<P>Uniform and extremely small-sized iron oxide nanoparticles (ESIONs) of < 4 nm were synthesized via the thermal decomposition of iron–oleate complex in the presence of oleyl alcohol. Oleyl alcohol lowered the reaction temperature by reducing iron–oleate complex, resulting in the production of small-sized nanoparticles. XRD pattern of 3 nm-sized nanoparticles revealed maghemite crystal structure. These nanoparticles exhibited very low magnetization derived from the spin-canting effect. The hydrophobic nanoparticles can be easily transformed to water-dispersible and biocompatible nanoparticles by capping with the poly(ethylene glycol)-derivatized phosphine oxide (PO-PEG) ligands. Toxic response was not observed with Fe concentration up to 100 μg/mL in MTT cell proliferation assay of POPEG-capped 3 nm-sized iron oxide nanoparticles. The 3 nm-sized nanoparticles exhibited a high <I>r</I>1 relaxivity of 4.78 mM–1 s–1 and low <I>r</I>2/<I>r</I>1 ratio of 6.12, demonstrating that ESIONs can be efficient <I>T</I>1 contrast agents. The high <I>r</I>1 relaxivities of ESIONs can be attributed to the large number of surface Fe3+ ions with 5 unpaired valence electrons. In the in vivo<I> T</I>1-weighted magnetic resonance imaging (MRI), ESIONs showed longer circulation time than the clinically used gadolinium complex-based contrast agent, enabling high-resolution imaging. High-resolution blood pool MR imaging using ESIONs enabled clear observation of various blood vessels with sizes down to 0.2 mm. These results demonstrate the potential of ESIONs as <I>T</I>1 MRI contrast agents in clinical settings.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2011/jacsat.2011.133.issue-32/ja203340u/production/images/medium/ja-2011-03340u_0002.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja203340u'>ACS Electronic Supporting Info</A></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja203340u'>ACS Electronic Supporting Info</A></P>

Background: The purpose of this study was to investigate the effect of 8 weeks of weight-bearing exercise on cognitive function, fitness, and health-related quality of life in older adults. Methods: A total of 30 adults with 65 years of age or older were randomly divided into two groups: an exercise group and a control group. The exercise group participated in an online video whole body exercise intervention and the control group was instructed to maintain daily activities unchanged. The mini mental status examination (MMSE), Seiner fitness test (SFT), and health-related quality of life (QL) questionnaire were performed before and after this intervention periods. An independent t-test was used for comparisons between the two groups, and a two-way repeated ANOVA was used to calculate the interaction between the two groups and across time. We used a paired-test to compare pre- and post-test data. Results: Participating in weight-bearing exercise increased attention, recall, and repetition of cognitive functions. Older adults in the exercise group increased their two-minute walk and chair sit and reach in the senior fitness test. Health-related quality of life increased, but there was no statistically significant change. Conclusion: Eight weeks of weight-bearing exercise may improve cognitive function, physical fitness, and health-related quality of life in older adults. Further studies examining the effects of exercise on more participants with cognitive decline should help us derive specific recommendations to maintain health in older adults.

Labeled (n = 3) and unlabeled (n = 2) islets were transplanted into the kidney capsules of streptozotocin-induced diabetic mice. Blood glucose levels and MR signal intensities were monitored for 30 days post-transplantation.</P><P><B>Results</B></P><P>There were no significant differences in viability or insulin secretion between labeled and unlabeled islets. A strong correlation (<I>r</I>2 > 0.94) was evident between the number of transplanted islets and T2 relaxation times quantified by MRI. Transplantation with labeled or unlabeled islets helped restore normal sustained glucose levels in diabetic mice, and nephrectomies induced the recurrence of diabetes. The MR signal intensity of labeled pancreatic islets decreased by 80% over 30 days.</P><P><B>Conclusion</B></P><P>The transplantation of SPIO-labeled porcine islets into the kidney capsule of diabetic mice allows to restore normal glucose levels, and these islets can be visualized and quantified using a 1.5T clinical MR scanner.</P>
