      • RFLP법을 이용한 사상체질의 유전적 분석 연구

        In Sasang medicine, humen are classified into four constitutions which are Taevang, Sovang, Taeum and Soum. Depending on each different constitution, the clinical and pharmacological application for the same disease might be different. In this study, genomic DNA of different constitutions(Taeum, Soyang and Soum) were analyzed by Restriction Fragment Length Polymorphism(RFLP) to provide scientific and objective references for Sasang classification. The DNA polymorphism for each constitution detected as differences in the length of DNA fragments, after digestion with restriction enzyme Hae III, was investigated using YNH24 as DNA probe. The allele size of Taeum, Soyang and Soum group detected by YNH24 ranged from 1.3 to 3.8 kb, 1.5 to 3.9 kb and 1.3 to 4.6 kb, respectively. However, the allele size distribution of YNH24 loci of different constitutions was shown to be too variable to be classified as 3 different constitution groups investigated. For further study, it is suggested that the number of each constitution samples for RFLP analysis should be increased and statistical analysis of the allele size distribution should be carried out.

        With a view to examining the habit and method in which old people maintain and improve their health, we consulted 551 men and 444 women who were over 65 years old, and concluded as follows: 1. Habit of eating It's been revealed that most of the old people keep a regular eating hour, control the quantity of meal or eat a little food, and prefer vegetables to meat. 2. Mental health Old people feel the necessity for mental health and most of them (about 80%) keep good mental health without becoming pessimistic. Their chief concenrs have turned out to be about their sons and daughters and their own health and finance. 3. Managing Good Health It has been revelaed that old people do not possess any special kind of secret to keep good health but taking plenty of rest, regular meal, and proper exercise, and many of them (about 60%) have been shown to eat invigorant food that they can easily get around their living area. 4. Physical Functions It has been revealed that many old people are conscious of some symptoms about their parts of body and about to percent of them suffers from neuralgia. Some measures like medical aid especially for old people need to be taken as soon as possible. As we have found that the health-keeping method of old people consists plenty of rest, proper exercise, and appropriate nutrition, we can conclude that the recreatonal and culture facilities and programs for keeping old people in good health should be developed. It is also necessary to study food that contribute to their health and develop ways to get rid of the stresses to which they are exposed in their daily life. Presides, it is natural that government found an administrative department to deal with these matters effectively.

      • 뇌출혈 동물모델에서 육미지황원의 효과 연구

        Intracerebral hemorrhage(ICH) is associated with a considerable proportion of strokes and head injuries. The mechanism of brain cell injury associated with hemorrhage may be different from that due to pure ischemia. Therefore, it is essential that models of intracerebral hemorrhage be developed and well characterized in animal model. Yukmijihwangwon (YM) has been known to reinforce the vital essence and have antioxidant activities. In this study, the protective effects of YM was investigated against ICH in animal models. Adult rats had 2 microliters saline containing 0.5 and 5 unit bacterial collagenase ifused into the right caudate nucleus. It was found out that YM was effective in protecting brain against ICH.

      • 저산소상태에서 육미지황원의 뇌신경세포 보호효과에 대한 연구

        Yukmijinhwangwon(YM) has been known to reinforce the vital essence and have antioxidant activities. This study was designed to examine the inhibitory effects of YM against in vitro hypoxia/reperfusion-induced inflammatory response. We have characterized the production of prostaglandin E₂and arachidonic acid during hypoxia/reperfusion in the human neuroblastoma SK-N-MC and human monocytic macrophage U937 cells and the ingibitory effect of YM on these inflammation-related substance formation has been found out in this study. To investigate inhibition of COX expression by YM during hypoxia in vitro. This result suggested that YM used in this experiment reinforced antiinflammatory potentials and protected cells against hypoxia/reperfusion induced inflammatory response.

      • RAPD(Random Amplified Polymorphic DNA)법을 이용한 한약재의 판별 연구

        Conventionally, identification and classification methods of natural products include the morphological survey and assay of chemical disposition. Using these methods, however, is not satisfying for the precise identification of natural products because they are often variable in the compositions, genomic DNA analysis such as RAPD, RFLP and Amp-FLP can be adopted for this purpose. In this study, various ginsengs and bear gall bladder were tested for the development of genetic identification and classification method. Varieties of ginsengs such as, P. ginseng, P. quinquefolium, P. japonicus and P. notohinseng, were genetically analyzed by RAPD. Also, DNA isolated form Bear blood and gall bladder, Ursus thibetanus, Ursus americanus and Ursus arctors, were analyzed by the same method. The results demonstrated that the identification and classification of bear gall bladder and various ginsengs were possible by RAPD analysis. Therefore, this method was thought to be used as a additional method for the identification of other natural products.

      • 허혈/재관류 세포 손상에서 청폐사간탕의 보호 효과

        Free radicals are though to be the most important cause of the reperfusion injury subsequent to ischemia. The antioxidant status of the tissue affected by ischemia-reperfusion is of great importance for the primary endogenous defense against the free radical induced injury. Therefore, antioxidant therapy has been shown to be beneficial in neurological disorders such as Alzheimer's disease and cerebral ischemia. In this study, the protective effects of Chungpesagan-tang(CST) was investigated against ischemia/reperfusion-induced cytotoxicity in SK-N-MC neuronal cells it was found out that low concentration of CST we highly effective in protecting neuronal cells against ischemia/reperfusion-induced cytotoxicity. The inhibitory effect of CST on malondialdehyde formation during ischemia/reperfusion-induced oxidative stress in SK-N-MC cells showed obvious dose-dependent responses. Also, CST showed relatively high inhibitory activity to xanthine oxidase induced by ischemia/reperfusion environment. Therefore, it is thought that CST has both antioxidant and xanthine oxidase inhibitory effect and can be used for clinical applications for protection of neuronal cells from ischemia/reperfusion injury.

        Characterization of lac^+ gal^+ Strains of Zymomonas mobilis for Ethanol Production from Lactose

        Previously RP1::Tn951 which is a derivative of RP1 containing the lactose transposon Tn951 was introduced into Z. mobilis strain ZM6100, and RP1::Tn951 was integrated into its genome to yield ZM6306. The galactose operon was incorporated into ZM6306 to yield ZM6307 for more efficient utilization of lactose. Batch culture study has been carried out on Z. mobilis strains, ZM6306 (lac^+) and ZM6307 (lac^+, gal^+), which can convert lactose directly to ethanol. Using a medium containing 80g·ℓ^-1 glucose and 40g·ℓ^-1 lactose, it was found that ZM6306 and ZM6307 produced maximum ethanol concentration of 40g·ℓ^-1 and 42g·ℓ^-1,respectively, whereas parent strain ZM6 produced 37g·ℓ^-1.

        Expression of lac and gal operons in Zymomonas mobilis

        Two Zymomonas mobilis strains (ZM63 and ZM6307), containing both lactose and galactose operons, were constructed. β-Galactosidase and galactokinase assays indicated that both operons were expressed in both strains. The transport systems available for lactose uptake by Zymomonas mobilis were investigated using ^14C-labelled lactose. After the outer membrane, which was considered to be a possible barrier to lactose uptake, was disrupted by treatment with EDTA and Ca^2+ ions, some increase in lactose uptake was observed in ZM6306 (lac^+) and ZM6307 (lac^+ gal^+), but not in the parent, ZM6. This suggested that the outer membrane of Zymomonas mobilis acts as a barrier to lactose uptake to some degree, and also that the lactose permease is operational in Zymomonas mobilis.

      • Development of RP-HPLC method for simultaneous determination of docetaxel and curcumin in rat plasma: Validation and stability

        <P>The purpose of the present research was to develop a suitable, simple, precise, accurate, robust, and reproducible RP-HPLC method for a reliable simultaneous quantification of docetaxel (DTX) and curcumin (CCM) in rat plasma samples using paclitaxel (PTX) as an internal standard. The samples were assayed by the Agilent 1260 Infinity HPLC instrument using a Capcell Pak C-8 column (4.6mmx 150 mm, 5 mu m) under isocratic conditions. The mobile phase consisted of acetonitrile and triple distilled water (40/60, v/v) with a flow rate of 1.0 ml/min. The eluent was monitored at 230 nm for simultaneous measurement of curcumin and docetaxel. The method was validated by determining system suitability, selectivity, sensitivity, linearity, inter-day and intra-day precision, accuracy, robustness, and stability in accordance with the guidelines of the United States Food and Drug Administration (FDA). The developed chromatographic method proved to be simple, precise, accurate, robust and reproducible. Moreover, the samples showed stability at room temperature over a period of 48 h.
