KR-32570 (5-(2-Methoxy-5-chlorophenyl)furan-2-ylcarbonyl)guanidine) is a new cardioprotective agent for preventing ischemia-reperfusion injury. Human liver microsomal incubation of KR-32570 in the presence of NADPH resulted in the formation of two metabolites, hydroxy- KR-32570 and O-desmethyl-KR-32570. In this study, a kinetic analysis of the metabolism of two metabolites from KR-32570 was performed in human liver microsomes, and recombinant CYP1A2, and CYP3A4. The metabolism for hydroxy- and O-desmethyl-KR-32570 formation from KR-32570 by human liver microsomes was best described by a Michaelis-Menten equation and a Hill equation, respectively. The Clint values of hydroxy- and O-desmethyl-KR-32570 formation were similar to each other (0.03 vs 0.04 µL/min/pmol CYP, respectively). CYP3A4 mediated the formation of hydroxy-KR-32570 from KR-32570 with Clint = 0.24 µL/min/pmol CYP3A4. The intrinsic clearance for O-desmethyl-KR-32570 formation by CYP1A2 was 0.83 µL/min/pmol CYP1A2. These findings suggest that CYP3A4 and CYP1A2 enzymes are major enzymes contributing to the metabolism of KR-32570.

Background: While adapalene and benzoyl peroxide have both been demonstrated to be effective for the treatment of acne vulgaris, they can also cause skin irritation, resulting in patient incompliance. In addition, the irritation may be severe when adapalene is co-administered with benzoyl peroxide. Objective: The intention of this study was to evaluate the efficacy and safety of adapalene 0.1%-benzoyl peroxide 2.5% fixed-dose combination gel, and adapalene 0.1% gel monotherapy, for the treatment of mild to moderate acne. Any changes in the skin barrier function were also measured. Methods: A total of 52 patients applied adapalene-benzoyl peroxide gel or adapalene gel once daily. The changes in lesion counts, investigator.s global assessment, patient satisfaction, and adverse events were recorded for 6 weeks. The biophysical profiles of the skin, including transepidermal water loss and redness, were measured for 3 weeks. Results: Both agents were effective for the treatment of acne lesions. Adapalene-benzoyl peroxide showed a faster onset of action than adapalene, especially on inflammatory lesions. However, at week 6, both agents showed a similar effect in reducing lesion counts. While most patients in both groups experienced skin irritation from week 1, this gradually reduced. Likewise, the changes in transepidermal water loss and redness showed peak rates at week 1 and 2, but decreased over time. Conclusion: Adapalene-benzoyl peroxide gel is effective for the treatment of acne with a safety profile comparable to adapalene gel. The skin irritation experienced may be associated with the changes in skin barrier disruption, which lasts for 1.2 weeks.

<P>Achiral poly(diphenylacetylene)s (PDPAs: pMe3, pEt3, piPr3, pMe2OD1, pMe2ODI, and mMe3) with different alkyl side chains at the para or meta position of the side phenyl ring were prepared to examine achiral-to-chiral transformations upon thermal annealing in a chiral solvent. All the para-substituted PDPAs showed significant circular dichroism (CD) enhancement upon annealing, whereas the meta-substituted polymer, mMe3, was inert to the same treatment. To investigate the kinetics, the asymmetric conformational change was monitored by observing the changes in the magnitude of circular polarization (g(CD)) or optical rotation. PDPAs with bulkier, round-shaped side groups (pEt3 and piPr3) had greater g(CD) values at equilibrium than pMe3 with a smaller side group. Moreover, the activation energy for the forward reaction (Ea(f)) was lower in the bulkier polymers than in pMe3 owing to enhanced miscibility with the chiral solvent. Similarly, the long alkyl chains of pMe2O1 and pMe2ODI acted as internal plasticizers to lower their Ea(f) values relative to that of pMe3, whereas their g(CD) values at equilibrium were smaller than that of pMe3. The kinetics of the achiral-to-chiral transformation is discussed in detail based on the spectroscopic changes observed during the annealing process.</P>

<P>Controlling and understanding the changes in metastatic cancer cell adhesion, shape, and motility are of paramount importance in cancer research, diagnosis, and treatment. Here, we used gold nanoparticles (AuNPs) as nanotopological structures and protein nanocluster forming substrates. Cell adhesion controlling proteins [in this case, fibronection (Fn) and ephrinB3] were modified to AuNPs, and these particles were then modified to the layer-by-layer (LbL) polymer surface that offers a handle for tuning surface charge and mechanical property of a cell-interfacing substrate. We found that metastatic cancer cell adhesion is affected by nanoparticle density on a surface, and ∼140 particles per 400 μm<SUP>2</SUP> (∼1.7 μm spacing between AuNPs) is optimal for effective metastatic cell adhesion. It was also shown that the AuNP surface density and protein nanoclustering on a spherical AuNP are controlling factors for the efficient interfacing and signaling of metastatic cancer cells. Importantly, the existence of nanotopological features (AuNPs in this case) is much more critical in inducing more dramatic changes in metastatic cell adhesion, protrusion, polarity, and motility than the presence of a cell adhesion protein, Fn, on the surface. Moreover, cell focal adhesion and motility-related paxillin clusters were heavily formed in cell lamellipodia and filopodia and high expression of phospho-paxillins were observed when the cells were cultured on either an AuNP or Fn-modified AuNP polymer surface. The ephrin signaling that results in the decreased expression of paxillin was found to be more effective when ephrins were modified to the AuNP surface than when ephrinB3 was directly attached to the polymer film. Estrogen-induced acute pancreatitis: A case report and literature review

Estrogens are commonly used in gynecologic area, such as oral contraception, hormone replacement therapy, and in vitro fertilization-embryo transfer. Although estrogen is a common cause of acute drug-induced pancreatitis, there has been paucity of report in Korea. Clinical course of estrogen-induced acute pancreatitis is usually mild to moderate, but fetal case can occur. In addition, there can be a latency from the first administration to the symptom. Therefore, physicians should consider the possibility of the disease when a woman taking estrogen or previous history of taking estrogen presents with acute abdominal pain. Here, we report a case of estrogen-induced acute pancreatitis that occurred during the preparation for embryo transfer.

We have investigated tunneling magnetoresistance (TMR) properties of Fe / Al₂O₃/ Co magnetic trilayer junctions sputtered on single-crystal Si(001) substrates. Al₂O₃ layers with thicknesses of 50~200Å were deposited directly on the bottom ferromagnetic layer by a reactive rf sputtering. For comparsion, we prepared Pt/Al₂O₃/Pt tunnel junctions whose current-voltage (I-V) characteristics measured at 300 K indicates that reactively sputtered Al₂O₃ is a particularly good material for thin insulating barriers and allows us to form pinhole-free tunnel barriers. The magnetic tunnel junctions exhibit changes of tunnel resistance of about 0.1 % at 300 K with an applied magnetic field and it was found that most junctions with Co as a top electrode have rather good I-V and TMR characteristics compared to those with Fe as a top electrode. These results were discussed in relation to their interfacial properties on the basis of those for Pt/Al₂O₃/Pt.

Background: There have been several studies about herpes zoster in the Korean dermatological literatures. However, there are few studies reported on disseminated herpes zoster. Objective: The aim of this study is to determine the epidemiology and clinical features of the disseminated herpes zoster as compared with herpes zoster. Methods: The authors retrospectively reviewed the medical records on 14 cases of disseminated herpes zoster and 402 cases of herpes zoster at the Department of Dermatology Busan Paik hospital during the 3 years from 2010 to 2012, and if necessary, telephone interviews were executed. Results: The rate of new disseminated herpes zoster outpatients at our dermatology clinic was 0.11% with the mean age of 67.7 years and the males to females ratio was 2.5. The most frequent involved dermatome was the lumbar (42.9%) area. The rate of patients with underlying diseases or immunosuppression, the rates of postherpetic neuralgia and the frequency of narcotic analgesics uses were significantly higher in disseminated herpes zoster as compared with herpes zoster. The patient treatment satisfaction was significantly higher in herpes zoster as compared with disseminated herpes zoster. Conclusion: These results demonstrated that the disseminated herpes zoster is associated with higher rates of underlying diseases, immunosuppression, frequent incidences of postherpetic neuralgia, more uses of narcotic analgesics and lower patient treatment satisfactions as compared with herpes zoster.
