Effect of feeding fresh Italian ryegrass, oat forage and corn silage were compared for milk yield, butter fat percentage and milk components during winter period. Nine milking Holstein cows at mid lactation stage were used in the experiment.

This paper proposes a measurement method for the surface of micro-parts by using AFM(Atomic Force Microscope). To this end, two techniques are presented to extend the capacity of AFM. First, the measurement range is extended by using an image matching method based on correlation coefficients. To account for the inaccuracy of the coarse stage implemented in AFM's, the image matching technique is applied to two neighboring images intentionally overlapped with each other. Second, a method to measure the shape of relatively large specimen is presented by using the inherent trigger mechanism due to the atomic force. The proposed method is proved effective through a series of experiments.

Sequential activation and inactivation of cell cycle-dependent kinases(cdk) ensure orderly progression of the cell cycle. In this study, we demonstrate that cdk2 regulates cdk1 and the G2 checkpoint through phosphorylation of the transcription factor, NF-Y. Cdk2, but not cdk1 or cdk4, phosphorylates two serine residues near the DNA-binding domain in the YA subunit of NF-Y during S and G2 phases of the cell cycle. This phosphorylation appears to be essential for the DNA binding activity of NF-Y and the expression of NF-Y targets, including the S- and G2specific genes, and consequently for cell cycle progression at both G1 and G2/M. DNA damage leads to a decrease in YA phosphorylation and NF-Y target gene expression in the presence of p21 gene. Our results facilitate the elucidation of the molecular mechanism employed by the p21-cdk2 pathway to regulate cdk1 activity throughout the cell cycle, and at G1 and G2 checkpoint arrests following DNA damage.
Genetic relationship between the SPT3 gene and ARS/cAMP pathway in yeast cell cycle control

The signal transduction pathways through the RAS gene product and adenyl cyclease play a critical role in regulation of the cell cycle in yeast, Saccharomyces cerevisiae. We examined the genetic relationship between the spt3 gene and ras/cAMP pathway. A mutation in the SPT3 gene suppressed cell cycle arrest at the G1 phase caused by either an inactivation of the RAS or CYR1 gene which encodes a yeast homologue of human ras proto-oncogene or adenyl cyclase, respectively. The phenotypes such as sporulation and heat shock resistancy, that resulted from a partial inactivation of the RAS or CYR1 genes, were also suppressed by the spt3 mutation. Expression of the SSA1 gene encoding one of th heat shock proteins (Hsp70) can be induced by heat shock or nitrogen starvation. Expression of this gene is derepressed in cry1-2 and spt3 mutants. The bcy 1 mutation repressed by the bcy1 mutation, but not in spt3 mutants. These results suggest that the SPT gene is involved in expression of genes that are affected by the RAS/cAMP pathway.
Regulatory role of p53 tumor suppressor gene in the cell cycle G2 check point and celluar senescence

Here, we show that senescence with specific associated markers can be induced by expression of wild-type p53 in human cancer cells lacking function of this gene. p53-induced senescence caused irreversible cell cycle arrest at G1 and G2/M, and the associated repression of the mitotic cyclins and telomerase activity. These finding demonstrate that wild type p53 can influence the intrinsic senescence program as well as the overlapping pathways of cellular transformation and programmed cell death.

The p53 tumor suppressor gene has been shown to play an important role in determining cell fate. Overexpression of wild-type p53 in tumor cells has been shown to lead to growth arrest or apoptosis. Previous studies in fibroblasts have provided indirect evidence for a link between p53 and senescence. Here we show, using an inducible p53 expression system, that wild-type p53 overexpression in EJ bladder carcinoma cells, which have lost junctional p53, triggers the rapid onset of G1 and G2/M growth arrest associated with p21 up-regulation and reprission of mitotic cyclins (cyclin A and B) and cdc2. Growth arrest in response to p53 induction became irreversible within 38-72 hours, with cells exhibiting morphological features as well as specific biochemical and ultrastructural markers of the senescent phenotype. These findings provide direct evidence that p53 overexpression can activate the rapid onset of senescence in tumor cells.

Two kinds of cDNA encoding mouse GalB1,3(4)GlcNAc α2,3-sialytransferase (mST3Gal III) and GalB1,4(3)GlcNAc α2,3-sialyltransferase (mST3Gal IV) were isolated from mouse brain cDNA library by means of a PCR-based approach. The cDNA sequences included an open reading frame coding for proteins of 374 and 333 amino acids. respectively, and the primary structure of these enzymes suggested a pultative domain structure consisting of four regions, like that in other glycosyltransferases. The deduced amino acid sequences of mST3Gal III and IV showed a 98% and 89% identity with rat ST3Gal III and human ST3Gal IV. respectively. Northern analysis indicated that the expression of mST3Gal III mRNA was abundant in heart. liver and adult brain, while that of mST3Gal IV mRNA was detected in all tissues tested except for testis. but the level was the highest in liver. Soluble forms of mST3Gal III and IV transiently expressed in COS cells exhibited enzyme activity toward acceptor substrates containing the terminal either GalB1.3GlcNAc or GalB1.4GlcNAc sequences. The substrate preferences of both enzymes were stronger for tetrasaccarides than for disaccarides.

Mutations of the tumor-suppressor gene p53 have been found in 30-50% cases of hepatocellular carcinoma (HCC). In the this study, E1-negative adenoviral vector encoding wild-type p53 under the control of the human cytomegalovirus promoter (AdCMV-p53w) was constructed to evaluate its therapeutic efficacy against tumor nodules developing after injection of HuH7 cell lines in the 10 nude mice.
