Vol.8 No.-
This experiment was conducted to determine a proper medium for callus induction of carrot root and to examine the formation of β-caratene which was the main component of carrot on the treatment of 2,4-dichlorophenoxyacetic acid and kinetin. It could be observed that Murashige and skoog medium containing kinetin 10^(-5)M÷2,4-D 10^(-6)M showed fastest growth rate from 9th to 18th day after inoculation. β-Carotone formation was accelerated in the Murashige and Skoog medium containing higher concentration of 2,4-D but was not influenced on the kinetin concentration.

It is essential to wash foreign materials on the surface of red peppers, because the red peppers are concerned with food. The purpose of this study was to develop a red pepper washer using ultrasonics and vertical rotary brush. To increase the washing rate of the red peppers, the optimum washing condition of the red pepper washer using ultrasonics and vertical rotary brush were tested and analyzed. The results of this study were as follows; 1.There was no significant difference of the absorption rate of red pepper by the washing water temperature and washing method. 2.The reasonable speed of vertical rotary brush was around 100rpm. 3.The optimum washing period of red pepper was two minutes. 4.The water temperature did not affect washing effect. 5.The washer using ultrasonics and vertical rotary brush had the best washing effect.

Type I diabetes is caused by destruction of the pancreatic beta cells responsible for insulin-secretion. It can be induced by the drugs alloxan or streptozotocin (STZ). Both an increase in reaction oxygen free radical species and a decrease in the antioxidant defense mechanism lead to the increase in oxidative stress in diabetes mellitus. Oxygen radicals scavengers are effective in preventing diagetes in animal models. Experiments were conducted by two system. The Sprague-Dawley rats were treated with STZ intravenous injection after oral administrated Betula's extract and quercetin for a week, and were administrated them for a week. In vitro, Betula's extracts and quercetin showed a tendency towards inhibition of free radical oxidation in all. In vivo, Betula's extracts group exhibited suppression morphologic change in pancreatic islet compare quercetin group on light microscopy. The Betula's extract significantly inhibited formation of MDA compared to the STZ-induced diabetic group in pancreas. They also significantly inhibited formation of Phosphatidylcholine hydroperoxide(PCOOH) compared to the STZ-induced diabetic group in pancreas and plasma, which considered defense against destruction in beta cells. The protection by Betula's extracts may be due to scavenging of deleterious and highly reactive free radical which is generated by STZ. In streptozotocin-induced diabetic rats, the effect of Betula's extract are greater than that of quercetin. Based on this result, we concluded that Betula's extract has the ability to effectively suppress in vitro and in vivo oxidation, thereby providing additional evidence for their potential beneficial type I diabetes.

A panel of 65 melon germplasm was used to screen for resistance to Fusarium oxysporum f. sp. melonis (Fom) and Monosporascus cannonballus. The screening test revealed 35 accessions that are highly resistant to Fom race 1 and 11 accessions with high resistance to M. cannonballus. A total of four accessions, ‘K134068’, ‘K133069’, ‘Wondae’ and ‘PI 414723’, showing resistance to both pathogens were selected as candidates for melon rootstock. Yield and quality of fruits harvested from ‘Earl``s elite’ (Muskmelon, Reticulatus Group) grafted onto the selected melon rootstocks were found comparable to or better than those of non-grafted melons. Nearly negligible incidence of fruit fermentation was observed when ‘Homerunstar’ (Honeydew type, Inodorus Group) was grafted with the selected melon rootstocks, unlike when it was grafted onto ’Shintozwa’ (Cucurbita spp.) rootstock. The selected melon accessions with resistance to Fusarium wilt and Monosporascus root rot are considered to be potential and valuable genetic resources for breeding program of melon.

In Korea, root-knot nematodes, Meloidogyne incognita, have caused serious damage to oriental melon production, especially in Gyeongsang province, Korea where oriental melon has been cultivated without fallowness. In this paper, its economic importance has been studied. The effect of pre-plant population levels (PI) of M. incognita at second stage juvenile (J2) on oriental melon was investigated. Thirty days after planting, the length of secondary vines and number of leaves tended to decrease with increase of population density of M. incognita. This indicates the lower Pl of M. incognita, the better growth of oriental melon acquired. The mean number of leaves was 14.6 at the low Pl whereas 12 at high Pl, indicating that the growth of oriental melon was negatively affected by high Pl. The Pl of M. incognita was negatively correlated with stem diameter, leaf area, and ovary length, while positively correlated with the day required for fruit set. The mean number of fruits per plant was 5.8 at low Pl, 5.4 at medium Pl, and 5.0 at high Pl. The mean number of marketable fruits decreased as Pl of M. incognita increased (5.1 at low Pl, 4.7 at medium Pl, and 3.0 at high Pl). The weight and width of fruits were negatively influenced by the increase of Pl levels. An appearance index, a major standard of evaluating fruit quality, decreased when the Pl levels increased but soluble solid contents were not affected by Pl levels.
