        IL-6-miR-210 Suppresses Regulatory T Cell Function and Promotes Atrial Fibrosis by Targeting Foxp3

        The aim of this study was to explore the role of IL-6-miR-210 in the regulation of Tregs function and atrial fibrosis in atrial fibrillation (AF). The levels of interleukin (IL)-6 and IL-10 in AF patients were detected by using ELISA. Proportions of Treg cells were detected by fluorescence activated cell sorting analysis in AF patients. The expression of Foxp3, α-SMA, collagen I and collagen III were determined by western blot. The atrial mechanocytes were authenticated by vimentin immunostaining. The expression of miR-210 was performed by quantitative real-time polymerase chain reaction (qRT-PCR). TargetScan was used to predict potential targets of miR-210. The cardiomyocyte transverse sections in AF model group were observed by H&E staining. The myocardial filaments were observed by masson staining. The level of IL-6 was highly increased while the level of IL-10 (Tregs) was significantly decreased in AF patients as compared to normal control subjects, and IL-6 suppressed Tregs function and promoted the expression of α-SMA, collagen I and collagen III. Furthermore, miR-210 regulated Tregs function by targeting Foxp3, and IL-6 promoted expression of miR-210 via regulating hypoxia inducible factor-1α (HIF-1α). IL-6-miR-210 suppresses regulatory T cell function and promotes atrial fibrosis by targeting Foxp3.

        Parameter Optimization of the LC filters Based on Multiple Impact Factors for Cascaded H-bridge Dynamic Voltage Restorers

        The cascaded H-Bridge Dynamic Voltage Restorer (DVR) is used for protecting high voltage and large capacity loads from voltage sags. The LC filter in the DVR is needed to eliminate switching ripples, which also provides an accurate tracking feature in a certain frequency range. Therefore, the parameter optimization of the LC filter is especially important. In this paper, the value range functions for the inductance and capacitance in LC filters are discussed. Then, parameter variations under different conditions of voltage sags and power factors are analyzed. In addition, an optimized design method is also proposed with the consideration of multiple impact factors. A detailed optimization procedure is presented, and its validity is demonstrated by simulation and experimental results. Both results show that the proposed method can improve the LC filter design for a cascaded H-Bridge DVR and enhance the performance of the whole system.

        Application of the French Codes to the Pressurized Thermal Shocks Assessment

        The integrity of a reactor pressure vessel (RPV) related to pressurized thermal shocks (PTSs)has been extensively studied. This paper introduces an integrity assessment of an RPVsubjected to a PTS transient based on the French codes. In the USA, the “screening criterion”for maximum allowable embrittlement of RPV material is developed based on theprobabilistic fracture mechanics. However, in the French RCC-M and RSE-M codes, whichare developed based on the deterministic fracture mechanics, there is no “screening criterion”. In this paper, the methodology in the RCC-M and RSE-M codes, which are used forPTS analysis, are firstly discussed. The bases of the French codes are compared with ASMEand FAVOR codes. A case study is also presented. The results show that the method in theRCC-M code that accounts for the influence of cladding on the stress intensity factor (SIF)may be nonconservative. The SIF almost doubles if the weld residual stress is considered. The approaches included in the codes differ in many aspects, which may result in significantdifferences in the assessment results. Therefore, homogenization of the codes inthe long time operation of nuclear power plants is needed.

        Differences in the methanogen community between the nearshore and offshore sediments of the South Yellow Sea

        The differences in methanogen abundance and community composition were investigated between nearshore and offshore sediments in the South Yellow Sea (SYS). Shannon, Simpson, and Chao1 indices revealed a higher diversity of methanogens in the nearshore sediments than in the offshore sediments. The Mann–Whitney U test demonstrated that the relative abundance of Methanococcoides was significantly higher in the offshore sediments, while the relative abundances of Methanogenium, Methanosarcina, Methanosaeta, Methanolinea, and Methanomassiliicoccus were significantly higher in the nearshore sediments (P < 0.05). The abundance of the mcrA gene in the nearshore sediments was significantly higher than that in the offshore sediments. Furthermore, a similar vertical distribution of the methanogen and sulfatereducing bacteria (SRB) abundances was observed in the SYS sediments, implying there is potential cooperation between these two functional microbes in this environment. Finally, total organic carbon (TOC) was significantly correlated with methanogen community composition.

        Research on intelligent launching control of dual clutch transmissions based on adaptive neural fuzzy inference system

        Vehicle launching has an important influence on driving performance of the vehicle. For vehicles with dual clutch transmissions (DCT), the clutch torque control is the key to the launching control. Therefore, a data-driven control method for DCT launching process based on adaptive neural fuzzy inference system (ANFIS) is proposed. Firstly, the vehicle test data during launching process is collected and the optimal clutch torque is obtained based on multi-objective particle swarm optimization (MOPSO). Afterward, to learn the launching control rules from optimization results, the combination of neural network and fuzzy logic algorithm, referred to as an ANFIS, is established. The dataset of the optimized launching clutch torque is utilized to train the ANFIS controller. Finally, the simulation and test results show that the datadriven control can accurately learn the launching control rules from the optimality, thereby achieving the optimal control for different launching intentions.

        Biosensors and chemosensors based on the optical responses of polydiacetylenes

        <P>Polydiacetylenes (PDAs), a family of conjugated polymers, have very unique electrical and optical properties. Upon environmental stimulation, such as by viruses, proteins, DNAs, metal ions, organic molecules <I>etc</I>., the blue PDAs can undergo a colorimetric transition from blue to red, which is accompanied by a fluorescence enhancement. Since the first report on polymerized diacetylene molecules as sensors of influenza virus, the development of efficient sensory systems based on PDAs continues to be of great interest.       • Colorimetric and Fluorometric Assays Based on Conjugated Polydiacetylene Supramolecules for Screening Acetylcholinesterase and Its Inhibitors

        <P>Polydiacetylene supramolecules (PDAs) are unique sensing materials. Upon environmental stimulation, blue PDAs can undergo a colorimetric transition from blue to red accompanied by fluorescence enhancement. In this paper, we report a new PDA system polymerized from a mixed liposome comprising 2-(2-(2-hydroxyethoxy)ethoxy)ethyl pentacosa-10,12-diynoate and pentacosa-10,12-diynoic acid at a 3:7 ratio. The PDA system provided new colorimetric and fluorometric assay methods for screening acetylcholinesterase and its inhibitors through three processes. First, myristoylcholine reacted with PDAs, which then underwent colorimetric and fluorometric transition. Second, acetylcholinesterase catalyzed the hydrolysis of myristoylcholine into tetradecanoic acid, which reduced the myristoylcholine concentration and led to faded color and fluorescence.         Homocysteine-targeting compounds as a new treatment strategy for diabetic wounds via inhibition of the histone methyltransferase SET7/9

        In hypoxia and hyperglycemia, SET7/9 plays an important role in controlling HIF-1α methylation and regulating the transcription of HIF-1α target genes, which are responsible for angiogenesis and wound healing. Here, we report the Ir(III) complex Set7_1a bearing acetonitrile (ACN) ligands as a SET7/9 methyltransferase inhibitor and HIF-1α stabilizer. Interestingly, Set7_1a could engage SET7/9 and strongly inhibit SET7/9 activity, especially after preincubation with homocysteine (Hcy), which is elevated in diabetes. We hypothesize that Set7_1a exchanges ACN subunits for Hcy to disrupt the interaction between SET7/9 and SAM/SAH, which are structurally related to Hcy. Inhibition of SET7/9 methyltransferase activity by Set7_1a led to reduced HIF-1α methylation at the lysine 32 residue, causing increased HIF-1α level and recruitment of HIF-1α target genes that promote angiogenesis, such as VEGF, GLUT1, and EPO, in hypoxia and hyperglycemia. Significantly, Set7_1a improved wound healing in a type 2 diabetic mouse model by activating HIF-1α signaling and downstream proangiogenic factors. To our knowledge, this is the first Hcy-targeting iridium compound shown to be a SET7/9 antagonist that can accelerate diabetic wound healing. More importantly, this study opens a therapeutic avenue for the treatment of diabetic wounds by the inhibition of SET7/9 lysine methyltransferase activity.

        Extended state observer-based finite time position control of rotational shell magazine via higher order sliding mode technique

        An adaptive higher order sliding mode controller based on a novel finite-time extended state observer is designed for rotational shell magazine position tracking control. The design of observer does not need to use the eigenvalue of a specific matrix and thus the observer design can be greatly simplified. Based on the observer, the controller does not rely on the bound of disturbance. The values of controller gains could be reduced and chattering will be restrained. Observation error and tracking error are both finite-time stable. The close-loop stability has been proven via Lyapunov theory. Simulation and experiment results show that compared with twisting controller and proportional-integral controller, the proposed scheme can reduce the overshoot, weaken the chattering and reach the steady state faster in the presence of time-varying disturbance, parameter uncertainties and measurement noise.
