        톱밥 발효사료 급여 및 분말어유의 첨가사료가 돈육의 이화학적 성질에 미치는 영향

        This study was conducted to investigate the influence of pork quality pigs which were fed with diets supplemented with fermented sawdust and sardin powder oil. The pigs were randomly assigned to one of the four treatments: a), control (commercial feed); b), T1 (30% fermented sawdust); c), T2 (10% sardin powder oil) d), T3 (30% fermented sawdust, l0% sardin powder oil and 30% limited amino acid) from 30 kg to 110 kg B. Wt. and slaughtered. The samples were stored at 0±1 C. The physico-chemical properties were determined at the dust of 2, 4, 6, 8 and 15. For the loin the pH's of all the treatments on the 15th day were higher than those of the 2nd day, and that of TI was higher than those of the others. The total moisture contents of all treatments decreased with the storage (P$lt;0.05), and there were no significant differences among treatments on the 15th day. The water soluble proteins of all treatments increased with time and were highest on the 8th day and decreased gianibicanth thereater. T1 had significantly higher water soluble protein than others(P$lt;0.05). The salt soluble proteins of all treatments increased with lure the days(P$lt;0.05). T1 had higher salt soluble protein than others during the storage period(P$lt;0.05). The brittleness, hardness, cohesiveness, gumminess and chewiness of all the treatments were not significantly different among during the period, but elasticity of them increased after from the 2nd day. The brittleness of control was higher than that of the others on the 15th day, and the hardness and chewiness of control were higher than those of the others on the 6th day(P$lt;0.05). Forthebelly the pH's of all the treatments on the 15th day were higher than those of the at 2nd day(P$lt;0.05). T1 showed higher pH than the others during the period(P$lt;0.05). The total moisture contents of control and T1 were significantly decreased with time and there were no significant differences between T2 and T3. T1 showed the highest value. The water soluble proteins of all the treatments decreased with time and was lower on the 15th day(P$lt;0.05). That of control and T1 was significantly higher than that of others on the 2nd day, but there were no significant differences among the treatments on the 15th day. The salt soluble proteins of all treatments increased with time (P$lt;0.05), and they were highest on the 15th day. T1 had significantly higher salt soluble protein than the others on the 8th and 15th days.

        톱밥 발효사료 및 분말어유의 첨가사료가 돈육의 선도에 미치는 영향

        This study was conducted to investigate the influence feeding fermented sawdust and powdered sardin oil on meat quality of pork. The pigs were randomly assigned to one of four treatments: a), control (normal feed); b), T1 (normal feed and fermented sawdust 30%); c), T2 (normal feed and sardin powder oil 10%); d), T3 (normal feed, fermented sawdust 30%, sardin powder oil 10% and limited amino acid 30%) from live weight 30 ㎏ to 110 ㎏ and slaughtered. The samples were stored at 0 ± 1℃. The shelf life was determined for on the days of 2, 4, 6, 8 and 15. The results obtained were summarized as follows: The thiobarbituric acid reaction substance(TBARS) values of all the treatments were increased with the storage periods (P$lt;0.05). T1 had lower TBARS value than the other treatments until the 8th day, there were no significant differences among treatments at the 15th day. There were no significant differences among volatile basic nitrogen (VBN) of the treatments with the storage periods. The peroxide value of all the treatments was decreased with days of the storage (P$lt;0.05), but was increased again after 8th day. That of T1 was significantly lower than that of others at the 15th day. The TBARS of all the treatments increased with days of the storage(P$lt;0.05), and they were not significantly different each others. There were no significant differences among VBN of treatments with the storage periods. The peroxide values of all the treatments were decreased with the storage periods(P$lt;0.05), but was increased again after 8th day. That of T1 was significantly lower than those of the others at the 15th day.

        한우 및 수입육의 품종간 특이성분의 검색에 관한 연구

        증류수, 식염수, SDS, Triton X-100을 이용하여 한우 및 에버딘앵거스육으로부터 추출한 단백질을 SDS-PAGE에 의하여 분석한 결과, 한우육 및 에버딘앵거스육의 구성 단백질에는 큰 차이가 없음이 확인되어 SDS-PAGE 분석에 의한 한우육과 에버딘앵거스육의 식별은 불가능하였다. 증류수로 추출한 육 단백질을 이용하여 lectin 염색을 실시한 결과, LCA, PNA, 그리고 UEA-1에서는 그 차이를 발견할 수 없었으나, DBA의 경우 약 32.1kDa의 성분이 한우육에서 많이 생합성되는 것으로 나타났다. 내열성 단백질의 함량을 측정한 결과, 온도가 상승함에 따라 그 변성도가 심하여 졌으며, 이러한 경향은 한우육보다 에버딘앵거스육이 격심한 것으로 확인되었다. 100℃에서 30초 가열한 후, 내열성 단백질을 이용하여 LCA lectin 염색을 실시한 결과, 한우 특이적인 성분을 검색할 수 있었다. 또한 에버딘앵거스육에서는 한우육 특이성분보다 분자량이 다소 적은 성분이 특이적으로 농축되었으며, 이 성분은 한우육에는 없는 것으로 나타났다. Sodium dodecyl sulfate-polyacrylamide gels electrophoresis (SDS-PAGE) of muscles extracted with distilled water, saline solution, SDS or Trition X-100 resulted in similar protein patterns between Hanwoo and Aberdeen Angus (Angus) meat, indicating that SDS-PAGE cannot be used for identification between Hanwoo and Angus meat. Lectine staining analysis of muscle extracted with distilled water demonstrated that Hanwoo and Angus meat had similar affinities for lens culinaris agglutinin (LCA), peanut agglutinin (PNA) or ulex europaeus agglutinin (UEA-1) lectins. However, approximately 32.1 kDa component of Hanwoo meat showed high affinity for dolichos biflorus agglutinin (DBA) lectin. Hanwoo meat-specific Also the component of heat-denatured meat at 100℃ for 30 sec, lectin staining of heat-denatured meat at 100℃ for 30 sec, which was slightly smaller than Hanwoo meat-specific component, was concentrated specifically in Angus meat.

        실험 무창육계사의 환경효율 분석

        본 연구는 무창계사에 대한 효율 비교분석 연구가 전무한 실정으로 무창계사의 환경효율을 검증하고자 하였다. 본 실험은 축산기술연구소 신축 실험계사에서 2002년 5월 9일에서 5월 30일 사이에 실시하였으며 실험결과는 다음과 같다. 1. 무창계사는 외부의 기온은 9.6∼21.2℃로 편차는 약 11.6℃를 나타내었는데 비하여 단열이 우수한 무창계사에서는 병아리 사육적온기인 32∼33℃ 범위를 유지하여 약간의 보조열원으로 낮과 밤의 편차는 2℃ 이상 나타나지 않아 온도효율이 높았다. 2. 공기풍속에 있어서는 무창계사는 상부 0.57 m/sec, 중앙 0.22 m/sec, 하부 0.04 m/sec로 각각 나타나, 계사내 적정 풍속유지를 위한 덕트입기구의 천공으로 인하여 계사내 전체공간에서 공기의 유속흐름 분포가 고르게 나타났다. 이상의 실험결과를 종합해 볼 때 조사대상이된 무창계사의 온도 및 습도 환경이 우수하게 나타났으며, 특히 공기속도는 계사 공간 내에서 아주 고른 분포를 가져와 측벽 덕트 입기시설과 측벽 배기시스템이 우수하였다. The experiment was conducted to evaluate a ventilation system, which was devised to encourage farmers to use the enclosed poultry housing system. The study was observed in the National Livestock Research Institute from May 9 to May 30 in 2002. The main results of the experiment are as follows : 1. Although the outside temperature of the enclosed poultry house was 9.6∼21.2℃ with 11.6℃ variation, the house with an excellent heat insulation was maintained at 32∼33℃ in a variation of 2℃ which is within the range of the optimal temperature for broiler, being aided with two small electric heaters. 2. The average of air flow rates of the upper, middle and low parts of the room in the broiler house were detected at 0.57, 0.22 and 0.04 m/sec, respectively. The air flow in the whole room was distributed uniformly by a perforated duct. In conclusion, heat and humidity could be controlled without any problem in this enclosed housing system. Especially, air flow in all parts of the room was detected in uniform rates, resulting in the better ventilation performance with air inhalation through the duct and air exhaust through the side walls of the house.

        SDS - PAGE 기법을 이용한 우육 , 돈육 및 계육의 축종 판별에 관한 연구

        The objectives of the present study were to investigate 1) effects of various extraction solution on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) patterns of muscle proteins, 2) species-specific muscle protein components among beef; pork, and chicken meat and 3) hexose amounts and lectin staining patterns of muscle proteins from Hanwoo and Holstein beef. Although great amount of approximately 1.1 kDa and 7.5 kDa protein components were extracted from Hanwoo beef with 0.5% sodium dodecyl sulfate (SDS) or saline solution and Triton X-100, respetively, there were no differences in SDS-PAGE patterns of muscle proteins from Hanwoo or Holstein beef which were extracted with distilled water, 0.5% SDS, 0.5% Triton X-100 or saline solution. The SDS-PAGE patterns of muscle proteins from various parts of Hanwoo beef did not differ from those of Holstein beef. However, SDS-PAGE patterns of muscle proteins among beef, pork and chicken meat differ significantly. However those of 68, 47, 43 and 28 kDa were observed particularly in beef but not in pork or chicken meat. However those of 40 and 34 kDa were identified in pork but not in beef or chicken meat. The components of 50 and 38 kDa were identified in chicken meat but not in beef or pork. Observations that amount of hexose at various parts of Hanwoo and Holstein beef did differ in a certain degree, and that lectin staining patterns[in particular, affinities for Dolichos diflorus agglutinin (DBA) and Ulex europaeus agglutinin (UEA-1)]did differ between Hanwoo and Holstein beef suggest that unique sugar chain may be synthesized in breed-specific manners in beef.

        Bilateral versus unilateral placement of metal stents for inoperable high-grade malignant hilar biliary strictures: a multicenter, prospective, randomized study (with video)

        <P>Background and Aims: The efficacy of palliative biliary drainage by using bilateral or unilateral self-expandable metal stents (SEMSs) for a malignant hilar biliary stricture (MHS) remains controversial. This prospective, randomized, multicenter study investigated whether bilateral drainage by using SEMSs is superior to unilateral drainage in patients with inoperable MHSs. Methods: Patients with inoperable high-grade MHSs who underwent palliative endoscopic insertion of bilateral or unilateral SEMSs were enrolled. The main outcome measurements were the rate of primary reintervention for malfunction after successful placement of SEMSs, stent patency, technical and clinical success rates, adverse events, and survival duration. Results: A total of 133 pathology-diagnosed patientswere randomized to the bilateral group (n=67) or the unilateral group (n=66). The primary technical success rates were 95.5%(64/67) and 100%(66/66) in the bilateral and unilateral groups, respectively (P = .244). The clinical success rates were 95.3% (61/64) and 84.9% (56/66), respectively (P Z.047). The primary reintervention rates based on the per-protocol analysis were 42.6% (26/61) in the bilateral group and 60.3% (38/63) in the unilateral group (P = .049). The median cumulative stent patency duration was 252 days in the bilateral group and 139 days in the unilateral group. The risk of stent patency failure was significantly higher in the unilateral group (log-rank test; P < .01). In a multivariate Cox proportional hazardmodel to assess stent patency, bilateral SEMS placement was a favorable factor (adjusted hazard ratio 0.30, 95% confidence interval, 0.172-0.521; P <.001). Survival probability and late adverse events were not different between the 2 groups. Conclusions: Unilateral and bilateral drainage strategies by using SEMSs had similar technical success rates, but bilateral drainage resulted in fewer reinterventions and more durable stent patency in patients with inoperable high-grade MHSs.</P>

        Syntenin regulates TGF-β1-induced Smad activation and the epithelial-to-mesenchymal transition by inhibiting caveolin-mediated TGF-β type I receptor internalization

        <P>Syntenin, a tandem PDZ domain containing scaffold protein, functions as a positive regulator of cancer cell progression in several human cancers. We report here that syntenin positively regulates transforming growth factor (TGF)-beta 1-mediated Smad activation and the epithelial-to-mesenchymal transition (EMT) by preventing caveolin-1-mediated internalization of TGF-beta type I receptor (T beta RI). Knockdown of syntenin suppressed TGF-beta 1-mediated cell migration, transcriptional responses and Smad2/3 activation in various types of cells; however, overexpression of syntenin facilitated TGF-beta 1-mediated responses. In particular, syntenin knockdown abolished both the basal and TGF-beta 1-mediated repression of E-cadherin expression, as well as induction of vimentin expression along with Snail and Slug upregulation; thus, blocking the TGF-beta 1-induced EMT in A549 cells. In contrast, overexpression of syntenin exhibited the opposite effect. Knockdown of syntenin-induced ubiquitination and degradation of T beta RI, but not TGF-beta type II receptor, leading to decreased T beta RI expression at the plasma membrane. Syntenin associated with T beta RI at its C-terminal domain and a syntenin mutant lacking C-terminal domain failed to increase TGF-beta 1-induced responses. Biochemical analyzes revealed that syntenin inhibited the interaction between caveolin-1 and T beta RI and knockdown of syntenin induced a massive internalization of T beta RI and caveolin-1 from lipid rafts, indicating that syntenin may increase TGF-beta signaling by inhibiting caveolin-1-dependent internalization of T beta RI. Moreover, a positive correlation between syntenin expression and phospho-Smad2 levels is observed in human lung tumors. Taken together, these findings demonstrate that syntenin may act as an important positive regulator of TGF-beta signaling by regulating caveolin-1-mediated internalization of T beta RI; thus, providing a novel function for syntenin that is linked to cancer progression.</P>
