        Ginsenoside Rg1 augments oxidative metabolism and anabolic response of skeletal muscle in mice

        Background: The ginsenoside Rg1 has been shown to exert various pharmacological activities with health benefits. Previously, we have reported that Rg1 promoted myogenic differentiation and myotube growth in C2C12 myoblasts. In this study, the in vivo effect of Rg1 on fiber-type composition and oxidative metabolism in skeletal muscle was examined. Methods: To examine the effect of Rg1 on skeletal muscle, 3-month-old mice were treated with Rg1 for 5 weeks. To assess muscle strength, grip strength tests were performed, and the lower hind limb muscles were harvested, followed by various detailed analysis, such as histological staining, immunoblotting, immunostaining, and real-time quantitative reverse transcription polymerase chain reaction. In addition, to verify the in vivo data, primary myoblasts isolated from mice were treated with Rg1, and the Rg1 effect on myotube growth was examined by immunoblotting and immunostaining analysis. Results: Rg1 treatment increased the expression of myosin heavy chain isoforms characteristic for both oxidative and glycolytic muscle fibers; increased myofiber sizes were accompanied by enhanced muscle strength. Rg1 treatment also enhanced oxidative muscle metabolism with elevated oxidative phosphorylation proteins. Furthermore, Rg1-treated muscles exhibited increased levels of anabolic S6 kinase signaling. Conclusion: Rg1 improves muscle functionality via enhancing muscle gene expression and oxidative muscle metabolism in mice.

        Identification of Hepatitis E Virus in Bovine and Porcine Raw Livers

        Several animal species including pigs are directly involved in the zoonotic transmission of the hepatitis E virus (HEV) to humans. This study was conducted to detect HEV in bovine and porcine raw livers by nested reverse transcription-polymerase chain reaction. Zoonotic HEV strains were identified in 1.0 and 3.0% of the tested bovine and porcine livers, respectively. HEV-4 was detected in the bovine livers, but both HEV-3 and HEV-4 were identified in the porcine livers. These results indicate that zoonotic transmission of HEV may occur via consumption of raw or undercooked livers of pigs and cattle.

        급성백혈병에서 다제약제내성 인자(P-glycoprotein)의 표현과 치료예측

        연구배경 : 급성백혈병은 진단후 1차 관해유도 약물치료에 의해 60~80에서 완전관해가 유도되나 1년내에 50%의 환자에서 재발하며, 이들에게 재차 관해유도 치료가 시도되어도 관해율은 30~50%에 불과하다. 그리고 1차 관해유도치료에 불응성인경우 2차 약제로 관해유도 약물요법이 시행되나 그 성적 역시 저조하여 백혈병환자의 5년 무병생존율은 25%에 불과하다. 이같이 백혈병 치료실패율이 높은 원인중 항암약제내성이 중요인자이며, 약제내성중에서도 특히 다제 약제 내성 유전자(multidrug resistant gene; MDR1)의 활성화에 의해서로 고차내성이 없는 4가지 약제에 내성을 나타내는 다제약제내성이 중용한 원인이다. 방법 : 본 연구에서는 급성 백혈병환자에서 MDR1 유전자의 다제약제내성이 표현되는 과정중 DNA와 mRNA는 dot blot 분석으로, 최종산물인 P-glycoprotein은 면역조직화학적염색법(immunohistochemical staining method)으로 유전자의 증폭(amplification) 혹은 과표현(overexpression) 여부를 관찰하여 다제약제내성의 존재를 증폭 혹은 과표현 과정중 용이하게 진단할 수 있는 방법을 조사하고, 아울러 다제약제내성의 발현도를 백혈병 각 질병상태별로 조사하였다. 그리고 다제약제내성의 환자 치료결과 예측 및 치료약제 선택기준으로서의 임상적 유용성을 조사하기 위하여 다제약제내성 발현여부와 치료결과와의 상관성 및 다제약제내성 발현여부와 생체외 약제감수성 검사에 의한 약제내성과의 상관성을 조사하여 다음과 같은 결과를 얻었다. 결론 : 1) 다제약제내성은 DNA에서는 0%(0/26), mRNA에서는 7.7%(2/26), P-glycoprotein에서는 55.4%(41/74)관찰되었다. 2) P-glycoprotein 과표현은 원발성 백혈병 진단시 33.3%, 재발시 78.6%, 치료 불응성시 88.9%에서 관찰되어 약제에 노출시 발현빈도는 증가하였다(p<0.01). 3) 속발성 백혈병은 원발성 백혈병에 비해 P-glycoprotein 과표현 빈도가 높은 경향이었다(p=0.18). 4) 진단시 P-glycoprotein을 과표현하는 경우 관해유도율이 낮은 경향이었다(p=0.11). 5) P-glycoprotein을 이용한 관해유도실패 예측도는 수용할 만 하였으나(84%), 관해유도 예측도는 낮았다(54%). 6) P-glycoprotein 과표현시 adriamycin, vincristine에 대한 약제내성 발현 빈도가 P-glycoprotein을 과표현하지 않는 경우보다 높았다(p=0.44). 7) P-glycoprotein을 이용한 adriamycin, vincristine에 대한 약제내성 예측도는 수용할 만 하였으나(각각 75%, 81%), 약제 감수성 예측도는 낮았다(각각 57%, 43%). 8) P-glycoprotein을 과표현시 verapamil 병용에 의한 다제약제내성의 변화는 adriamycin에서 33.3%, vincristine에서 16.7%였다. 결론 : 이상의 결과로 면역조직화학적염색에 의한 P-glycoprotein 과표현 여부 관찰이 다제약재내성 존재 확인의 간편한 검사방법이며, 이를 치료결과 예측의 유용한 기준으로 사용할수 있을 것으로 생각된다. 그리고 다제약제내성이외의 약제내성 기전을 고려하여 생체외약제 감수성 검사를 병행, 적절한 약제선택을 함으로써 급성백혈병의 치료효과를 증가시킬 수 있을 것으로 기대된다. Background : The complete remission rate of the first induction chemotherapy is 60~80% in acute leukemia. Most patients who achieve complete remission will ultimately relapse within the first year and are much less responsive to the therapy with a brief second remission in a minority. Acquisition of drug resistance in cellular level is the major cause of treatment failure. Therefore, the identification of the mechanisms of drug resistance is an important goal of current treatment. One mechanism that has been extensively studied in vitro is a multidrug resistance (MDR) mediated by P-glycoprotein which is the product of the MDR1 gene, but there is a little information on it`s clinical application. Methods : In order to find out the suitable method for detecting MDR, and furthermore, to see if the detection of MDR can be used in predicting treatment results, here, the existence of MDR1 amplification/overexpression in DNA, RNA and P-glycoprotein level was studied. Results : P-glycoprotein overexpression by immunohis-tochemical staining method was more sensitive (55.4%) than KNA amplification (0.0%) and mRNA overexpression (7.7%) in detecting MDR in acute leukemic patients. The incidence of P-glycoprotein overexpression was higher in relapsed (78.6%) and in refractory patients (88.9%) than in initial state (33.3%). There was a tendency of low complete remission rate in patients with primary P-glycoprotein overexpression. The incidence of acquired P-glycoprotein overexpression was 71.4%. When comparing the correlation between the P-glycoprotein overexpressin and the clinical results, the predictability for th treatment failure in P-glycoprotein overexpressing patients was acceptable (84%), but the predictability for the remission in P-glycoprotein non-expressing patients was low (54%). These predictability were similar to the predictability for drug resistance by comparing the correlation between the P-glycoprotein overexpression and in vitro cytotoxic assay results; the predictability for drug resistance to adriamycin in P-glycoprotein overexpressing patients was acceptable (75%), but the predictability of drug sensitivity in P-glycoprotein non-expressing patients was low (57%). Combination of verapamil and cytotoxic drug reversed MDR in 33.3% for adriamycin and 16.7% for vincristine in P-glycoprotein overexpressing patents. Conclusions : These results suggest the P-glycoprotein by immunohistochemical staining method could be a useful method for predicting treatment results. However, the low predictability for the remissionn in P-glycoprotein non-expressing patients, suggested by some other drug resistance mechanisms, can be overcome by combination with in vitro cytotoxic assay. This fact might contribute to the better treatment results in acute leukemia by optimization of drug selection.

        앎과 지식 구성을 통한 PBL지식창출 공간 고찰과 대학무용교육 모듈

        As society migrates toward post industrial, we live in rapidly changing world with massive information. The effective knowledge in which this society requires, is no longer staying with traditional epistemology. Recently, it is argued that acquired knowledge through university is not usable sufficiently to solve complex problems generated by working environment. Therefore, in this study offers Problem- Based Learning module that begins with solving problems in real working life in dance curriculum. The way of solving real world problems require various skills development through self-learning and interdisciplinary. PBL offers acquisition of knowledge of those elements through interaction and cooporation with other in community. This study awares of the importance of PBL approach in university curriculum especially for dancers who are used to acquiring knowledge with their bodies. Therefore, this study aims to offer the validity to PBL approach through exploring what is our knowledge and knowing based on epistemology and ontology. This study examines, firstly, exploring knowledge constructivism through analyisng the type of knowledge and the process of knowing. Secondly, analysing the validity of constructivism learning and constructivism knowledge. Thirdly, exploring the knowledge producing space within PBL approach and offers effective dance curriculum based on PBL.

        트랜스리터러시 비대면 무용교육 사례를 통한 디지털 정체성 고찰

        Over the past 20 years, academic interest in body and embodiment has become a key topic, with a sharp increase in social science, cognitive science, and neuroscience. The contemporary era is undergoing major changes in our daily lives through digital technology and globalised networks. Digital technology promotes widespread shifts and interaction between people, language, and artifacts on a previously unimaginable scale, and is rapidly changing our environment. The ability to read, write, and communicate for this should not just be the ability to operate media, but should be able to interact with each other, reflecting the process of creating a person’s existence, thinking, and meaning in a critical and human way. Contemporary communication crosses the media and huge information in the digital world is made quickly and in a variety of ways. Now that the world is constantly moving rapidly in the digital world, education cannot rule out the communication flow of reading and writing for this movement. Text for education is no longer written letters. The body can not only allow external experiences through these digital media tools, but also influence our internal experiences with the help of digital media. Now that the digital world is spreading at a rapid pace, it is essential to consider whether it is a contemporary bodily knowledge, and to consider what changes dance education should interact with digital space in a time of rapid change in communication. Therefore, this study aims to recognise the need for change in dance education, depict the body’s cognitive embodiment in the digital world, and consider digital identity expression through this, and examine untact dance education cases as follows. First, for the discussion of digital embodiment, the digital existence and body through transliteracy are considered. Second, in order to consider the expression of digital identity, experience through untact dance education cases is discussed along with transliteracy dance text.

        트랜스리터러시와 무용의 문화적 텍스트 고찰

        Our life in 21th century has an unseparable relation with objects which have screens and internet such as a mobile phone and a computer screen. These tools are ubiquitous and globally networked and so, vast informations spread rapidly through the tools. However, reading these media are dependent on the effectiveness of technology therefore, screen image cannot be understood without having the specific recognition and the cultural literacy. Literacy has to develop continuously to read social transforms and acquire the method of engaging the world where the students live. Literacy as a main method to interpret, manage and product the world, is placed within our social practice and ideology. It develops continuously according with the phases the times through analysing, acting, depicting and exploring the interaction between human and human, and human and objects. Therefore, this study aims to expand the cultural role of transliteracy with metaphorical exploration of how dancing body text communicates within social·cultural practice and ideology. As a primary method to interpret and product the world in media era for communication process to understand contemporary culture, this study interrelates how transliteracy culturally conceptualise through speaking, auditory, language, and different sign system, and how different message produces the culture in multiple media. This study explores, firstly, it considers the knowledge that literacy contains as a primary method to interpret, manage and produce the world and describes the type of communicative knowledge of transliteracy. Secondly, it depicts producing culture through interaction between human and human, and human and objects which are grounded on social·cultural practice as a learning to understand media in digital media era. Lastly, it explores the characteristics of transliteracy text and dancing body text for the comparative analysis on exploring the communicative spaces of transliteracy.

        트랜스미디어를 통한 무용학습 디자인과 컨버젼스 공간 고찰

        The advent of massmedia introduces the change of perspectives with offering expanded the spectrum of the learning methods through various languages of technology, platform, communication format, and different contents. This allows great challenges to the various disciplines especially the education sphere. There is learning, which is under the environment of rapid changes, through the enhanced interaction, participation, ubiquitous information and communication penetration. Consequently, it causes the occurrence of the new forms of expression through media literacy and digital literacy. This means it is inevitable that we have to be aware of reading and writing through media technology. Transmedia is able to develop the contents of communication through learning of the media technology method of the application. It becomes popular as a learning method internationally because it adapts the space between learners, learners and teachers into a new sphere with amplifying the synergy. Therefore, this study aims at offering the principles and the foundation of dance learning design. This study considers that, Firstly, it explores the elements of the consideration during the process of constructing story to contemplate transmedia space as learning, and the potential capacities of learning, which contains the characteristics of each media, to offer the validity of transmedia learning. Secondly, it examines the notion and the role of text to operate transmedia learning and explores various strategies for the text. Finally, it discusses what is transmitted and what design is approachable to multimedia design and a learning process in dance. It illustrates a transmedia learning design for dance through exploring the narrative elements of dance to solve the problem in online as well as the physical environment.

        논문 6 : 무용언어의 기표, 기의 간극의 표현을 위한 연구방법론 고찰

        As dance is an instrument of expressing one’s emotion, it exists almost every society in various forms. It is also an aesthetic art form to allow to feel and express emotion of which more magnificent range than when it is in everyday life. Dance is able to deliver various and rich feeling because it expresses and stimulates emotion with human body through perception of multiple senses, but it is hardly explained with one definite word. Dancer shows the intention through the movement, which is consist of her/his inner experience and the audience sees the long process of dancer’s embodiment even though it is covered by codified movements. Therefore, this study begins with interrogation of how well interpret the expression intention which involves dancer’s inner experience that cannot explain with clear words and moreover, how to document such a subjective feeling and experience with socialised objective language in dance study. Dance reveals aspecific process within semiotic because its image through which enculturated human movement, pervades with social and cultural codes. Therefore, this study analyses the relationship between language and dance using associated with semiotics in that dance posits the exterior movement as signifiant and its interior embodied long process as signifie. This study aims to discuss the potential of various dance studies with suggestion of proper methodology to argue dancer’s indefinite inner experience to reveal semiotic function of dance. This study considers that Firstly, to analyse how dancer and audience communicate, it discusses kinesthetic empathy throughout a dance performance. Secondly, dance movement as significant, it depicts the notion of truth in the text, which dance study uses, to explore proper methodology based on the criterion of the postmodernism through its epistemology and ontology. Finally, it explains the destruction of hypothetic binary within the relationship of dancer and audience to proposes methodologies to deliver dancer’s a sort of signifiespace and its value to document with seemingly objective texts.
