        Validation of Combs prognostic scoring system in Indian recurrent glioma patients treated with re-radiation

        Purpose: Retrospective audit of recurrent glioma patients treated by different fractionation schedules and to validate the modified Combs prognostic score in Indian patient cohort. Materials and Methods: Between Jan 2009 and June 2022, 66 recurrent gliomas patients treated with standard adjuvant treatment—radiation (RT) ± temozolomide (chemotherapy)—and re-treated with RT (± chemotherapy) were categorized as per modified Combs prognostic criteria and outcomes were compared. Results: Sixty-six patients with recurrent gliomas who received reirradiation (re-RT) were audited—53% males; 61% Karnofsky performance status (KPS) ≥80 at time of re-RT; median age 41.5 years (range, 6 to 70 years); 67% <50 years; primary histology low-grade glioma in 33% ; grade III 27%, grade IV 40%; initial median dose of 60 Gy equivalent dose in 2 Gy fractions (EQD2); maximum safe resection at recurrence 41%; mean and median follow-up 78 ± 51 months and 66 months. Mean time interval between RT was 46.4 ± 39 months. Mean planning target volume (PTV) volume in conventional RT (Conv-RT), hypofractionated RT (Hypo-RT), and ultra-hypofractionated RT (UF-RT) was 226.1 ± 140.7 mL, 162.8 ± 123.3 mL, and 143.3 ± 145.8 mL. Mean dose for Conv-RT, Hypo-RT, and UF-RT was 50 Gy (range, 40 to 60), 31 Gy (range, 20 to 40), and 20 Gy (range, 10 to 30). Mean overall survival (OS) in Conv-RT, Hypo-RT, and UF-RT cohort was 18.8 months (range, 2.4 to 76.8); 6.6 months (range, 2 to 17.4), and 13.9 months (range, 3 to 131.9). Median OS as per Combs criteria were 16.6 months (Group a), 24.6 months (Group b), 4.6 months (Group c), and 3 months (Group d). Significant survival benefit was with good KPS score (KPS >80 vs. <80; 20.46 vs. 5.25 months; p < 0.001), patients receiving salvage chemotherapy (20.46 vs. 6.96 months; p = 0.001), and patients received re-RT biological equivalent dose (BED3) >80 Gy (16.62 vs. 5.48 months; p = 0.03). Median overall survival (OS) in our patient cohort and Combs cohort in Group a was 16.6 and 19.5 months; Group b was 24.6 and 11.3 months; Group c was 4.7 and 8.1 months, and Group d was 2 and 5.5 months, respectively. Six months survival in our patient cohort and Combs cohort in Groups a, b, c, d were 100%, 92%, 34%, 17% and 94%, 79%, 70%, 41%, respectively. Twelve months survival in our patient cohort and Combs cohort in Groups a, b, c, d were 88%, 74%, 22%, 0% and 88%, 47%, 22%, 7%, respectively. Conclusion: Modified Combs prognostic factors predicts OS and is applicable in Indian subcontinent patient population.

      • Evidence of Multimeric Forms of HSP70 with Phosphorylation on Serine and Tyrosine Residues - Implications for Roles of HSP70 in Detection of GI Cancers

        Background: Heat-shock protein70 (HSP70) are intracellular protein chaperones, with emerging evidence of their association with various diseases. We have previously reported significantly elevated plasma-HSP70 (pHSP70) in pancreatic cancer. Current methods of pHSP70 isolation are ELISA-based which lack specificity due to cross-reactivity by similarities in the amino-acid sequence in regions of the protein backbone resulting in overestimated HSP70 value. Materials and Methods: This study was undertaken to develop a methodology to capture all isoforms of pHSP70, while further defining their tyrosine and serine phosphorylation status. Results: The methodology included gel electrophoresis on centrifuged supernatant obtained from plasma incubated with HSP70 antibody-coupled beads. After blocking non-specific binding sites, blots were immunostained with monoclonal-antibody specific for human-HSP70, phosphoserine and phosphotyrosine. Conclusions: Our novel immunocapture approach has distinct advantages over the commercially available methods of pHSP70 quantification by allowing isolation of molecular aggregates of HSP70 with additional ability to precisely distinguish phosphorylation state of HSP70 molecules at serine and tyrosine residues.

      • Control of a shimmy vibration in vehicle steering system using a magneto-rheological damper

        <P>Vehicle stability largely depends on the vibration of the steering system. A four degrees of freedom dynamic model of an automotive steering system with a magneto-rheological damper is presented in this study. Firstly, an equivalent mathematical model of the steering system is developed. The nonlinear equation of motion obtained from the dynamic model is then linearized around its equilibrium point to make it suitable for the design of an appropriate controller for vibration suppression. In this work, a new type of adaptive sliding mode controller is designed for control of the magneto-rheological damper and hence to control unwanted vibration. It is shown that the proposed control logic is very effective for settling steering motion near the equilibrium position. The shimmy vibrations of the wheels are reduced by a considerable amount and the steering system becomes stable. In addition, a comparative work is undertaken between the proposed controller and an ordinary sliding mode controller to demonstrate the advantage of the proposed methodology.</P>

        Evaluation of turbulence models in rough-wall boundary layers for hydroelectric applications

        The accuracy of turbulence models for the Reynolds-Averaged Navier-Stokes (RANS) equations in rough-wall flows is evaluated using data from large-eddy simulations (LES) of boundary layers with favourable and adverse pressure gradients. Some features of the flow (such as flow reversal in the roughness sublayer) cannot be captured accurately by any model, due to the fundamental model formulation. In mild pressure gradients most RANS models are sufficiently accurate for engineering applications, but if strong favourable or adverse pressure gradients are applied (especially those leading to separation) the model performance rapidly degrades.

        Grey Wolf Optimizer Based PID Controller for Speed Control of BLDC Motor

        A BLDC motor is superior to a brushed DC motor, as it replaces the mechanical commutation unit with an electronic one; hence improving the dynamic characteristics, effi ciency and reducing the noise level marginally. Maximum BLDC motor drives use PID controller to control the speed of the machine; because it is simple in structure, relatively cheaper and exhibits good performance. But the main problem associated with PID controller is adjusting its parameters during implementation. In recent works, it has been observed that Particle Swarm Optimization (PSO) technique showed good performance in tuning PID controller. For this purpose, in this article, “Grey Wolf Optimization” (GWO) algorithm is introduced; which is used to optimally tune the PID controller parameters. The objective of this article is to compare the results obtained for tuning of PID controller based on of GWO and PSO technique and a conclusion has been derived that the proposed approach yields superior dynamic performance for BLDC motor.

      • A Nonlinear Acoustic Technique for Crack Detection in Metallic Structures

        <P>A crack detection technique based on nonlinear acoustics is investigated in this study. Acoustic waves at a chosen frequency are generated using an actuating lead zirconate titanate (PZT) transducer, and they travel through the target structure before being received by a sensing PZT wafer. Unlike an undamaged medium, a cracked medium exhibits high acoustic nonlinearity which is manifested as harmonics in the power spectrum of the received signal. Experimental results also indicate that the harmonic components increase nonlinearly in magnitude with increasing amplitude of the input signal. The proposed technique identifies the presence of cracks by looking at the two aforementioned features: harmonics and their nonlinear relationship to the input amplitude. The effectiveness of the technique has been tested on aluminum and steel specimens. The behavior of these nonlinear features as crack propagates in the steel beam has also been studied.</P>

      • Efficacy of some new genearation insecticides and a botanical against mustard aphid and their toxicity to coccinellid predators and foraging honeybees

        <P>Field studies were carried out to evaluate the efficacy of four new generation insecticides along with a botanical against mustard aphid (Lipaphis erysimi Kalt.) and their toxicity to coccinellid beetles and foraging honeybees during 2014-15 at Bangladesh Agricultural Research Institute (BARI), Gazipur, Bangladesh. Buprofezin 40 SC was found to be the most effective against aphid offering the lowest aphid population (1.56/ top10cm central twig) at 7 days after spraying (DAS) which was statistically identical to Diafenthiuron 500SC (1.85/ top10cm central twig). Among the treatments, Azadiractin 1EC appeared to be safest to coccinellid beetles and foraging honeybees because it recorded the highest number of beetle (7.50 /5 plants) and honeybee (9.64 /plot/5 min) population at 7 DAS, although honeybee population did not vary statistically with that of Buprofezin 40 SC and Lufenuron 5EC treated plots. Indoxacarb 145SC was found to be the most toxic against honeybees. However, the highest yield was obtained from Buprofezin 40 SC (1.57 t ha-1) treated plot although this was statistically identical to that Diafenthiuron 500SC (1.52 t ha-1) and Azadiractin 1EC (1.48 t ha-1) treated plots.Bangladesh J.       • <i>In vitro</i> synergistic effect of gentamicin with the anti-inflammatory agent diclofenac against <i>Listeria monocytogenes</i>

        <P>Abstract</P><P>Aims: </P><P>A total of nine <I>Listeria monocytogenes</I> strains (seven serotypes) were studied to ascertain whether the non-steroidal anti-inflammatory drug diclofenac (Dc) used in combination with the conventional antilisterial antibiotic gentamicin (Gm) or ampicillin (Am) synergistically augments the efficacy of the antibiotic <I>in vitro</I>.</P><P>Methods and Results: </P><P>The effect of combination was evaluated by the checkerboard method to obtain a fractional inhibitory concentration (FIC) index followed by kill curves. Dc was synergistic with Gm (FIC 0·37) and there was indifference with Am (FIC 1) against <I>L. monocytogenes</I> ATCC 51774. The magnitude of the differences between killing by a single agent and the combination observed at 24 h was significant (<I>P</I> < 0·05) for Dc plus Gm but not Dc plus Am.</P><P>Conclusions: </P><P>Thus, the ability of extended antibiotic therapy may be improved with the help of this synergistic drug pair in listeriosis.</P><P>Significance and Impact of the Study: </P><P>Such findings may indicate parallel administration of anti-inflammatory and anti listeriosis drugs.</P>

        Influence of Food Allowance on Nutrient Utilization of Oat (Avena sativa) Hay by Goats and Feeding Value of Leftovers from High Levels of Offer in Sheep

        Two experiments were conducted to determine the effect of feeding levels on intake and nutrient utilization of oat (Avena sativa) hay (OH) by goats and potential feeding value of leftovers from high levels of offer in sheep. In experiment 1, the goats (15) were offered OH at three levels of feeding to give leftovers of about 20% (T-1), 35% (T-2) and 50% (T-3) of DM offered. A marked effect of refusal rate of OH on intake and digestibility of nutrients was evident. Allowing selective consumption at higher levels (T-2 and T-3), the intake of DCP and TDN from OH was found not only to meet the maintenance requirement but provided surplus nutrients for moderate production. In experiment 2, Sheep (3) were offered one of the three dietary treatments viz OH, refusals left uneaten by goats in T-2 and T-3 (OHR) and 3% urea treated oat hay refusals (UTR) in a $3{\times}3$ latin square design. Though the potential feeding value of leftovers (OHR and UTR) was lower than OH, the results confirm the possibility of their effective use. It was concluded that feeding above the conventional ad libitum level could be an alternative to improve the performance of ruminants fed feeds like oat hay. The potential impact of the strategy would, however, depend on the effective reutilization of leftovers for feeding other animals.
