        Synthesis and Properties of N,N'-Bis(5-Aminosalicyl)-L-Cystine as a Colon-Specific Deliverer of 5-Aminosalicylic Acid and Cystine

        <P> N,N'-bis(5-aminosalicyl)-L-cystine (5-ASA-Cys) was prepared by a simple synthetic route. 5-ASA-Cys was not degraded in pH 1.2 and 6.8 buffer solutions, and in the homogenates of the upper intestine. In marked contrast, 5-ASA-Cys was deconjugated extensively to liberate 5-ASA in the cecal contents. Upon oral administration of 5-ASA-Cys to rats, the plasma concentration of 5-ASA-Cys was extremely low and the urinary recovery of 5-ASA-Cys was ∼ 10% of the dose. These results suggest that 5-ASA-Cys administered orally is delivered efficiently to the large intestine followed by deconjugation to liberate 5-ASA and cystine.</P>

      • Oxidative stress attenuates Fas-mediated apoptosis in Jurkat T cell line through Bfl-1 induction

        Many types of mammalian cells produce ROS in response to many different stimuli to modulate a number of cellular functions, including apoptosis. However, the correlation between ROS and apoptosis remains controversial, and the mechanisms whereby ROS-induced signals are propagated to critical downstream targets remain largely undefined. Here, we demonstrate that hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>) upregulates the expression of Bfl-1, an antiapoptotic member of the Bcl-2 family, and that this is responsible for the antiapoptotic activity of ROS. When Jurkat, human leukemic T cells, were pretreated with 100 μM H<SUB>2</SUB>O<SUB>2</SUB> and then treated with anti-Fas antibody, apoptosis was impaired without change of cell surface Fas expression. An investigation of the expression patterns of Bcl-2 family genes revealed that H<SUB>2</SUB>O<SUB>2</SUB> treatment induced Bfl-1 gene expression, but left other genes unchanged, and this Bfl-1 expression and H<SUB>2</SUB>O<SUB>2</SUB>-induced antiapoptotic effect was inhibited by antioxidants or NF-κB inhibitor. In addition, an electromobility shift assay revealed that the p65/p50 subunits of NF-κB activated by H<SUB>2</SUB>O<SUB>2</SUB> bound to a bfl-1 promoter. Neither the induction of Bfl-1 nor the antiapoptotic effect of H<SUB>2</SUB>O<SUB>2</SUB> was detected in Bfl-1-knockdown Jurkat cell line containing Bfl-1 antisense (Bfl-1AS). These data indicate that oxidative stress induces the expression of Bfl-1 via NF-κB activation, and this early-response gene protects cells from Fas-mediated apoptosis.       • Hypometabolism and altered metabolic connectivity in patients with internet gaming disorder and alcohol use disorder

        <P><B>Abstract</B></P> <P>Internet gaming disorder (IGD) has become the subject of growing concern as an addictive behavior and has been compared with substance/non-substance-related addiction. Although IGD show clinical impairments and social dysfunction, neurobiological alterations in IGD have not been clearly elucidated. We used <SUP>18</SUP>F-fluorodeoxyglucose PET to investigate differences in glucose metabolism and metabolic connectivity in young men [thirty-six patients with IGD, twenty-six patients with alcohol use disorder (AUD) and thirty-nine healthy controls (HC)].</P> <P>Compared with the HC, IGD showed hypometabolism in the anterior cingulate cortex (ACC), temporal, frontal, parietal and striatum and AUD exhibited hypometabolism in the occipital, temporal and parietal lobule. Furthermore, IGD showed negative correlations between the ACC and game duration and between the orbitofrontal cortex and impulsivity. Also, IGD had lower metabolic connectivity between temporal and limbic regions and between the motor area and occipital region. And AUD showed greater metabolic connectivity between the orbitofrontal and parietal regions, and between the somatosensory or parietal and temporal regions, but lower metabolic connectivity in the fronto-striatal or fronto-limbic regions. Our results provide evidences that hypometabolism and altered metabolic connectivity in IGD might be related to the abnormal sensory function by longtime gaming and dysfunction of impulsive/motivational states.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We assessed with PET in patients with internet gaming disorders (IGD) or alcohol use disorder (AUD). </LI> <LI> Addiction duration was associated with hypometabolism in IGD. </LI> <LI> Abnormal metabolism and different metabolic connectivity were observed in IGD and AUD. </LI> </UL> </P>

        Intravenous albumin for the prevention of contrast-induced nephropathy in patients with Liver cirrhosis and chronic kidney disease undergoing contrast-enhanced CT

        Background: The purpose of this study was to evaluate the incidence of contrastinduced nephropathy (CIN), and the effect of intravenous albumin for prophylaxis of CIN in patients with liver cirrhosis (LC) and chronic kidney disease (CKD). Methods: We conducted a retrospective study of 81 subjects with LC and CKD (estimated glomerular filtration rate (eGFR)o60 mL/min/1.73 m2) who underwent contrast-enhanced computed tomography (CT). Patients received either isotonic sodium bicarbonate solution (3 mL/kg for 1 h before CT and 1 mL/kg/h for 6 h after CT) or albumin (20% albumin, 25 mL for 1 h before CT and 75 mL for 6 h after CT). CIN was defined as an increase of Z25% or Z0.5 mg/dL in serum creatinine level. Results: Overall, CIN developed in three patients (3.7%). Of the 81 subjects, 43 received sodium bicarbonate solution and 38 received albumin. Both groups were comparable with regard to age, sex, diabetes mellitus, and baseline eGFR. The albumin group showed a significantly poorer liver function profile. CIN incidence did not differ significantly between the groups: it occurred in one (2.3%) of the 43 subjects receiving sodium bicarbonate and two (5.3%) of the 38 subjects receiving albumin (P¼0.6). However, the albumin group showed a significantly smaller increase in body weight (P¼0.03). Conclusion: The incidence of CIN in patients with LC and CKD undergoing contrast-enhanced CT after preventive measures was relatively low. The incidence of CIN was not significantly different between sodium bicarbonate and albumin groups.

        Exploring the Determinants of Preference for Visiting Beauty & Health Multi-Shop Operate by One Person

        Purpose: A beauty health multi-shop is a place where two or more beauty and health-related professional services are managed, and it is divided into a multi-person multi-shop where multiple managers manage users and a one-person multi-shop that is operated by a single person. This study was conducted to conduct an objec-tive and in-depth exploration of how the preference determinants of beauty health one-person multi-shop cus-tomers are organized. Method: As a research method, the first open-ended survey was conducted, and based on this, preliminary questions were constructed through content analysis, and the second closed-ended survey was conducted. The first survey was conducted on 95 adults with experience in using beauty and health one-person multi-shops, and the second survey was conducted on 212 questionnaires with the same experience, and the final preference de-terminants were derived through exploratory factor analysis and reliability verification. Results: The results of the study were implied by 17 sub-constructs, 6 constructs and 1 other factor. The first three internal preference determinants were identified as manager expertise, customer care environment, and affordability, and the second three external preference determinants were identified as diversity, convenience, and member retention services. Conclusion: Currently, in Korea, the beauty health industry is based on the health industry and the beauty industry is being mixed and converged, creating new demands that are connected to health, culture, emotions, and aesthetic pursuits. The results of this study provide an objective explanation of the determinants of consum-ers' preference for visiting beauty and health one-person multi shops, so it is expected to be used as a resource for various operational measures in the future.

        지하수 히트펌프 시스템의 지중 환경관리를 위한 시추 슬라임의 원핵생물 유전자 다양성

        지하수의 열을 이용한 히트펌프 시스템에서는 열 효율 유지를 위한 클로깅 현상이 고려되어야 한다. 클로깅 현상은 토양·지하수 환경에서 이화학적 요인 외에도 미생물학적 요인으로 발생한다. 이번 연구에서는 안정적인 지하수 열원 냉난방 시스템 운영을 위하여, 대수층 수위강하의 영향을 받지 않는 지하 10 미터 지점에서 불교란 시추 슬라임의 초기상태 원핵생물 다양성을 조사하였다. 세균은 문 수준에서 Proteobacteria (20.8%), Acidobacteria (18.8%), Chloroflexi (16.9%) 및 Firmicutes (10.2%) 등으로 나타났고, 속 수준에서는 Koribacter속 등 144개속이 분석되었다. 고세균은 문 수준에서 Thaumarchaeota (42.8%), Crenarchaeota (36.9%) 및 Euryarchaeota (17.4%)이 나타났으며, 강 수준에서 약 69.4% 비율로 Miscellaneous Crenarchaeota Group (MCG), Finnish Forest Soil Type B (FFSB) 및 Thermoplasmata가 분석되었다. Operational taxonomic units (OTUs)는 세균 3,565 및 고세균 836 OTUs로 나타났고, 세균이 고세균에 비해 풍부하며 우점도가 낮게 나타났다. 또한, 관정 막힘 현상을 유발할 가능성 있는 세균 후보군 135개(1.9%) reads가 분석되었으며, 향후 클로깅 현상에 대한 연구에 자료로 활용할 수 있을 것으로 기대된다. Groundwater heat pump (GWHP) systems must consider phenomena such as clogging to improve system efficiency and maintenance. In this study, we evaluated the prokaryotic diversity in a boring slime sample obtained at a depth of 10 m, which represented an undisturbed sample not affected by aquifer drawdown. Bacteria belonging to the phyla Proteobacteria (20.8%), Acidobacteria (18.8%), Chloroflexi (16.9%), and Firmicutes (10.2%) were found. Additionally, 144 species were identified as belonging to the genus Koribacter. Archaeal phyla were detected including Thaumarchaeota (42.8%), Crenarchaeota (36.9%), and Euryarchaeota (17.4%) and the class level comprised the miscellaneous Crenarchaeota group (MCG), Finnish forest soil type B (FFSB), and Thermoplasmata, which collectively accounted for approximately 69.4% of the detected Archaea. Operational taxonomic units (OTUs) were analyzed to reveal 3,565 bacterial and 836 archaeal OTUs, with abundances of 7.81 and 6.68, and richnesses of 5.96E-4 and 2.86E-3, respectively. The distribution of the groundwater microbial community in the study area showed a higher proportion of non-classified or unidentified groups compared to typical communities in surface water and air. In addition, 135 (approx. 1.9%) reads were assigned to a bacterial candidate associated with clogging.

        Exploring the Experiences of Automobile Manufacturing Workers Participating in Rehabilitation Exercise Programs after Industrial Accidents

        Purpose: This study was analyzed with a qualitative research method by applying a phenomenological method to examine the experiences and perceptions and attitudes about health care of workers participating in rehabil-itation exercises after suffering an industrial accident while working in the K automobile manufacturing industry in Korea. Method: As for the selection of research participants, a total of 12 participants were selected as the final research participants, including 1 female and 11 male, who could provide a wealth of information on the subject of the study. For data collection and analysis, Colaizzi's six-step analysis method was used to identify the partic-ipants' experiences and cognitive changes who participated in the rehabilitation program. Also, the participants' facial expressions and actions were recorded so that they could be used as additional observational data and reflected in the results of further research. Results: As a result of the study, 5 categories, 13 topic collections, 32 topics, and 97 construct meanings were finally confirmed. Specifically, the importance of strengthening the intrinsic motivation of the participants was emphasized in order to continuously practice health promotion after an industrial accident in the automobile manufacturing industry. In addition, it appears that a method of self-awareness of the necessity of exercise, giving fun of the complex program, and preventing and managing it in daily life is needed. Conclusion: The contents derived from the results of this study represent the changing perception of workers toward continuous health promotion management and improvement after an industrial accident. Through the research results, it was an opportunity to confirm the changing perception of health care among workers, and it is expected to be actively used in the future by developing rehabilitation exercise programs at the workplace in the automobile industry.
