated the BKP correction and was more frequently seen in patients with unstable vertebral fractures.
