Genes encoding pathogenesis-related proteins,such as degrading enzymes of fungal cell wall polysaccharides,have been used to confer enhanced resistance to fungal pathogens of various plants. A new type of endo-bmannanase gene, amn5A, was isolated from alkaliphilic Bacillus strain (JAMB-602) found in deep-sea sediment. The AMN5A mannanase is active over a wide pH range (pH 7–10) and stable at high temperature. In this study,transgenic tobacco plants expressing the amn5A gene were generated using Agrobacterium-mediated transformation. Polymerase chain reaction (PCR) analysis revealed that the amn5A gene was integrated into the genome of transgenic tobacco plants. Southern blot analysis showed that transgenic plants contained 1–6 copies of amn5A transgenes in their genome. Expression of the amn5A transgene was confirmed by reverse transcription-PCR analysis. Leaf extracts from the transgenic plants showed degradation activity of Konjak mannan. Antifungal assay of detached leaves and in vitro whole plantlets indicated that transgenic plants expressing amn5A gene acquired enhanced resistance to the soil-borne pathogenic fungus, Fusarium oxysporum,compared to untransformed control plants.

Various disorders cause hyperammonemia during childhood. Amongthem are those caused by inherited defects in urea synthesis and related metabolic pathways. These disorders can be grouped into two types: disorders of the enzymes that comprise the urea cycle, and disorders of the transporters or metabolites of theamino acids related to the urea cycle. Principal clinical features of these disorders are caused by elevated levels of blood ammonium. Additional disease-specific symptoms are related to the particular metabolic defect. These specific clinical manifestations are often due to an excess or lack of specific amino acids. Treatment of urea cycle disorders and related metabolic diseases consists of nutritional restriction of proteins, administration of specific amino acids, and use of alternative pathways for discarding excess nitrogen. Although combinations of these treatments are extensively employed, the prognosis of severe cases remains unsatisfactory. Liver transplantation is one alternative for which a better prognosis is reported.

We investigated the effects of various concentrations of ${\beta}$-hydroxybutyrate (BHB, 0, 0.1, 1 and 10 mM), a ketone body, added to chemically-defined maturation medium with or without energy substrates (glucose, pyruvate and lactate) on nuclear maturation rates up to the metaphase stage of the second meiotic division (M-II stage). In addition, we also assessed the influence of BHB on glutathione content, sperm penetration rate and embryonic development up to the blastocyst stage of oocytes matured under the presence of these energy substrates. Nuclear maturation rates up to the M-II stage of oocytes matured with BHB in each concentration group did not show a significant increase compared with the control (0 mM) groups in both the presence and absence of energy substrates. Although glutathione contents were not significantly different in each BHB concentration group, the sperm penetration rate in the 1 mM BHB group was significantly higher (p<0.05) and the embryonic development rate of oocytes up to the blastocyst stage was significantly lower (p<0.05) than the respective values of the control groups. These results suggest that BHB added to a chemically-defined maturation medium may stimulate sperm penetration while inhibiting embryonic development of porcine oocytes.

This article reports on a two-year study on the in-school experiences of 10 ethnically and generationally diverse Asian American LGBQ-identified adolescents who attended a racially integrated urban public high school in the Midwest. The conceptual framework draws on Queer of Colour Analysis (QOCA) to situate how heteronormative, heterosexist, and racially exclusionary practices impacted the youth in distinct ways due to the intersections of their gendered and racialized identities. Three major findings are discussed as follows: (a) the participants’ experiences with violence-based encounters at school including the complex reasons for their general reluctance to formally report these incidents, (b) gendered and racialized forms of heterosexism and homophobia, and (c) how the school’s White-dominated Gay-Straight Alliance (GSA) club because a site of erasure and exclusion. Specifically, their lived realities at school elevate the urgency of issues pertaining to school culture and climate as it relates to their personal safety, security, and well-being. The implications section will reflect on the ways by which K-12 schools could create more equitable, inclusive, and welcoming spaces for racially diverse LGBQ students.

This paper discusses the future of journalism, including social-media use. The findings within this paper are based on the results of the "Social Survey on Social Media Use in the Great East Japan Earthquake." The author conducted this Internet-based survey in June 2012, and the results discussed herein are based on a sample size of 1,000 persons. The main findings suggest that social-media use during and immediately following the March 11, 2011 earthquake in Japan has been developing in a mutually complementary manner with traditional media.
