        Effect of Anthonotha macrophyla (P. Beauv) leaf extract on carrageenan-induced paw oedema, oxidative stress makers and hyperalgesia in murine models

        Anthonotha macrophylla leaf (AML) is employed in the management of pain, inflammation, and some diseases associated with oxidative stress in African traditional medicine. This study investigated the anti-inflammatory and analgesic activities, the acute toxicity and anti-oxidant properties of AML, characterised the phytochemical constituents, and quantified its total phenolic and flavonoid contents. Anti-inflammatory and analgesic activities of AML extract were evaluated using carrageenan-induced oedema assay in rats’ paw and acetic acid-induced writhing assays respectively. Anti-oxidant effect of the extract was assessed in vivo by measuring liver antioxidant enzymes and in vitro by using DPPH radical scavenging assay and the acute toxicity of the extract was investigated in vivo in mice. The phytochemicals were characterised using basic phytochemical screening and total phenolics and flavonoids were quantified using established methods. The AML extract exhibited significantly high anti-inflammatory activity and moderate analgesic activity compared to diclofenac sodium. The extract exhibited moderate DPPH scavenging activity, significantly increased Superoxide Dismutase (SOD) and Glutathione peroxidase (GPX) activities, and did not have any effect on lipid peroxidation. AML was found to contain high phenolic and low flavonoids. The LD50 of the extract was above 5000 mg/kg body weight. In conclusion, the AML extract possessed significant anti-inflammatory and moderate analgesic activities, which may be mediated through its strong antioxidant properties due to its phenolic contents. The LD50 value showed that the extract was safe in the short-term utilization. The extract may therefore serve as a source of anti-inflammatory, analgesic and antioxidant agents.

        Endocuff-Assisted Colonoscopy—A Novel Accessory in Improving Adenoma Detection Rate: A Review of the Literature

        Endocuff (Arc Medical Design) is a U.S. Food and Drug Administration-approved device that is attached like a cap to the distal tip of thecolonoscope; it is used to improve adenoma detection rates during colonoscopy. The aim of this review was to summarize and evaluatethe clinical and technical effcacy of Endocuff in improving adenoma detection rate. A comprehensive literature review was performed toidentify studies describing this technique. In this review article, we have summarized case series and reports describing Endocuff use andresults. The reported indications, results, limitations, and complications are discussed.

        In Vitro Activity of Extract and Fractions of Natural Cocoa Powder on Plasmodium falciparum

        Several flavonoids isolated from certain plants have demonstrated antiplasmodial activity, after their initial indigenous use in malaria treatment. Cocoa has been found to be a rich food source of flavonoids in comparison with many common foods and beverages. The aim of this work was to investigate the in vitro activity of natural cocoa powder on the growth of Plasmodium falciparum. Prepared crude methanol extract was partitioned successively with petroleum ether, ethyl acetate,chloroform, and butanol. Total flavonoid concentration in the crude methanol extract and fractions was measured by the AlCl3colorimetric assay. Direct inhibitory activity of the natural cocoa powder was assessed by culturing extract and fractions with P. falciparum in vitro. Greater antiplasmodial activity was observed in nonpolar solvent fractions (chloroform, ethyl acetate, and petroleum ether) compared with polar solvents. The chloroform fraction was most active, with mean – SEM 50% and 90%inhibition concentrations of 48.3 – 0.9 and 417 – 7.8 lg/mL, respectively. The study showed a weak association between total flavonoid concentration and antiplasmodial activity. Early trophozoite (ring-stage) synchronized cultures treated with the chloroform fraction of natural cocoa powder showed a decline in growth. Further reduction in parasitemia was also observed for other erythrocytic stages. These results suggest that natural cocoa powder has measurable direct in vitro inhibitory effect on P. falciparum and support the anecdotal reports of its ability to prevent malaria as a result of regular intake as a beverage.
