      • 갑상선 세포에서 전사보조활성인자인 CBP와 CIITA의 인터페론-감마 활성 부위에 대한 조절 : CBP and CIITA on GAS(Interferon- γ Activated Site)-Mediated Transcription in Thyroid Cells

        Background : In the previous stuides, we identified that the interferon-r activated sequence (GAS) in the 5-flanking region of rat ICAM-1 gene is major element for interferon-r-inducible expression of the gene in rat thyroid cells, FRTL-5. We here, investigated the role of transcriptional coactivators, CBP(CREB binding protein) and CⅡTA (class Ⅱ transactivator) in the modulation of the activity of GAS which could interacts with signal transducers and activators of transcription-1 and 3 (STAT1 and STAT3). Methods : The expression of CBP RNA and protein were quantitated in FRTL-5 after stimulation with interferon-r (IFN-r), thyroid stimulating hormone (TSH), forskolin and methimazole. Direct association of CBP with STAT were analyzed by immunoprecipitation. The transcriptional roles of CBP and CⅡTA in the regulation of GAS were assessed by the cotransfection with their expression vectors with reporters; 5-deletion constructs of rat ICAM-1 promoter or 8xGAS-luc constructs, into FRTL-5 thyroid cells. Results : The level of CBP RNA and protein were not changed by the treatment with TSH, IFN-r, forskolin and methimazole in FRTL-5, FRT and BRL liver cells. The CBP could be directly associated with STAT1. Furthermore, the overexpression of CBP significantly increases the both promoter activities; rat ICAM-1 gene promoter which has GAS element and 8xGAS-luc cassette constructs. However, the cotransfection of CⅡTA decreased the constitutive and CBP-mediated transactivation of rat ICAM-1 promoter and 8xGAS-luc cassette constructs. Conclusion : We identified that the two tanscriptional coactivators; CBP and CⅡTA has differential roles in the regulation of transcriptional activity of GAS drived promoter. CBP increases the GAS activity through the direct binding with STAT1, but CⅡTA inhibited the CBP-mediated transactivation of GAS activity.

        Quantification of reproductive effort of the triploid Pacific oyster, <i>Crassostrea gigas</i> raised in intertidal rack and bag oyster culture system off the west coast of Korea during spawning season

        <P><B>Abstract</B></P> <P>Oyster culture forms one of the major produce of aquaculture industry in Korea. Due to the demand for production, it is necessary to maintain a steady culture process. However, as a result of oil spill accident and other environmental problems in the west coast, there is a need to find a quick recovery in the production of oysters. While oyster culture in Korea is dominated by use of diploid oysters, an alternative to fast recovery for oyster industry is to introduce the use of triploid oysters. Triploid oysters have advantage in terms of faster growth and negligible quantity of reproductive effort during spawning, resulting in shorter grow-out period and improved meat quality compared to diploid oysters. In this study, we determined the reproductive effort in triploid females of Pacific oyster, <I>Crassostrea gigas</I> using an indirect enzyme-linked immunosorbent assay (ELISA). Two years old triploid and diploid female oysters raised in rack and bag oyster culture system on the west coast of Korea were collected during spawning and post-spawning season in 2013. Histology revealed that the diploid females spawned from June to September, with a peak in June. Unlike diploids, the triploid females exhibited no sign of spawning as the diploids were actively engaged in spawning. During post-spawning season, the triploid oysters reabsorbed the mature remnant eggs in their follicles. Gonad somatic index (GSI), determined using ELISA, of the diploid females reached its peak in June as 22.2%. In contrast, GSI of the triploids recorded during spawning peak was only 3.4%. Carbohydrate and glycogen contents in the triploid oyster tissues during spawning season were much higher than in diploids, suggesting that the triploids used minimum energy for reproduction in summer season, containing very small amount of gonad and high level of glycogen.</P> <P><B>Statement of relevance</B></P> <P>Reproductive efforts (amount and number of eggs) of the diploid and triploid Pacific oysters measured in this study clearly demonstrated sterility of the triploid oysters during spawning season in summer.         서해안 갯벌의 춘·하계 대형저서동물의 군집구조 및 건강도 평가

        본 연구는 서해안 갯벌의 퇴적물 환경 및 대형저서동물 군집의 특성과 건강도를 파악하기 위해 수행되었다. 조사는 강화도부터 전남 목포까지 2017년 4월(춘계)과 8월(하계), 2018년 4월(춘계)에 실시하였다. 충남 권역의 퇴적물 입도는 조립하였으며, 경기·인천 권역, 전북 권역, 전남 권역의 퇴적물은 세립하였다. 대형저서동물은 총 140종으로, 대형저서동물 군집유사도를 분석한 결과, 대형저서동물군집은 4개의 그룹으로 나뉘었다. Group2는 바지락 어장 정점이 포함되었으며, 바지락(Ruditapes phillipinarum)과 남방백금갯지렁이(Nephtys polybranchia)와 짧은다리송곳갯지렁이(Lumbrineris nipponica)가 우점했다. Group4에는 퇴적상이 세립한 정점이 포함되었으며, 작은부채발갯지렁이(Eteone longa)와 유형동물(Nemertea unid.)이 높은 빈도로 출현하였다. 서해안 갯벌의 건강도 분석 결과, ISEP, BHI지수에서 대체로 'High (가장 양호)'상태를 나타냈으며, AMBI지수는 'Good (양호)'상태인 것으로 확인되었다. 따라서 서해안 갯벌의 건강도는 양호한 것으로 판단된다. 하지만 일부 지역에 오염지시종인 아기반투명조개(Theora lata), 등가시버들갯지렁이(Capitella capitata) 등이 출현하므로 갯벌 환경이 악화되지 않도록 주기적인 관리가 필요하다. This study investigated the characteristics of a macrobenthos community and sediment environment and assessed the health of tidal flats along the west coast of Korea. A survey was conducted from Ganghwa-do to Mokpo, Jeollanam-do in April (spring) and August (summer) 2017, and April (spring) 2018. The sediment grain sizes in the Chungcheongnam-do region were coarser, and the sediment in the Gyeonggi-do·Incheon, Jeollanam-do, and Jeollabuk-do regions were finer. A total of 140 macrobenthic species were collected from this study and using a cluster similarity analysis of the macrobenthos community, they were divided into four groups. Group2 was associated with Manila clam farm stations, and Ruditapes phillipinarum, Nephtys polybranchia and Lumbrineris nipponica were dominant. Group4 included some sites with finer sediment composed relatively, and Eteone longa and Nemertea unid. appeared at a high frequency. From the health assessment of the western tidal flat, the ISEP and BHI indices had a "High status," and the AMBI index had a "Good status." In conclusion, the tidal flats along the west coast of Korea have good ecological health. However, pollution indicator species such as Theora lata and Capitella capitata have appeared in some areas. Therefore, periodic administration and interventions are necessary to prevent deterioration of the tidal flat environment.

        일 대학병원 정신과 외래를 방문한 우울증 환자에서의 하지불안증후군

        Objectives : This study was designed to investigate the clinical characteristics, relationship with antidepressants and incidence of restless legs syndrome(RLS) in depressed outpatients. Methods : Total 160 adult patients(male 32, female 128) of psychiatric outpatient were enrolled in this study. We use the diagnostic criteria of International RLS Study Group(IRLSSG) for RLS. Also WHO quality of life scale abbreviated version(WHOQOL-BREF), Hamilton Depression Rating Scale-17(HAM-D) and Pittsburgh Sleep Quality Index(PSQI) was used to investigate the relationship between RLS and depression severity, quality of life, sleep quality. Results : In 39(24.4%) out of the total 160 patients were diagnosed as having RLS. Among those 39 RLS patient, 19 patients(48.7%) was reported to have developed their RLS after starting depression treatment. The RLS group showed higher PSQI and HAM-D scores(p=0.02, p<0.01) and lower WHOQOL-BREF score(p=0.06) compared to those of non-RLS. Compared to those of non-RLS group, the bupropion group showed a significantly low rate of RLS group (p=0.043) and the venlafaxine group showed a significantly high rate of RLS group(p=0.018). HAM-D score 7 or more group caused RLS 4.5 times compared those of less than HAM-D score 7 group. Conclusion : In this study, depressed outpatients showed high incidence rate of RLS and RLS had influence on sleep quality, depression severity and quality of life. Moreover, certain antidepressants had potential to induce RLS. Therefore, these results suggest that appropriate treatment for depression is required to clinicians and development of RLS from depressed patients should be carefully monitored. Further research about the relationship between antidepressant and RLS is also necessary.

      • 성인의 상하악 치조 전돌증의 교정 : 외과적 악교정 2례

        저자들은 영남대학교 의과대학 부속병원 치과에 상하악 치조전돌을 주소로 내원한 2명의 환자에게 상하악의 전치부 분절골절단술과 교정치료를 병행하여 심미적으로 좋은 결과를 얻었기에 치료결과를 아래와 같이 요약했다. 1. 전치부를 후방 위치시키는 외과적 술식으로 제 1소구치 발치 공간을 이용해 Wunderer, Kole방법을 사용했다. 2. crowding이 있었던 증례 2에서는 술전 교정으로 이를 해소했고 intrusion시키는 단계에서 외과적 시술을 시행하였다. 3. 술전 paper surgery, cast surgery에 의해 술후 결과를 어느 정도 예상할 수 있었다. 4. cast상에서 resin splint를 제작하여 6주동안 악내고정하였고 이에 따라 악간고정은 필요치 않았다. 5. 증례 2에서는 수술 후 남아있는 deep bite를 개선하기 위해 술후교정을 시행했다. 6. 각각 3개월, 5개월 관찰하였는 바 아직까지는 relapse와 합병증이 나타나지 않았으며 심미적인 면에서 개선된 양상을 보였다. Two patients, sought treatment for chief complaints of protruding frontal tooth and desired treatment to reduce the prominance of lips, were diagnosed as bimaxillary protrusion via clinical and cephalometric analysis. The authors corrected them by combined surgical and orthodonic treatment. As pre-surgical survey, paper and cast surgery were performed and wafer and resin sprint were constructed. We performed anterior maxillary and mandibular osteotomies in first premolar site to retract the maxillary and mandibular dentoalveolar segment in order to; 1) decrease prominance of upper and lower lips. 2) create proper lower incisor intrusion. By use of intramaxillary fixation, prompt oral intake was possible. We made good result of esthetic improvement and there was no evidence of relapse and any complication.

        Background: Neutrophil is an important inflammatory cell in severe asthma and associated with steroid-resistance in asthma therapy. In addition, interleukin (IL)-8 is a key chemokines recruiting and activating neutrophils. Methods: The aim of this study was to explore the therapeutic potential of aromatherapy using essential oils derived from plant extracts in asthma treatment. Essential oils from citron, pine tree, Chamaecyparis pisifera, and Chamaecyparis obtusa were extracted by water-steam distillation. The promoter activity and secretion of IL-8 were measured by using luciferase assay and enzyme-linked immunosorbent assay after pretreatment in tumor necrosis factor-α stimulated A549 and human bronchial epithelial cells (BEAS-2B) cell lines with diluted essential oils. Results: IL-8 transcription was suppressed with 1 : 500 concentration of each essential oil (P<0.05). IL-8 secretion in A549 cell was suppressed with 1 : 1,000 or higher concentrations of oils except from citron (P<0.05). IL-8 secretion in BEAS-2B cells was decreased after exposure to 1 : 500 concentration of pine tree, Chamaecyparis pisifera and 1 : 2,000 or higher concentrations of Chamaecyparis obtusa (P <0.05). Conclusion: Essential oils from pine tree and Chamaecyparis obtusa may have a therapeutic potential for asthma by regulating neutrophil-mediated inflammation.         2007년∼2011년 하절기에 양식 넙치, 조피볼락, 참돔, 새우의 세균 및 기생충 감염 현황

        2007년∼2011년 하절기에 국내 동·서·남해안과 제주의 육상양식장, 가두리 및 축제식 양식장에서 사육중인 넙치 (Paralichthys olivaceus), 조피볼락(Sebastes schlegeli), 참돔 (Pagrus major), 흰다리새우(Litopenaeus vannamei) 의 총 2,413마리에 대하여 세균과 기생충의 감염현황을 조사하였다. 조사기간(2007년∼2011년) 사이 어종별 병원체에 대한 감염양상을 종합하면, 기생충과 세균의 단독감염이 혼합감염에 비하여 비교적 높게 나타났으며, 단독감염 가운데 기생충이 세균보다 감염률이 높은 경향을 나타내었다. 조사기간 동안 넙치, 조피볼락, 새우에서 Vibrio spp. (V. harveyi, V. ichthyoenteri, Vibrio sp.)가 가장 우점종으로 검출된 세균의 위치를 차지하였으며, 참돔에서는 Vibrio sp.와 P. damselae가 우점종 세균으로 검출되었다. 조사기간 동안 가장 우점종으로 검출된 기생충은 넙치에서 스쿠티카충 (M. avidus)과 트리코디나충 (Trichodina sp.), 조피볼락에서 아가미흡충 (M. sebastes), 참돔에서 아가미흡충 (M. tai), 새우에서 섬모충 (Zoothamnium sp.) 으로 나타났다. Diagnostic monitoring in fish farms with land-based tanks and netpen cases were conducted in eastern, western, southern and Jeju island of Korea during summer of 2007∼2011. In total, 2413-fish samples of 4 marine fish species were tested for the detection of bacteria and parasite. Fish species tested were olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), red sea bream (Pagrus major), pacific white shrimp (Litopenaeus vannamei). During the diagnostic monitoring from 2007 to 2011, the infection rates by single infection of bacterial or parasitic pathogens were relatively higher than the mixed infections. The main bacterial pathogens in olive flounder, black rockfish and pacific white shrimp were Vibrio spp. (V. harveyi, V. ichthyoenteri, Vibrio sp.). The main bacterial pathogens in red sea bream were also Vibrio sp. and Photobacterium damselae subsp. damselae. The main parasitic pathogens were both Miamiensis avidus and Trichodina sp. in olive flounder, Microcotyle sebastes in black rockfish, Microcotyle tai in red sea bream and Zoothamnium sp. in pacific white shrimp.
