        Characterization of two novel γ-gliadin genes encoded by K genome of Crithopsis delileana and evolution analysis with those from Triticeae

        By acid polyacrylamide gel electrophoresis (A-PAGE) analysis,it was indicated that the electrophoresis mobility of gliadins from Crithopsis delileana (Schult) Roshev (2n=2x=14,KK) had obvious difference with those from common wheat in α, γ and ω region. Using homologous primers, two γ-gliadin genes (gli-Kr1 and gli-Kr2) were isolated from C. delileana,which had been deposited in the GenBank under accession numbers EU283818 and EU283821, respectively. Two γ-gliadin genes of C. delileana had the similar primary structures to the corresponding gene sequences from other wheat related species. The differences were mainly resulted from substitutions,insertions and deletions involving single amino acid residues or motifs of γ-gliadins. The repetitive domains of gli-Kr1 and gli-Kr2 from C. delileana are shorter than most of other sequences. By the alignment of γ-gliadin genes from A, B, D, Am, Au, S, Sl, Ssh, Ss and Sb genomes of Triticum and Aegilops, R genome of Secale (γ-secalin), Ee genome of Lophopyrum and K genome of Crithopsis in Triticeae, phylogenetic analysis indicated that two γ-gliadin genes of C. delileana could be clustered together with a γ-gliadin genefrom Ssh genome of Aegilops by an interior paralleled branch. It was the first time that the γ-gliadin genes encoded by K genome of C. delileana were characterized. These could offer precious information for better understanding the qualities associated with gliadins, the response in coeliac disease and studying the evolutionary relationship of gliadins in Triticeae.

        MicroRNAs (miRNAs) are endogenous non-coding genes that participate in post-transcription regulation by either degrading mRNA or blocking its translation. It is considered to be very important in regulating insect development and metamorphosis. Insects are the largest group of animals and are extremely valuable in biological and agriculture research. Insects are also important pests to human health and agriculture, and efforts are necessary protect both humans and plants from disease and damage. Despite their importance, insects lag behind mammals, nematodes, and plants in miRNA research. At present, only 279 insect miRNAs have been identified from Drosophila melanogaster, Anopheles gambiae, Apis mellifera, Bombyx mori, and D. pseudoobscura in miRBase, and most of these miRNAs were computationally predicted without experimental validation. Functional analysis of insect miRNAs has only been conducted in D. melanogaster.

        Effectiveness and Safety of Apatinib Plus Programmed Cell Death Protein 1 Blockades for Patients with Treatment-refractory Metastatic Colorectal Cancer: A Retrospective Exploratory Study

        This study aimed to investigate the efficacy and safety of apatinib plus programmed cell death protein 1 (PD-1) blockades for patients with metastatic colorectal cancer (CRC) who were refractory to the standard regimens. In this retrospective study, patients with metastatic CRC who received apatinib plus PD-1 blockades in clinical practice were included. The initial dosage of apatinib was 250 mg or 500 mg, and PD-1 blockades were comprised of camrelizumab, sintilimab and pembrolizumab. Efficacy and safety data were collected through the hospital’s electronic medical record system. From October 2018 to March 2022, a total of 43 patients with metastatic CRC were evaluated for efficacy and safety. The results showed an objective response rate of 25.6% (95% CI, 13.5%-41.2%) and a disease control rate of 72.1% (95% CI, 56.3%-84.7%). The median progression-free survival (PFS) of the cohort was 5.8 months (95% CI, 3.81-7.79), and the median overall survival (OS) was 10.3 months (95% CI, 5.75-14.85). The most common adverse reactions were fatigue (76.7%), hypertension (72.1%), diarrhea (62.8%), and hand-foot syndrome (51.2%). Multivariate Cox regression analysis revealed that Eastern Cooperative Oncology Group (ECOG) performance status and location of CRC (left or right-side) were independent factors to predict PFS of patients with metastatic CRC treated with the combination regimen. Consequently, the combination of apatinib and PD-1 blockades demonstrated potential efficacy and acceptable safety for patients with treatment-refractory metastatic CRC. This conclusion should be confirmed in prospective clinical trials subsequently

        Effect of Heat Treatment on Microstructure and Properties of FGH4096M Superalloy Processed by Selective Laser Melting

        A self-designed nickel-based superalloy, designated as FGH4096M, was prepared by selective laser melting (SLM). Differentheat treatments were performed to improve the mechanical properties of the SLM alloy through optimizing the microstructuresand the features of γ′ precipitates. Compared with the as-deposited alloy, the columnar grains with dendritic structuresand equiaxed structures were retained in the alloy after direct aging, but a large amount of tertiary γ′ phase precipitated,especially around the sub-grain boundaries, resulting in the highest tensile strength but the lowest elongation. During solidsolution and aging treatment (SSA), the recovery and recrystallization occurring in the alloy facilitated the grains to beequiaxed with the increase of solution temperature. For lower solution temperature (below 1100 ℃), the secondary γ′ precipitatesdecreased with the increase of solution temperature, while the tertiary γ′ precipitates from the subsequent agingprocess gradually increased; for higher solution temperature over 1100 ℃, exceeding the complete dissolution temperatureof the γ′ phase, only tertiary γ′ precipitates from the subsequent aging process were uniformly distributed in the alloy. Afterdouble solid solution (1170 ℃ + 1050 ℃) + aging heat treatment (DSSA), there were three sizes of γ′ precipitates in thealloy. In general, the SLM + SSA (1130 ℃) alloy obtained the best comprehensive properties, which could be related tothe homogenized microstructures and the uniform and dense distribution of single sized tertiary γ′ precipitates in the alloy.

      • Serum Carbohydrate Antigen 19-9 as an Indicator of Liver Metastasis in Colorectal Carcinoma Cases

        Purpose: The liver is the organ to which colorectal carcinomas (CRCs) most commonly metastasize, and surgical resection has been established as the most effective and potentially curative treatment for CRC with liver metastasis (LM). Therefore, surveillance of LM is vital for improvement of prognosis of CRC patients. In this study, we aimed to explore the potential value of carbohydrate antigen 19-9 (CA 19-9), carcinoembryonic antigen (CEA), and marker enzymes in indicating LM with CRC. Methods: Three groups of eligible patients with metastatic cancers were retrospectively included: CRC patients with LM (CRC-LM) or without LM (CRC-NLM), and non-CRC patients with LM (NCRC-LM). All metastatic lesions were identified by CT or MRI. Data on characteristics of the patients, the primary site, the locations of metastasis, CA 19-9, CEA, and biochemical parameters were collected for analysis. Results: A total of 493 patients were retrospectively included. More alcohol consumption was found in CRC-LM than CRC-NLM. Some biochemical enzymes were found to be significantly higher in groups with LM than without (CRC-LM or NCRC-LM v.s CRC-NLM). Both CEA and CA 19-9 were much higher in CRC-LM than CRC-NLM or NCRC-LM. For CRC patients, CA 19-9, ${\gamma}$-glutamyl transpeptidase, CEA and alcohol consumption were identified as independent factors associated with LM. Conclusion: Our analysis suggested the CA 19-9 might be a potential valuable indicator for LM of CRC in the clinic.

        A novel semi-IPN hydrogel: Preparation, swelling properties and adsorption studies of Co (II)

        A range of super-absorbent semi-IPN hydrogels—polyvinyl alcohol/poly (acrylic acid-co-acrylic amide)(PVA-P(AA-co-AM)) were synthesized via free radical polymerization method under ultrasound-assistedcondition. L16 (45) orthogonal experiments were designed to optimize the synthesis conditions and thesuccess was characterized by FTIR, SEM and TGA. Swelling capacities were studied in various pH solutionand saline solution whose results indicated that pH in solution shows an obvious influence on it and thesalt resistance is greater in salt solution of low valence relative to that of high valence. Meanwhile, itsswelling behavior was evaluated in water solution, which revealed that the swelling process conformedto the Schott model and the diffusion type was non-Fickian diffusion. Moreover, the adsorption on cobalt(II) from aqueous solutions was also investigated in the paper. The optimumpH value was found close to4 for the cobalt (II) adsorption and it was discovered that adsorption kinetics and adsorption isothermsfor cobalt (II) were in accordance with the pseudo-second-order model and the Freundlich modelrespectively. Besides, thermodynamic parameters were calculated for the cobalt (II) adsorption and thefindings revealed that the adsorption behavior is spontaneous and endothermic.

      • Retinoid Receptors in Gastric Cancer: Expression and Influence on Prognosis

        Background: Gastric cancer is frequently lethal despite aggressive multimodal therapies, and new treatment approaches are therefore needed. Retinoids are potential candidate drugs: they prevent cell differentiation, proliferation and malignant transformation in gastric cancer cell lines. They interact with nuclear retinoid receptors (the retinoic acid receptors [RARs] and retinoid X receptors [RXRs]), which function as transcription factors, each with three subclasses, ${\alpha}$, ${\beta}$ and ${\gamma}$. At present, little is known about retinoid expression and influence on prognosis in gastric cancers. Patients and Methods: We retrospectively analyzed the expression of the subtypes RARa, $RAR{\beta}$, $RAR{\gamma}$, RXRa, $RXR{\beta}$, $RXR{\gamma}$ by immunohistochemistry in 147 gastric cancers and 51 normal gastric epithelium tissues for whom clinical follow-up data were available and correlated the results with clinical characteristics. In addition, we quantified the expression of retinoid receptor mRNA using real-time PCR (RT-PCR) in another 6 gastric adenocarcinoma and 3 normal gastric tissues. From 2008 to 2010, 80 patients with gastric cancers were enrolled onto therapy with all-trans-retinoic acid (ATRA). Results: RARa, $RAR{\beta}$, $RAR{\gamma}$ and $RXR{\gamma}$ positively correlated with each other (p < 0.001) and demonstrated significantly lower levels in the carcinoma tissue sections (p < 0.01), with lower $RAR{\beta}$, $RAR{\gamma}$ and RXRa expression significantly related to advanced stages (p < =0.01). Tumors with poor histopathologic grade had lower levels of RARa and $RAR{\beta}$ in different histological types of gastric carcinoma (p < 0.01). Patients whose tumors exhibited low levels of RARa expression had significantly lower overall survival compared with patients who had higher expression levels of this receptor (p < 0.001, HR=0.42, 95.0% CI 0.24-0.73), and patients undergoing ATRA treatment had significantly longer median survival times (p = 0.007, HR=0.41, 95.0% CI 0.21-0.80). Conclusions: Retinoic acid receptors are frequently expressed in epithelial gastric cancer with a decreased tendency of expression and RARa may be an indicator of a positive prognosis. This study provides a molecular basis for the therapeutic use of retinoids against gastric cancer.

      • Factors Potentially Associated with Chemotherapy-induced Anemia in Patients with Solid Cancers

        Purpose: Chemotherapy-induced anemia (CIA) is one of the most important causes of anemia in cancer patients. This study was conducted to describe the prevalence and characteristics of CIA in solid cancer patients in the Chinese population, and to explore the relationship of white blood cell (WBC) or platelet decrease with CIA. Methods: Data on age, gender, tumor diagnosis, anti-cancer treatment and blood cell analyses were available from 220 untreated non-anemic cancer patients who received at least 2 cycles of chemotherapy, and the data were analyzed to assess their relationship with CIA or its severity. Results: 139 patients (63.2%) presented anemia, most being Grade 1 or 2. Esophageal and lung cancers were associated with a high prevalence. G3/4 leucopenia and decrease of platelets were identified as independent risk factors for the occurrence of CIA. Moreover, G3/4 leucopenia, decrease of platelet and G3/4 thrombocytopenia were found to be also associated with the severity of CIA. Cisplatin-containing regimens were a main potential factor in causing CIA, although significant association was only found on univariate analysis. Conclusion: Anemia or decrease in hematoglobin are common in Chinese cancer patients receiving chemotherapy. Cisplatin-containing regimens might be an important factor influencing the occurrence of CIA. Our analysis firstly described some risk factors, such as decrease of platelets or WBCs, severity of leucopenia or thrombocytopenia, associated with the occurrence and severity of CIA.
