Applications of Yeast Flocculation in Biotechnological Processes

A review on the main aspects associated with yeast flocculation and its application in biotechnological processes is presented. This subject is addressed following three main aspects-the basics of yeast flocculation, the development of "new" flocculating yeast strains and bioreactor development. In what concerns the basics of yeast flocculation, the state of the art on the most relevant aspects of mechanism, physiology and genetics of yeast flocculation is reported. The construction of flocculating yeast strains includes not only the recombinant constitutive flocculent brewer's yeast, but also recombinant flocculent yeast for lactose metabolisation and ethanol production. Furthermore, recent work on the heterologous $\beta$-galactosidase production using a recombinant flocculent Saccharomyces cerevisiae is considered. As bioreactors using flocculating yeast cells have particular properties, mainly associated with a high solid phase hold-up, a section dedicated to its operation is presented. Aspects such as bioreactor productivity and culture stability as well as bioreactor hydrodynamics and mass transfer properties of flocculating cell cultures are considered. Finally, the paper concludes describing some of the applications of high cell density flocculating bioreactors and discussing potential new uses of these systems.e systems.
Digital Video Festivals and Mathematics: Changes in the Classroom of the 21st Century

This paper aims to present how video production is impacting the classroom, mathematics students and teachers in Brazil, as well as mathematics knowledge production developed with this media. Digital video Festivals with mathematical content are being implemented in Brazil, locally and nationally. One of them is Mathematics Education Digital Video Festival, organized by the Research Group in Informatics, Other Media and Mathematics Education, GPIMEM. In this century, video is earning space as a pedagogical approach in either face-to-face or distance education, even before the COVID-19 pandemic. Currently, considering social distance, the use of videos became an imposed reality to students and teachers. Students and teachers with different level of experience or age started to realize possible positive effects regarding the reorganization of the classroom by the presence of mathematical videos. However, other aspects provided negative effects: for example, inequalities in homes and mobile access impacted equal opportunity to all. This paper will summarize the use of mathematical video in the last century and discuss what is happening this century. We will revise some research developed, mainly in Brazil, that shows how the classroom does not fit in a parallelepiped model. Internet, videos and software combined are “things” that have agency and are co-participating in learning and teaching mathematics. The organization of festivals of videos have become important not only in Brazil but elsewhere in many formats either nationally or internationally, in a way to create challenges, exhibitions, cultural mix that aim to show mathematics applications to the general public. In particular, this article reports on parts of a qualitative research that investigated participants of the first edition of the referred national festival. As a result, we look for the comprehension of following the discussions of qualitative transformations regarding the use of videos in classrooms and mathematics knowledge production, as this new setting suggests that digital technologies arrived in XXI century to collaborate with learning. This article will be supported by a theoretical perspective on technology based on the notion of humans-with- media. In such a perspective knowledge is seen as being constructed by humans and different media and different artifacts. This article shows how such a perspective may help us to cope with the classroom of the XXI century, including the classroom that during the pandemic include students’ and teachers’ homes.

<▼1><P><B>Summary</B></P><P>Pattern recognition receptors are activated following infection and trigger transcriptional programs important for host defense. Tight regulation of NF-κB activation is critical to avoid detrimental and misbalanced responses. We describe Pickle, a <I>Drosophila</I> nuclear IκB that integrates signaling inputs from both the Imd and Toll pathways by skewing the transcriptional output of the NF-κB dimer repertoire. Pickle interacts with the NF-κB protein Relish and the histone deacetylase dHDAC1, selectively repressing Relish homodimers while leaving other NF-κB dimer combinations unscathed. Pickle’s ability to selectively inhibit Relish homodimer activity contributes to proper host immunity and organismal health. Although loss of <I>pickle</I> results in hyper-induction of Relish target genes and improved host resistance to pathogenic bacteria in the short term, chronic inactivation of <I>pickle</I> causes loss of immune tolerance and shortened lifespan. Endoscopy Timing in Patients with Acute Upper Gastrointestinal Bleeding

Background/Aims: The role of very early (≤12 hours) endoscopy in nonvariceal upper gastrointestinal bleeding is controversial. We aimed to compare results of very early and early (12–24 hours) endoscopy in patients with upper gastrointestinal bleedingdemonstrating low-risk versus high-risk features and nonvariceal versus variceal bleeding. Methods: This retrospective study included patients with nonvariceal and variceal upper gastrointestinal bleeding. The primaryoutcome was a composite of inpatient death, rebleeding, or need for surgery or intensive care unit admission. Endoscopy timing wasdefined as very early and early. We performed the analysis in two subgroups: (1) high-risk vs. low-risk patients and (2) variceal vs. nonvariceal bleeding. Results: A total of 102 patients were included, of whom 59.8% underwent urgent endoscopy. Patients who underwent very earlyendoscopy received endoscopic therapy more frequently (p=0.001), but there was no improvement in other clinical outcomes. Furthermore, patients at low risk and with nonvariceal bleeding who underwent very early endoscopy had a higher risk of thecomposite outcome. Conclusions: Very early endoscopy does not seem to be associated with improved clinical outcomes and may lead to poorer outcomesin specific populations with upper gastrointestinal bleeding. The actual benefit of very early endoscopy remains controversial andshould be further clarified.

Biofilm formation and antibiotic resistance are important determinants for bacterial pathogenicity. Ribonucleases control RNA degradation and there is increasing evidence that they have an important role in virulence mechanisms. In this report, we show that ribonucleases affect susceptibility against ribosome-targeting antibiotics and biofilm formation in Salmonella.

Background/Aims The diagnosis of functional heartburn is important for management, however it stands on fragile pH monitoring variables, ie, acid exposure time varies from day to day and symptoms are often few or absent. Aim of this study was to investigate consistency of the diagnosis of functional heartburn in subsequent days using prolonged wireless pH monitoring and its impact on patients’ outcome. Methods Fifty proton pump inhibitotor refractory patients (11 male, 48 years [range, 38-57 years]) with a diagnosis of functional heartburn according to Rome III in the first 24 hours of wireless pH monitoring were reviewed. pH variables were analysed in the following 24-hour periods to determine if tracings were indicative of diagnosis of non-erosive reflux disease (either acid exposure time > 5% or normal acid exposure time and symptom index ≥ 50%). Outcome was assessed by review of hospital files and/or telephone interview. Results Fifteen out of 50 patients had a pathological acid exposure time after the first day of monitoring (10 in the second day and 5 in subsequent days), which changed their diagnosis from functional heartburn to non-erosive reflux disease. Fifty-four percent of non-erosive reflux disease vs 11% of functional heartburn patients (P < 0.003) increased the dose of proton pump inhibitors or underwent fundoplication after the pH test. Outcome was positive in 77% of non-erosive reflux disease vs 43% of functional heartburn patients (P < 0.05). Conclusions One-third of patients classified as functional heartburn at 24-hour pH-monitoring can be re-classified as non-erosive reflux disease after a more prolonged pH recording period. This observation has a positive impact on patients’ management. (J Neurogastroenterol Motil 2015;21:265-272)
