        Effect of high-fat diet on the various morphological parameters of the ovary

        Increased food consumption rich in fat and carbohydrate and sedentary lifestyle have seriously increased the rates of obesity and obesity-associated diseases in developed countries. Female mice with diet-induced obesity exhibit infertility and thus can serve as a model for human polycystic ovary syndrome. The aim of the present study was to examine how ovary is affected by diet-induced obesity. The effects of high-fat diet (HFD) on ovary morphology in mice fed with HFD were investigated using unbiased stereological methods. The ovary of mice fed with HFD (n=8, C1090-60, Altromine) for 9 weeks, were compared with that of mice fed with standard chow diet (n=8, C1090-10, Altromine). Stereological parameters were obtained in diestrus cycle. The samples were processed through routine and standard paraffin embedding and were serially sectioned in 5-μm thickness then, every 10th section was saved, stained with Crossman’s triple stain for counting and measuring. In all sampled sections mean follicle numbers, diameters, total ovarian volume cortex to medulla ratio (Vv), ovum to cell ratio in secondary follicle were examined in all sampled sections. The present results showed that weight of ovarian and amount of intraperitoneal adipose tissue and the body weight markedly increased in obese mice when compared with control groups. Moreover, follicle numbers (except primordial follicles) and diameters were significantly increased in obese mice. Cortex to medulla ratio (Vv) and ovum to cell ratio in secondary follicle were also considerably different between experimental and the control groups. The present findings indicate that obesity adversely affects overall ovarian morphology.

        Herein, an algorithm for efficient evaluation of oscillatory Fourier-integrals with Jacobi-Cauchy type singularities is suggested. This method is based on the use of the traditional Clenshaw-Curtis (CC) algorithms in which the given function is approximated by the truncated Chebyshev series, term by term, and the oscillatory factor is approximated by using Bessel function of the first kind. Subsequently, the modified moments are computed efficiently using the numerical steepest descent method or special functions. Furthermore, Algorithm and programming code in MATHEMATICA<sup>®</sup> 9.0 are provided for the implementation of the method for automatic computation on a computer. Finally, selected numerical examples are given in support of our theoretical analysis.

        Protective Effects of the Nuclear Factor Kappa B Inhibitor Pyrrolidine Dithiocarbamate on Experimental Testicular Torsion and Detorsion Injury

        Testicular torsion results with the damage of the testis and it is a surgical emergency. Pyrrolidinedithiocarbamate (PDTC) is a low-molecular-weight antioxidant and potent inhibitor of nuclear factorkappa B (NF-κB) activation. In this study, we aimed to investigate the effects of PDTC to testiculartorsion-detorsion (T/D) injury. Forty adult male Sprague-Dawley rats were separated into four groups. A sham operation was performed in group I. In group II, torsion is performed 2 hours by 720 degreeextravaginally testis. In group III, 4 h reperfusion of the testis was performed after 2 h of testiculartorsion. In group IV, after performing the same surgical procedures as in group III, PDTC (100 mg/kg,intravenous’s) was administered before 30 min of detorsion. The testes tissue malondialdehyde (MDA),superoxide dismutase (SOD) catalase (CAT) level was evaluated. Histological evaluations were performedafter hematoxylin and eosin staining. Testicular tissue MDA levels were the highest in theT/D groups compared with treatment group. Administration of PDTC prevented a further increase inMDA levels. Significant decrease occurred in CAT and SOD levels in treatment group compared withthe control group. The rats in the treatment group had normal testicular architecture. The resultssuggest that PDTC can be a potential protective agent for preventing the biochemical and histologicalchanges related to oxidative stress in testicular injury caused by testis torsion.

      • Expression of Cox-2 and Bcl-2 in Paget's Disease of the Breast

        Background: Paget's disease (PD) is a rare form of intraepithelial adenocarcinoma that involves breast and extramammarian tissues. It is often associated with ductal carcinoma in situ and/or invasive ductal cancer. Molecular pathways that play a role in development of Paget's disease are stil unclear. Expression patterns of Cox-2 and bcl-2 were therefore assessed. Materials and Methods: Patients with a histopathological diagnosis of Paget's disease were included in this study. Patient files were analysed retrospectively. Results: Invasive cancer was diagnosed in 35 (76.1%) of the patients, 7 (15.2%) had ductal carcinoma in situ and 4 (8.7%) patients had no associated neoplasm. Twenty four (52.2%) patients showed COX-2 expression in Paget cells whereas no expression was seen in 22 (47.8%) patients. No relation was found between COX-2 expression and the lesion underlying Paget's disease (p=0.518). Bcl-2 expression in Paget cells was found positive in 12 (26.1%) and negative in 27 (58,7%) cases. There was no relation between Bcl-2 expression and the lesion accompanying Paget's disease (p=0.412). No relation was observed between COX-2 expression and Bcl-2 expression (p=0.389). Conclusions: In breast cancer, COX-2 expression is associated with poor prognostic factors. As COX-2 expression increases the tendency to metastasize also increases. In our study we found a significantly high COX-2 expression in Paget's disease of the breast. We suggest that COX-2 expression and inflammatory processes may play a role in pathogenesis of the Paget's disease of the breast.
