        흰쥐 해마박편에서 acetylcholine이 gamma-aminobutyric acid 유리에 미치는 영향에 관한 연구

        Present study was performed to clarify the effect of acetylcholine on the release of gamma-aminobutyric acid(GABA) employing hippocampal slices. Hippocampal slices (300∼400㎛ thick) were prepared by the method of Kim et al(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. Pre-equilibrated slices were incubated in fresh KBM and then potassium(50mM)-containing KBM for 5 min period. Basal and potassium-induced release of GABA were determined from recovered medium by HPLC. After 30min resting period, in the presence of physostigmine(20μM) slices were reincubated in acetylcholine-containing KBM and acetylcholine plus potassium-containing medium consecutively for 5min period each to investigate the effect of acetylcholine on basal or potassium-induced GABA release from hippocampal slices. The observed results were as follows: 1. The release of GABA induced by the first and second 5 min-exposure of 50mM potassium was 107.3±8.2 nmol and 90.6±3.2nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 4.6 and 4.6-fold increase respectively. 2. Physostigmine(20μM) had no significant effect on the spontaneous release of GABA. 3. Acetylcholine(10-1000μM) increased spontaneous and potassium-induced GABA release in a dose-dependent manner.

        흰쥐 해마박편에서 veratrine과 고농도 포타슘자극시 칼슘이온이 gamma-aminobutyric acid 유리에 미치는 영향에 관한 연구 : A role of calcium

        Present study was performed to clarify the effect of calcium on the release of gamma-aminobutyric acid (GABA) employing hippocampal slices. Hippocampal slices(300-400㎛ thick) were prepared by the method of Kim et al(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. In case of veratrine-induced GABA release, pre-equilibrated slices were incubated in fresh KBM and then veratrine (25μM)-containing KBM for 10min period in the presence or absence of 2.5mM Ca^2+. In case of potassium-induced GABA relaese, pre-equilibrated slices were incubated in fresh KBM and then potassium(50mM)-containing KBM for 5min period in the presence or absence of 2.5mM Ca^2+. Basal and veratrine and potassium-induced release of GABA was determined from recovered medium by HPLC. The observed results were as follows: 1. The release of GABA induced by the 10min-exposure of 25μM veratrine and 5min-exposure of 50mM potassium in the presence of 2.5mM Ca^2+ was 228.9±11.2 nmol and 100.1±8.9nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 6.8 and 4.6-fold increase respectively. 2. The release of GABA induced by the 10min-exposure of 25μM veratrine and 5min-exposure of 50mM potassium in the absence of Ca^2+ was 381.4±30.2 nmol and 55.1±4.1 nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 11.3 and 2.4-fold increase respectively.

        검도 게재연구논문에 활용된 통계 기법의 타당성 평가

        The purpose of this study were to evaluate validity for statistic technique applied to Kendo studies published at Domestic Journals. To achieve this purpose, We entered “Kendo” to search engine of web-site RISS(Research Information Sharing Service: www.riss.kr), selected 33 quantitative paper related “Kendo“ in this study, finally. And we evaluated these papers via index suggested Kim. and Kim(1990), Choi. et al.(1990). Statistic technique used in these studies related “Kendo” appeared to apply frequency analysis, chi-square, t-test, ANOVA, correlation analysis, regression analysis, factor analysis, and structural equation model. and We could find statistical error in selected papers mostly. And results of evaluation on validity of statistical technique applied as follows: First, a statistical assumption were ignored in most papers. Second, a statistical procedure were left out portion of papers. Finally, a statistic were left out portion of statistics techniques.

      • 형광검출 역상 액체크로마토그래피에 의한 Histamine의 정량

        Histamine을 정확하고 신속하게 정량하기 위해 9.Fiuorenylmethyl chloroformate를 형광유도체화제로 하여 역상 HPLC법으로 정량하였다. 히스타민을 형광유도체화할 때 반응액의 pH, 반응시간, 형광유도체화제의 농도 등 최적 반응 조건을 검토하였다. 이 방법으로 히스타민을 분석한 결과 0.1~0.5ug/ml의 농도범위에서 상관계수가 0.922인 양호한 직선성을 나타내었으며 검출한계는 0.01??/ml였다. A rapid and simple method for the determination of histamine by reverse-phase high performance liquid chromatography with fluorescence detection was established. 9-Fluorenylmethyl chloroformate(FMOC) was used as fluorescent derivative reagent. The optimum conditions for the derivatiation such as pH, reaction time and he concentration of FMOC were investigated. Linearity of calibration curve was obtained between 0.1ug/ml/and 0.5ug/ml(r=0.922) and the limit of detection was 0.01ug/ml.

      • Prostaglandin E_2 와 Phorbol Myristate Acetate가 조골세포의 활성에 미치는 영향

        PGE_2 has both anabolic and catabolic activities. Excess levels of PGE_2 have been implicated in a number of pathological states associated with bone loss such as a number of chronic inflammatory conditions such as periodontal disease and rheumatoid arthritis. PGE_2 and other arachidonic acid metabolites have been shown to be potent stimulators of osteoclastic bone resorption in organ culture. The anabolic effects of PGE_2 were first noticed when an increase in periosteal woven bone formation was seen after the infusion of PGE_2 into infants in order to prevent closure of the ductus arteriosus. The cellular basis for the catabolic actions of PGE_2 has been well characterized. PGE_2 increases osteoclast recruitment in bone marrow cell cultures. PGE_2 also has a direct action on osteoclast serving to inhibit activity and can also indirectly activate osteoclast via other cells in the vicinity, presumably osteoblast. PKC was reported to be involved in the action of osteotropic hormones. PTH primarily affects osteoblasts and increases irrtracellular cAMP level, and PTH-stimulated osteoblasts may produce and release the substances that activate osteoclasts. PKC may mediate the action of PTH in osteoblasts. Moreover, PKC increase the number of receptors for 1,25-(OH)_2D_3 in the osteoblasts. The purpose of this paper is to study the actions of PGE_2 and PMA on osteoblastic clone MC3T3E1 cells. The effect of PMA and PGE_2 on the induction of alkaline phosphatase(ALP) was investigated in osteoblastic clone MC3T3E1 cells clutured in medium containing 04% fetal bovine serum. PMA and PGE_2 stimulated ALP activity and MTT assay in the cells in a dose-dependent manner at concentrations of 10-500ng/ml. Cycloheximide, protein synthesis inhibitor, inhibited the stimulative effect of PGE_2 and PMA on ALP activiy in the cells. PGE_2 also increased the intracellular cAMP content in a dose-dependent fashion with a maximal effect at 500ng/ml. Our present results suggest that PGE_2 stimulate the differentiation of osteooblasts and are involved in bone formation in vivo, as well as in bone resorption.

        To maintain its functional integrity, bone is continuously remodelled by a process involving resorption by osteoclasts and bone formation by osteoblasts. In order to respond to changes in the physical environment or to trauma with the relevant action, this process is strictly regulated by locally synthesized or systemic fators. The effects of DBcAMP and PGE_2 on the induction of alkaline phosphatase(ALP) was investigated in osteoblastic clone MC3T3E1 cells cultured in medium containing 0.4% fetal bovine serum. DBcAMP and PGE_2 stimulated ALP activity and MTT assay in the cells in a dose-dependent manner at concentrations of 10-500ng/ml. Cycloheximide, protein synthesis inhibitor, inhibited the stimulative effect of PGE_2 and DBcAMP on ALP activity in the cells. PGE_2 also increased the intracellular cAMP content in a dose-dependent fashion with a maximal effect at 500ng/ml. Our present results suggest that PGE_2 stimulate the differentiation of osteoblasts and are involved in bone formation in vivo, as well as in bone resorption.

        자원봉사자들의 탈북자 지원 경험 분석 : 설문결과를 중심으로

        Objectives: Since 1994, the number of North Korean defectors, escaping North Korea and entering South Korea, has in creased rapidly.The Government has provided most of te support for the defectors' adaptation to their new society.But as the number of defectors has increased, and the support needs have become more various, the support of volunteers from non-governmental organization(NGO) has become very important.To raise the level of the success and effectiveness of the support from the volunteers, their experiences were surveyed and analyzed. Methods: This study surveyed 83 volunteers of 7 NGOs involved in helping North Korean defectors' adaptation to South Korean society-48 men and 34 women. Results: In their relationships with defectors, women volunteers have more difficulties in their first 3 months.However volunteers in their 50s and 60s tend to have more stable and better relationships with defectors than younger volunteers.Many volunteers statedexperiences of human difficulties with defectors, for example, mistrust and anxiety in human relationship (n=34, 41.0%), psychological maladjustment(n=19, 22.9%), different ways of thinking and culture(n=17, 20.5%), economic difficulties(n=10, 12.0%), and lack of independence spirit(n=6, 7.2%).The volunteers said they have feelings of goddwill toward defectors when defectors showed warm heart(n=30, 36.1%), politeness(n=30, 36.1%), positive and active character (n=24, 28.9%), strong will(n=17, 20.5%), and plainness (n=14, 16.0%).The volunteers stated an absence of goodwill feelings to defectors when defectors showed uncompromising and aggressive attitudes (n=29, 34.9%), lack of independent will (n=28, 33.7%), selfishness (n=18, 21.7%), and avoidance of making human relationship (n=16, 19.3%).as for adequate training before commencing activity, 5 volunteers(6.0%) said they had received enough education before their activity, 31(37.3%) relatively enough, 38 (45.8%) relatively not enough, and 9 (10.8%) absolutely not enough education.If they were the owners of al company, 27 volunteers (51.9%) sanid they would want to thire defectors even if the defectors did not have the required ability. Conclusion: These results showed that NGO's volunteers have a friendlier attitude to defectors than policemen n charge and public officials, which is their strong point.Women volunteers experience more difficulties in their early relationships with defectors, suggesting the need for a specific pre-education program for women volunteers.Furthermore, NGOs would benefit from an increase in the number of volunteers in their 50's and 60's.Both volunteers and defectors need to understand each othe to a deeprer extent and the characteristics to which South Korean volunteers feel goodwill need to be used n their education.

      • 상업Sports시설의 지도자와 경영자의 전문적 능력에 관한 조사 연구

        A significant change in the curriculum in Korean physical education colleges can be observed recently. The major reason for this change were the decreasing demand for school physical education teachers and the increasing demand for instructors or managers in the commercial sport sector. The purpose of this study was to investigate the way the physical education college ought to be and to investigate the future direction of physical education college curricular, clarifying the professional knowledge and abilities required for both an instructor and manager in the collegial sport management. A survey was conducted using a random sample of employers and instructors in the commercial sport facilities such as swimming pools and tennis courts. The results of the survey show the majority of sport instructors are required not only to teach but to do task involved in sport management. Nevertheless, they are not really ready to do all the management tasks required. Therefore the future task in the physical education colleges is to develop the curriculum to include the knowledge and field experience, necessary in sport management.
