      • Macrophage-secreted Exosomes Delivering miRNA-21 Inhibitor can Regulate BGC-823 Cell Proliferation

        Exosomes, membranous nanovesicles, naturally carry bio-macromolecules or miRNA and play impoetant roles in tumor pathogenesis. Here, we showed that macrophages cell-derived exosomes can function as vehicles to deliver exogenous miR-21 inhibitor into BGC-823 gastric cancer cells. Exosomes loaded with miR-21inhibitor significantly increased miR-21 levels in BGC-823, but miR-21inhibitor loaded in exosomes exerted an opposite effect. miRNA transfected with exosomes had less cellular toxicity to host cells compared to conventional transfection methods. The miR-21inhibitor loaded exosomes promoted the migration ability and reduced apoptosis of BGC-823 gastric cancer cells. These observations indicate that miR-21 acts as a tumor promoter by targeting the PDCD4 gene and preventing apoptosis of gastric cancer cells through inhibition of PDCD4 expression. Furthermore, exosome -mediated miR-21 inhibitor delivery resulted in functionally more efficient inhibition and less cellular toxicity compared to conventional transfection methods. Similar approaches could be useful in modification of target biomolecules in vitro and in vivo. These findings contribute to our understanding of the functions of miR-21 and exosomes as a carrier for therapy of gastric cancer.

        Catalytic oxidative desulfurization of benzothiophene with hydrogen peroxide over Fe/AC in a biphasic model diesel-acetonitrile system

        Catalytic oxidative desulfurization (Cat-ODS) of benzothiophene (BT) in n-octane has been investigated with hydrogen peroxide (H2O2) over catalysts of activated carbon (AC) supported iron oxide under mild conditions. The catalyst was characterized by N2 adsorption, XRD, SEM/EDS, TPR and XPS. Under the best operating condition for the catalytic oxidative desulfurization--temperature 60 oC, atmospheric pressure, 0.15 g Fe/AC, 18 molar ratio of hydrogen peroxide to sulfur, using acetonitrile as extraction solvent for double extraction--the sulfur content in model diesel fuel (MDF) was reduced from 700 ppmw to 30 ppmw with 95.66% of total sulfur was removed.

        Optimal Heterogeneous Distributed Storage Regenerating Code at Minimum Remote-Repair Bandwidth Regenerating Point

        Recently, a product-matrix (PM) framework was proposed to construct optimal regenerating codes for homogeneous distributed storage systems (DSSs). In this paper, we propose an extended PM (EPM) framework for coding of heterogeneous DSSs having different repair bandwidths but identical storage capacities. Based on the EPM framework, an explicit construction of minimum remote-repair bandwidth regenerating (MRBR) codes is presented for a specific heterogeneous DSS, where two geographically different datacenters with associated storage nodes are deployed. The data reconstruction and regeneration properties of the MRBR code are proved strictly. For the purpose of demonstration, an example implementation of MRBR code is provided. The presented MRBR code is the first optimal strict-regenerating code for heterogeneous DSSs. In addition, our proposed EPM framework can be applied to homogeneous systems also.

        SYNTHESIS OF YTTRIA-STABILIZED CUBIC ZIRCONIA NANOCRYSTALS BY ULTRASONIC–MICROWAVE ROUTE

        The yttria-stabilized zirconia (YSZ) nanocrystals with uniform size, high purity, and high degree of crystallinity, were prepared by ultrasonic–microwave-assisted method. The structure, optical properties and morphologies of YSZ nanocrystals were characterized by X-ray powder diffraction (XRD), Raman spectroscopy, UV–vis absorption, scanning electron microscope (SEM) and transmission electron microscopy (TEM). The SEM and TEM images of the YSZ nanocrystals indicate that the product is a mono-dispersion structure with an average particle size of about 25 nm.

        Collaborative Sub-channel Allocation with Power Control in Small Cell Networks

        For enhancing the coverage of wireless networks and increasing the spectrum efficiency, small cell networks (SCNs) are considered to be one of the most prospective schemes. Most of the existing literature on resource allocation among non-cooperative small cell base stations (SBSs) has widely drawn close attention and there are only a small number of the cooperative ideas in SCNs. Based on the motivation, we further investigate the cooperative approach, which is formulated as a coalition formation game with power control algorithm (CFG-PC). First, we formulate the downlink sub-channel resource allocation problem in an SCN as a coalition formation game. Pareto order and utilitarian order are applied to form coalitions respectively. Second, to achieve more availability and efficiency power assignment, we expand and solve the power control using particle swarm optimization (PSO). Finally, with our proposed algorithm, each SBS can cooperatively work and eventually converge to a stable SBS partition. As far as the transmit rate of per SBS and the system rate are concerned respectively, simulation results indicate that our proposed CFG-PC has a significant advantage, relative to a classical coalition formation algorithm and the non-cooperative case.

        OPE molecular junction as a hydrogen gas sensor

        Oligo(phenylene ethynylene) (OPE) molecular junction has been suggested as a H2 molecule sensor based on calculations using the first principles of density–functional theory and non-equilibrium Green's function. The electronic transport properties of the OPE molecule between two Au electrodes with or without adsorbed H2 molecules are investigated. Results show that the adsorbed H2 molecule significantly changes the characteristics of the current–voltage curve of the OPE molecular junction. The pure OPE molecular junction exhibits a significant negative differential resistance, but this kind of phenomenon will disappear or weaken after hydrogen molecules are adsorbed. The conductance of the junction also obviously decreases in the bias range of [−0.4, 0.4] V after adsorbing H2 molecules. These effects can be used to design a H2 molecule sensor.

        Chatter reliability prediction of side milling aero-engine blisk

        Reliability analysis of a dynamic structural system is applied to predict chatter of side milling system for machining blisk. Chatter reliability is defined as the probability of stability for processing. A reliability model of chatter was developed to forecast chatter vibration of side milling, where structure parameters and spindle speed are regarded as random variables and chatter frequency is considered as intermediate variable. The first-order second-moment method was used to work out the side milling system reliability model. Reliability lobe diagram (RLD) was applied to distinguish reliable regions of chatter instead of stability lobe diagram (SLD). One example is used to validate the effectiveness of the proposed method and compare with the Monte Carlo method. The results of the two approaches were consistent. Chatter reliability and RLD could be used to determine the probability of stability of side milling.

      • Serum Tumor Marker Levels might have Little Significance in Evaluating Neoadjuvant Treatment Response in Locally Advanced Breast Cancer

        Background: To determine the potential value of serum tumor markers in predicting pCR (pathological complete response) during neoadjuvant chemotherapy. Materials and Methods: We retrospectively monitored the pro-, mid-, and post-neoadjuvant treatment serum tumor marker concentrations in patients with locally advanced breast cancer (stage II-III) who accepted pre-surgical chemotherapy or chemotherapy in combination with targeted therapy at Fudan University Shanghai Cancer Center between September 2011 and January 2014 and investigated the association of serum tumor marker levels with therapeutic effect. Core needle biopsy samples were assessed using immunohistochemistry (IHC) prior to neoadjuvant treatment to determine hormone receptor, human epidermal growth factor receptor 2(HER2), and proliferation index Ki67 values. In our study, therapeutic response was evaluated by pCR, defined as the disappearance of all invasive cancer cells from excised tissue (including primary lesion and axillary lymph nodes) after completion of chemotherapy. Analysis of variance of repeated measures and receiver operating characteristic (ROC) curves were employed for statistical analysis of the data. Results: A total of 348 patients were recruited in our study after excluding patients with incomplete clinical information. Of these, 106 patients were observed to have acquired pCR status after treatment completion, accounting for approximately 30.5% of study individuals. In addition, 147patients were determined to be Her-2 positive, among whom the pCR rate was 45.6% (69 patients). General linear model analysis (repeated measures analysis of variance) showed that the concentration of cancer antigen (CA) 15-3 increased after neoadjuvant chemotherapy in both pCR and non-pCR groups, and that there were significant differences between the two groups (P=0.008). The areas under the ROC curves (AUCs) of pre-, mid-, and post-treatment CA15-3 concentrations demonstrated low-level predictive value (AUC=0.594, 0.644, 0.621, respectively). No significant differences in carcinoembryonic antigen (CEA) or CA12-5 serum levels were observed between the pCR and non-pCR groups (P=0.196 and 0.693, respectively). No efficient AUC of CEA or CA12-5 concentrations were observed to predict patient response toward neoadjuvant treatment (both less than 0.7), nor were differences between the two groups observed at different time points. We then analyzed the Her-2 positive subset of our cohort. Significant differences in CEA concentrations were identified between the pCR and non-pCR groups (P=0.039), but not in CA15-3 or CA12-5 levels (p=0.092 and 0.89, respectively). None of the ROC curves showed underlying prognostic value, as the AUCs of these three markers were less than 0.7. The ROC-AUCs for the CA12-5 concentrations of inter-and post-neoadjuvant chemotherapy in the estrogen receptor negative HER2 positive subgroup were 0.735 and 0.767, respectively. However, the specificity and sensitivity values were at odds with each other which meant that improving either the sensitivity or specificity would impair the efficiency of the other. Conclusions: Serum tumor markers CA15-3, CA12-5, and CEA might have little clinical significance in predicting neoadjuvant treatment response in locally advanced breast cancer.

        Plasma extracellular vesicle delivery of miR-210-3p by targeting ATG7 to promote sepsis-induced acute lung injury by regulating autophagy and activating inflammation

        Extracellular vesicles (EVs) can be used for intercellular communication by facilitating the transfer of miRNAs from one cell to a recipient cell. MicroRNA (miR)-210-3p is released into the blood during sepsis, inducing cytokine production and promoting leukocyte migration. Thus, the current study aimed to elucidate the role of plasma EVs in delivering miR-210-3p in sepsis-inducedacute lung injury (ALI). Plasma EVs were isolated from septic patients, after which the expression of various inflammatory factors was measured using enzyme-linked immunosorbent assay. Cell viability and apoptosis were measured via cell counting kit-8 and flow cytometry. Transendothelial resistance and fluorescein isothiocyanate fluorescence were used to measure endothelial cellpermeability. Matrigel was used to examine the tubulogenesis of endothelial cells. The targeting relationship between miR-210-3p and ATG7 was assessed by dual-luciferase reporter assays. The expression of ATG7 and autophagy-related genes was determined to examine autophagic activation. A sepsis mouse model was established by cecal ligation and puncture (CLP)-induced surgery. The level of miR-210-3p was highly enriched in septic EVs. MiR-210-3p enhanced THP-1 macrophage inflammation, BEAS-2B cell apoptosis, and HLMVEC permeability while inhibiting angiogenesis and cellular activity. MiR-210-3p overexpression reduced ATG7 and LC3II/LC3I expression and increased P62 expression. Improvements in vascular density and autophagosome formation, increased ATG7 expression, and changes in the ratio of LC3II/LC3I were detected, as well as reduced P62 expression, in adenovirusanti-miR-210-3p treated mice after CLP injury. Taken together, the key findings of the current study demonstrate that plasma EVs carrying miR-210-3p target ATG7 to regulate autophagy and inflammatory activation in a sepsis-induced ALI model.
