        Theoretical study of UHPCC composite column behaviors under axial compression

        To improve the durability and service life of reinforced concrete column such as bridge piers, an advanced composite column made of Ultra High Performance Cementitious Composites (UHPCC) permanent form is proposed. Based on elasticity plasticity theory, axial compression behavior of the composite column was studied theoretically. The first circumferential cracking load and ultimate limit loading capacity are derived for the composite column. Short composite column compression tests and numerical simulations using FEM method were carried out to justify the theoretical formula. The effects of UHPCC tube thickness on the axial compression behavior were studied. Using the established theoretical model and numerical simulation, the large dimension composite columns are calculated and analyzed with different UHPCC tube thickness. These studies may provide a reference for advanced composite column design and application.

        Structural behaviors of sustainable hybrid columns under compression and flexure

        Structural behaviors of a sustainable hybrid column with the ultra high performance cementitious composites (UHPCC) permanent form under compression and flexure were studied. Critical state and failure stage characters are analyzed for large and small eccentricity cases. A simplified theoretical model is proposed for engineering designs and unified formulas for loading capacity of the hybrid column under compression and flexure loads are derived, including axial force and moment. Non-linear numerical analysis is carried out to verify the theoretical predictions. The theoretical predictions agree well with the numerical results which are verified by the short hybrid column tests recursively. Compared with the traditional reinforced concrete (RC) column, the loading capacity of the sustainable hybrid column is improved significantly due to UHPCC confinements.

        Identification of novel reference genes using sika deer antler transcriptome expression data and their validation for quantitative gene expression analysis

        The most commonly used normalization strategyfor quantitative real-time reverse transcription-polymerasechain reaction (RT-qPCR) is to select a stablereference gene. However, to date, no suitable referencegenes have been identified in sika deer antler tissues. Thus,the aim of this study was to identify the most stable gene ora set of genes to be used as reference genes for RT-qPCRanalysis in sika deer antler tissues. We first selected candidatereference genes using sika deer antler gene expressiondata from an Illumina sequencing platform (Hiseq2000); twenty-one reference genes from the antler tips ofChinese sika deer were selected to test for the normalizationof expression levels during different growth stages. These genes were tested by RT-qPCR and ranked accordingto the stability of their expression using two differentmethods (implemented in geNorm and NormFinder). Based on different algorithms and analytical procedures,our results clearly indicate RPL40 and Gpx as the moststable reference genes of our pool.

        Isolation and Preliminary Identification of a Novel Microorganism Producing Aspartame from Soil Samples

        To develop a new method for producing aspartame (APM), a simple and efficient strategy for the isolation of certain microorganisms producing APM from soil samples was designed. A newly strain with secreting certain specific dipeptidase to directly synthesize APM from the substrates of L-aspartic acid (L-Asp) and Lphenylalanine methyl ester (L-PM) without protection for amino acid side chains was screened from soil samples. APM concentration in reaction mixture was quantified by high performance liquid chromatography (HPLC), yield reached 0.015 g/L. Examination of the general morphological characteristics and data from the sequence analysis of the rDNA-ITS gene led to identification of the isolate as a strain of fungal endophyte spp. The newly isolated strain had a high potential for application in industrial processes for APM production. In particular, this new method was low cost for synthesis of APM during the reaction due to avoiding protection for amino acid side chains and optical resolution of the mixtures. As we known it, this is first report that a newly strain with a high potential for selective synthesis of the APM was isolated from soil.
