市乳의 貯藏期間에 따른 耐冷性 微生物 및 化學的 特性의 變化에 關한 硏究

The present study was conducted to predict shelf life of pasteurized milk with analysis of volatile compounds and sensory evaluation of milk to growth of psychrotrophic bacteria. Pseudomonas fluorescens KFRI 00194 inoculated concentration of 10^4 cfu/㎖ to observe of changes on sensory evaluation and volatile compounds of pasteurized milk. The results obtained in these experiments were summarized as follows; The psychrotrophic bacteria number at 4℃ were increased 6.39×10 exp(4)cfu/㎖ until first day and 2.20×10 exp(6)cfu/㎖, 1.17×10 exp(7)cfu/㎖ after 5 and 10days, respectively. The volatile compounds were shown the decreasing tendancies in acetaldehyde, ethanol depending on storage days, especially ethanol contents were significantly decreased between 5 and 10days storage(P<0.05). Diacetyl, 2-heptanone, 2-butanone and 3-methyl-1-butanol were shown the increasing tendencies of contents. Diacetyl, 2-heptanone and 2-butanone were significantly increased between 5 and 10days, 1 and 5days, 5 and 10days and 1 and 5days, respectively(P<0.05). However, 3-methyl-1-butanol was shown a increasing tendancy depending on storage days but the significantly different was not recognized. The changes of sensory evaluation were rapidly decrease from 6.8point first day to 4.2point after 10days and the decrease of organoleptic acceptability was significantly different recognized(P<0.05), excepted of 1 and 2days. The organoleptic acceptability according to increasing of psychrotrophic bacteria numbers were decreased. The organoleptic acceptability depending on changes of volatile compounds were shown to a conversed correlation coefficient with the exception of acetaldehyde and acetone. Especially, 2-heptanone, 2butanone and 3-methyl-1-butanol were gradually decreased of acceptability by the contents.
Functional Analysis of a Histidine Auxotrophic Mutation in Gibberella zeae

A plant pathogenic fungus, Gibberella zeae (anamorph: Fusarium graminearum), not only generates economic losses by causing disease on cereal grains, but also leads to severe toxicosis in human and animals through the production of mycotoxins in infected plants. Here, we characterized a histidine auxotrophic mutant of G. zeae, designated Z43R1092, which was generated using a restriction enzyme-mediated integration (REMI) procedure. The mutant exhibited pleiotropic phenotypic changes, including a reduction in mycelial growth and virulence and loss of sexual reproduction. Outcrossing analysis confirmed that the histidine auxotrophy is linked to the insertional vector in Z43R1092. Molecular analysis showed that the histidine requirement of Z43R1092 is caused by a disruption of an open reading frame, designated GzHIS7. The deduced product of GzHIS7 encodes a putative enzyme with an N-terminal glutamine amidotransferase and a C-terminal cyclase domain, similar to the Saccharomyces cerevisiae HIS7 required for histidine biosynthesis. The subsequent gene deletion and complementation analyses confirmed the functions of GzHIS7 in G. zeae. This is the first report of the molecular characterization of histidine auxotrophy in G. zeae, and our results demonstrate that correct histidine biosynthesis is essential for virulence, as well as sexual development, in G. zeae. In addition, our results could provide a G. zeae histidine auxotroph as a recipient strain for genetic transformation using this new selectable marker.

Ovarian cancer is a cancerous growth arising from the ovary and with poor prognosis that usually have resistant to all currently available treatments. Whether (E)-2,4-bis(p-hydroxyphenyl)-2-butenal (butenal) synthesized by Maillard reaction from fructose-tyrosine, has potential therapeutic activity against human ovarian cancer was investigated using two ovarian cancer cell lines (PA-1, SK-OV-3). We found that butenal could inhibit NF-${\kappa}B$/STAT3 activity, thereby inducing apoptotic cell death of ovarian cancer cells. We treated with several concentration of butenal each cell line differently (PA-1; 5, 10 and $15{\mu}g/ml$, SK-OV-3; 10, 20 and $30{\mu}g/ml$). First, ovarian cancer cell lines exhibited constitutively active NF-${\kappa}B$, and treatment with butenal abolished this activation as indicated by DNA binding activity. Second, butenal suppressed activation of signal transducer and activator of transcription-3 as indicated by decreased phosphorylation and inhibition of Janus kinase-2 phosphorylation. Third, butenal induced expression of pro-apoptotic proteins such as proteolytic cleavage of PARP, Bax and activation of caspase-3, -8 and -9. Lastly, combination of butenal and TRAIL causes enhanced induction of apoptosis. Overall, our results indicate that butenal mediates its anti-proliferative and apoptotic effects through activation of multiple cell signaling pathways and enhances the TRAIL-induced apoptosis. These data suggested that butenal may be a potential anti-cancer agent in ovarian cancer.
Validity of the Clinical Frailty Scale in Korean older patients at a geriatric clinic

Background/Aims: We aimed to assess the validity of the Korean translated version of the Clinical Frailty Scale (CFS) in determining the frailty status in geriatric outpatients. Methods: The records of 123 ambulatory outpatients who had undergone CFS and comprehensive geriatric assessments (CGAs) including measurements for the Cardiovascular Health Study (CHS) frailty scale and the frailty index (CGA-FI) were analyzed. Correlations between CFS, CHS frailty scale, and CGA-FI were assessed. The ability of CFS to classify frailty status was calculated using the CHS frailty scale and CGA-FI as references. Results: The mean CFS score was 3.2 in the study population, with a mean age of 77.49 years (45.5% men). Individuals with higher CFS scores were older, had a greater burden of chronic diseases, and worse daily functions and cognitive performance. CFS scores positively correlated with CGA-FI (B = 0.78, p < 0.001) and CHS frailty scale (B = 0.67, p < 0.001) scores. For CFS, C-statistics to classify frailty by CGA-FI and CHS scale were 0.905 and 0.826, respectively. The cut-off value of CFS ≥ 4 maximized Youden’s J to classify frailty by both the CHS scale and CGA-FI. Conclusions: The CFS is a valid screening tool to assess the frailty status in out-patients of a geriatric clinic in Korea. As a simple and quick measure, the CFS may facilitate frailty assessments in real-world clinical practice.

<P><B>Abstract</B></P> <P>The challenge of developing epoxy-foam adhesives for automotive applications lies in the difficulties associated with finding the optimal composition ensuring both high expansion ratio and mechanical strength. Herein, epoxy-foam adhesives based on epoxy composites were prepared and evaluated for automotive applications. The curing behavior was studied by differential scanning calorimetry, and the morphology was characterized non-destructively by micro X-ray computed tomography. Increasing the content of rubber-modified epoxy (RME) retarded the curing reaction by increasing the expansion ratio and affected the lap shear strength. A high expansion ratio and suitable mechanical strength was observed for epoxy-foam adhesives containing 10 or 15 wt% RME.</P>
