      • 악성림프종에서 Phospholipase C의 발현

        Phosphatidylinositol-specific phospholipase C(PLC) plays a central role in transmembrane signal transduction pathways in response to the interaction between various ligands and cell-surface receptors in most eukaryotic cells. Triggered PLC catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate to yield the intracellular second messengers, diacyiglycerol and inositol 1,4,5-trisphosphate. It has been found that PLC-γ₁ is overexpressed in some cancer cells including breast, liver, and colorectal cancer, but data on the expression of PLC isozymes other than γ₁ have not been reported. Furthermore, there have been no reports on the expression of PLC in lymphomas. In this study, we examined the patterns of PLC isozymes expression in normal lymph node and lymph node samples from 15 patients with malignant lymphoma by western blot analysis and immunohistochemistry. The levels of PLC-β₁ expression were found to be similar between tumor samples and normal lymph node. PLC-β₂ was found to be overexpressed in 12 lymphoma samples showing, marked increase in 4 and slightly increased patterns in 8. PLC-β₃ was abnormally expressed quantitatively and qualitatively in all the lymph nodes from lymphoma patients compared to the normal lymph node. PLC-b3 showed a strong overexpression pattern in the lymphoma tissues, and the mobility of this protein was slightly retarded in all samples on the SDS/polyacrylamide gel. There were no detectable changes in PLC-γ₁ or δ₁ expression, but PLC-γ overexpression was observed in 10 of 15 lymphoma samples. The fact that lymphoma tissues showed an overexpression of PLC-β₂, -β₃, and -γ₂ indicates that the alteration of the signal transduction pathway is associated with the pathogenesis of lymphoma.

        With the discovery of cyclins and cydin-dependent kinases (cdk), key regulators of cell cyde, it is now possible to test whether the deregulation of any or all of these molecules could lead to oncogenesis. Information regarding the dinical significance of the cyclins and cdks is beginning to unfold. It has been found that certain cyclins or cdks are overexpressed in some tumor tissues including liver, parathyroid, and breast cancer. However, data on the expression of cyclins or cdks in lung cancer have not been reported. In this study, we examined the expression of cydins and cdks in various primary lung cancer tissues by Western blot analysis to determine if there was an altered expression in those tissues. Human lung cancer tissues consisted of 16 adenocarcinomas, 18 squamous cell carcinomas, 2 small cell carcinomas, 1 large cell cardnoma, and 1 with sarcoma. Western blot analysis showed that the levels of cyclin A and B1, and cdk2 expression were not elevated as compared to normal tissues. However, overexpression of cyclin Dl (8/38, 21%), cdk4 (8/38, 21%), and cdc2 (12/38, 32%) was observed in cancer tissues as compared to normal tissues. The expression of cyclin E was higher in all the cancer tissues tested than in normal tissues. These data implicate dysregulated expression of several cyclin and cdk genes, particularly cyclin E, as a potential factor in the pathogenesis of lung cancer.

      • Increased Expression of Phospholipase C-γ1 in Human Gastric Cancer Tissues

        Phospholipase C (PLC) plays a central role in signal transduction and is important in cellular growth, differentiation and transformation. There are currently ten known mammalian isozymes of PLC reported to dale. However, their biological implication in carcinogenesis and in disease is not fully understood. Here we examined the relative content of various PLC isozymes present in 50 paired surgical specimens from human gastric cancer patients. Western blot analysis demonstrated considerablely higher levels of PLC-γ₁ protein in gastric cancer tissues from 41 of 50 specimens as compared to that of normal mucosa. On immunohistochemistry, distinct speckles or clumps of PLC-γ₁ staining were observed in the cytoplasms of primary malignant lesions as well as the invading cancer cell nests, in contrast to the normal tissues which were negative. Little differences in other isozymes were found. Only PLC-δ₁ was slightly increased in about two thirds of the patients, however, to a much lesser extent. In conclusion, although the causes of increased PLC-γ₁ in gastric carcinomas remain to be explored further, the elevated level of PLC-γ₁ suggests an altered signal transduction pathways in gastric carcinomas and may indicate an important role played by PLC-γ₁ in pathogenesis and proliferation of cancer cells.

      • Differential Translocation of Protein Kinase C ∈ during HeLa Cell Adhesion to a Gelatin Substratum

        The spreading of HeLa cells, following attachment to a collagen or gelatin substratum, requires the activation of protein kinase C (PKC). Membrane-bound PKC was previously shown to be activated during cell attachment and in response to the activation of a series of lipid second messengers turned on by the ligation of β1-integrin collagen receptors. HeLa cells express the α,γ,ε,ζ,λ, and ι isozymes of PKC as determined by Western blotting with specific antibodies. Only PKCε redistributed from the cytosol to the membrane during cell adhesion. Most of the PKCε in cells that were in suspension was in the cytosolic fraction. During cell attachment to a gelatin matrix, all of the PKCε moved out of the cytosol, with most going to the membrane fraction. After the cells became fully spread, PKCε began to reappear in the cytosol. Translocation of PKCε was not observed during the adhesion of cells to culture dishes where cells nonspecifically attach but do not spread. The conventional PKCα and -γ isozymes were translocated from the cytosol to the membrane only when phorbol ester was present at a concentration that increases the rate and extent of cell spreading. Under normal conditions, i.e. in the absence of phorbol ester, PKCε appears to be the PKC isozyme responsible for the regulation of HeLa cell adhesion to the extracellular matrix.

        인터넷 포털산업의 재편

        네이버의 전략적 혁신이 있기 전까지, 선도기업 다음의 서비스 모델은 인터넷 포털 산업에서 성공을 위한 산업규범(industry norm)으로 여겨졌다. 경쟁기업들은 다음의 서비스 모델을 벤치마킹하여 웹메일과 커뮤니티 서비스를 강화하고 배너광고의 수익을 올리는 것에 집중하였다. 그러나 2002년 네이버는 인터넷 포털산업 내에 형성되어 있던 산업규범의 고객, 제품, 운영방식을 재정의하는 전략적 혁신을 단행한다. 그 결과 새로운 수익원인 검색광고를 출시하고, 주요 고객을 대기업에서 중소기업 및 자영업자로 전환할 수 있었다. 또한 변화의 성과를 극대화하기 위해 네이버는 모든 기업의 자원과 역량을 검색서비스를 개선하는데 투자하여 지식검색이라는 새로운 개념의 검색 서비스를 개발하고 네트워크 효과(Network Effect)에 따른 시장 선점 효과를 누리게 된다. 결과적으로 네이버의 전략적 혁신은 인터넷 포털산업의 새로운 규범이 되었으며, 현재 네이버는 시장을 선도하는 기업의 위치에 올라서게 되었다. Before the success of Naver, Daum’s profit model was regarded as the formula for success by other firms in the industry. Daum continued to dominate, and most firms focused on benchmarking the service model to develop web mail services and community service to maximize the price of banner advertisements, their main product. Naver was one of the third tier companies in an industry following such norms. Then in 2002, Naver implemented a strategic innovation to break the prevailing rules of the industry and pursue an economic takeoff. They defined the market in a totally different way, based on “who”, “what” and “how”. By focusing on “search advertisements”, which was a new notion at the time, Naver changed its main customers from big companies to small and private companies. To maximize the performance of its changes, Naver concentrated its resources and capabilities (R&Cs) to strengthen the search engine. The firm developed a new concept of the Internet search service named “Knowledge Search” and benefited from the first mover advantage by gaining in on the network effect. As a result, Naver set a new industry norm and now dominates the Internet portal industry market.

        아급성형 Gaucher 씨 병에서의 동종골수이식과 유세포 분석기를 이용한 Glucocerebrosidase 활성화 추적 1 예

        Gaucher's disease (GD) is the most common inherited lysosomal storage disease, manifested by generalized accumulation of glucocerebroside in macrophages of the reticuloendothelial system due to a deficient lysosomal β-glucocerebrosidase (GC). It is inherited by an autosomal recessive pattern in which three clinical phenotypes have been described based on the presence and severity of neurologic involvement. GD is treated possible by GC enzyme replacement therapy, allogeneic bone marrow transplantation (BMT), and gene therapy. We here report the experience of successful allogeneic BMT in a 16-year-old female patient with GD type III which was demostrated markedly increased Gaucher cells in bone marrow and absence of GC activity in peripheral blood monocytes by FACS using 5'- pentafluorobenzoylaminofluorescein-di-β-D-glucoside (PFBFDGlu) as substrate. Donor marrow engraftment was confirmed by chromosome analysis using microsatellite and by bone marrow examination. Assay of GC activity using FACS revealed normalevel of enzyme activity. She remains alive and well after 12 months of BMT.(Korean J Med 61:195-200, 2001)
