      • 성별판별을 위한 한국인 성인 방패연골의 판별분석

        방패연골은 목뿔뼈 바로 아래 위치하며 후두부를 구성하는 연골 가운데 가장 큰 연골이다. 본 연구는 한국인 방패연골의 형태분석을 통해 얻은 결과를 이용하여 성별판별을 할 수 있는지 알아보고자 하였다. 20대이상 한국인에서 109(남자69, 여자 40)개의 방패연골을 대상으로 26가지 항목을 계측하여 통계학적으로 검증하여 다음과 같은 결과를 얻었다. 1. 방패연골너비, 방패연골높이, 방패연골길이, 연골판높이 등은 남자의 계측치가 컸다 (p<0.01). 2. 연골판각, 방패연골아래뿔각, 위방패패임각 등은 여자의 게측치가컸다. (p<0.01). 3. 방패연골길이와 연골판높이 항목을 이용하여 남녀판별함수를 얻었다. 이상의 결과에서 한국인 방패연골은 성별에 따라 차이가 있었으며, 20대 이상 한국인의 경우 방패연골의 형태분석에 의해 성별을 판별할 수 있을 것이다.

      • 토끼 눈물샘에서 Nitric Oxide Synthase의 면역조직화학적 연구

        Endogenous nitric oxide(NO) has been known to regulate many physiological and pathological process, especially the salivary secretion and glandular blood flow. However, nitric oxide synthase(NOS) responsible for NO synthesis has not been well studied in rabbit lacrimal gland. The present study was performed to investigate the distribution of nitric oxide synthase isoforms(endothelial, neuronal, and inducible NOS). Immunohistochemistry, using monoclonal mouse anti-endothelial NOS, anti-neuronal NOS, and anti-inducible NOS, was performed in inferior lacrimal of the rabbit. Endothelial NOS(eNOS)-positive immunoreactivities were observed in arterial endothelia, intralobular ducts and interlobular ducts of the inferior lacrimal gland of the rabbit. eNOS-positive immunoreactivities were most prominent in the interlobular and intralobular ducts. Immunoreactivities were well concentrated in cytoplasm of columnar epithelial duct cells. However, weak eNOS-positive immunoreactivity was observed in the intercalated ducts and vascular endothelial cells. Neuronal NOS(nNOS)-positive immunoreactivity was seen in the nerve fibers of inferior lacrimal gland. nNOS-positive immunoreactivy was not detected in duct systems or in acinar cells. Inducible NOS-positive immunoreactivity was not seen. Especially, the expression of the nNOS in the nerves of lacrimal gland suggest that NO may play a role in modulating tear production. These results reveal the presence of eNOS and nNOS in the lacrimal gland of the rabbit, which may be related with regulation of the glandular secretion and blood flow through the gland.

      • 공업용 접착제 증기 흡입이 흰생쥐 기관상피에 미치는 영향 : 조직화학적 및 미세구조적 연구

        Industrial glues, known as Bonds in Korea, contain many kinds of organic solvents, and glue sniffing of youths became one of the social problems in Korea. Mixed exposures to solvents by glue sniffing may induce chronic toxicities different from those induced by exposures to the solvent of single component. In this study, toxicological insult of the inhalation of glue vapor on the primary target organ, the tracheal epithelium of the respiratory system was morphologically worked out. Vapor samples of organic solvents were collected for analysis, and the components found were aceton, n-hexane, methyl cyclopentane, c-hexane, and toluene by gas mas. In order to inhale the organic solvents of adhesives to the mouse, experimental animals were exposed to vapors of the glues(acetone 5212 ppm, n-hexane 139 ppm, c-hexane 3757 ppm, methylcyclopentane 1820 ppm, and toluene 2.2 ppm) in a chamber for 20 min/day for 3, 5, 7, 14 consecutive days, in a whole body inhalation chamber. Control groups were exposed to room air. Animals were sacrificed and tracheal tissues were collected and fixed in 10% neutral formalin for light microscopy and in 2.5% glutaradlehyde - 1.5% paraformaldehyde for transmission electron microscopy and in 2.5% glutaraldehyde for scanning electron microscopy. The results are as follows : 1. In the tracheal epithelium of a normal mouse, ciliated cells, nonciliated cells and basal cells were found, but goblet cells and submucosal glands were not found. 2. In the tracheal mucosa of a normal mouse, alcianophilic bands were detected, but PAS positive bands were not found, but alcian blue-PAS positive cells were found in the mucosae exposed to glue vapor for 7 days and 14days. 3. Distributing of ciliated and nonciliated cells in the epithelium were roughly comparable during experimental periods; the longer the experimental period(2 week), higher the percentage of non-ciliated cells. 4. The mouse of organic solvent inhaled group showed three distinct forms of nonciliated cells, which were apparently transitional types between nonciliated cells and ciliated cells. It is suggested that the role of the nonciliated cell might be a developmental functioning involved in the epithelial cell turnover. Nonciliated cells show a great deal of plasticity; change to ciliated cells during the early phase; and revolving of nonciliated cells during the repopulation phase after injury. The results suggest that ultrastructural morphologic changes in tracheal epithelia may occur following glue sniffing, and those structural changes were analysed as an adaption to the environment.

        The purpose of this study was to examine the ultrastructural characteristic of the normal pylorus mucosa, and their structural changes induced by the ligation of common bile duct of the rabbit. Common bile duct ligation probably causes the dysfunction of the pyloric surface mucous cells that results in delayed mucous formation and secretion, but recovered mucous secretory activity is recovered on the late stages.

      • 5-Fluorouracil이 생쥐 콩팥소체의 미세구조에 미치는 영향

        The experiment was performed to study the morphological responses of the renal glomeruli of the mice after administration of 5-fluorouracil. Healthy adult mice weighing about 25gm each were divided into normal and 5-fluorouracil treated groups. 5-fluorouracil(60mg/kg) were injected subcutaneously to the animals every other day, and animals were sacrificed at 4 days or 7days following the first injections. Pieces of tissues were taken from renal coriticies, prefixed with 2.5% glutaraldehyde- 1.5% paraformaldehyde(0.1M Millonig's phosphate buffter, pH 7.3), and followed by post-fixation with 1% osmium tetroxide(0.1M Millonig's phosphate buffer, pH 7.3). Fixed blocks were dehydrated with alcohol and acetone, embedded in araldite mixture, cut with LKB V-ultrotome. The ultrathin sections stained with uranyl acetate and lead citrate were observed with a JEM 100CX-Ⅱ electron microscope. In the fourth day following the first injection of 5-fluorouracil, components of the renal glomeruli of the mice are looked compact since they were filled with the widened the mesangium, and showed narrowing lumen of glomerular capillaries and of urinary spaces. The cisternae of the rough endoplasmic reticulum of the podocytes were slightly swollen, whereas flattened cistrernae of the Golgi complexes were frequently observed. And in the fourth day after the first injection of 5-fluorouracil, the compact arragement of renal golmeruli of the mice were stil observed, but morphological changes of glomeruli were generally recovered in the seventh day. From the above results, it was concluded that the large amout of 5-fluorouracil(60mg/kg)induces slightly cytotoxic effects of the renal glomeruli, but fine structural changes occurred inthe renal glomeruli after injection of 5-fluorouracil were recovered to normal apperance within 1 week.

        흰쥐 침샘의 Nitric Oxide Synthase에 관한 면역전자현미경적 연구

        산화질소는 생물체내에서 생리적이나 병리학적으로 중요한 역할을 한다고 알려져 있으며, 특히 침샘조직에서 침분비작용과 샘혈류 조절에 중요한 인자의 하나로 관여함이 알려져 있다. 산화질소합성효소 (nitric oxide synthase, NOS)는 동위효소로서 내피산화질소합성효소 (endothelial NOS, eNOS), 신경산화질소합성 효소 (neuronal NOS, nNOS)와 유도산화질소합성효소 (inducible NOS, iNOS)가 있으며, 세포내에서 내인성산화질소를 합성한다고 알려져 있다. 그러나 산화질소합성효소의 세포내 분포에 관한 전자현미경적 연구는 매우 드물며, 흰쥐 침샘에서의 산화질소생산효소(NOS)에 대한 전자현미경적 연구는 없었다. 흰쥐 침샘에서 NOS의 세포내 분포를 규명하기 위하여 면역전자현미경방법을 이용한 금입자표지법을 시행하여 아래와 같은 결과를 얻었다. eNOS에 양성 면역반응을 보이는 구조는 침샘의 분비세포 중장액세포에 있는 전자밀도가 높은 분비과립이었으며, 점액분비세포의 점액분비과립에서는 비교적 약한 면역반응성이 관찰되었다. 즉 턱밑샘과 혀밑샘을 구성하고 있는 두 종류의 분비세포 중장액세포의 분비과립에 금입자가 비교적 많이 표지되었으며, 점액세포의 분비과립에서는 적은 수의 금입자가 관찰되었고, 침샘의 소엽속관(intralobular duct)의 전자밀도가 높은 분비과립에서도 금입자가 표지된 것이 관찰되었다. 귀밑샘에서도 장액세포의 분비과립과 소엽속관의 분비과립에 금입자가 표지되었다. nNOS의 양성 면역반응은 턱밑샘에서 점액세포의 분비과립에서만 약간의 금입자가 관찰되었으며, 턱밑샘, 혀밑샘 및 귀밑샘의 분비세포와 분비관세포에서는 iNOS에 대한 양성 면역반응이 관찰되지 않았다. 흰쥐 침샘에서 산화질소합성효소 중 eNOS는 침샘분비세포의 분비과립에 존재하며, 특히 전자밀도가 높은 장액성분비과립에 주로 분포하고 있으며, 분비관 중에서 소엽속관에도 분포하고 있는 것이 관찰되었으나, 다른 동위효소인 nNOS와 iNOS는 거의 관찰되지 않았다. 산화질소합성효소가 흰쥐 침샘분비세포의 분비과립과 소엽속관의 분비과립에 분포하고 있는 것으로 보아 침샘에서 산화질소가 침의 생산과 분비에 중요한 역할을 담당하는 것으로 생각된다. Immunoelectron microscopic study, using monoclonal mouse anti-endothelial NOS, anti-neuronal NOS, and anti-inducible NOS, was performed in the salivary gland of rat. Endothelial NOS (eNOS)-positive immunoreactivities were most prominent in the secretory granules of serous cells of the salivary gland of the rat. Immunoreactivities were well concentrated on serous secretory granules in the serous cells. However, weak eNOS-positive immunoreactivity was observed in the mucous secretory granules of the mucous cells. Positive endothelial NOS (eNOS) immunoreactivities were most prominent in the secretory granules of intralobular ducts. Ductal secretory granules and acinar serous secretory granules have a similar pattern of labeling as eNOS suggestings. Neural NOS (nNOS)-positive immunoreactivity was not detected in duct systems or in acinar cells. Inducible NOS (iNOS)-positive immunoreactivity was not seen in acinar and ductal cells. These results reveal the presence of eNOS in the salivary gland of the rat, which may be related with regulation of the glandular secretion and blood flow through the gland.

Healthy adult mice weighing about 25gm each were divided into normal and experimental groups. Mitomycin-C(400㎍/kg) was injected subcutaneously to the animals every other day, and animals were sacrificed at 4 days or 6 days following the first injections. Pieces of tissues were taken from renal corticies, prefixed with 2.5% glutaraldegyde- 1.5% paraformaldehyde(0.1M Millonig's phosphate buffer, pH 7.), and followed by post-fixation with 1% osmium tetroxide(0.1M Millonig's phosphate buffer, pH 7.3). Fixed blocks were dehydrated with alcohol and acetone, embedded in araldite mixture, cut with LKB V-ultrotome. The ultratihin sections stained with urany1 acetate and lead cirtrate were observed with a JEM 100CX-Ⅱ electron microscope. In the fourth and seventh day following the first injection of mitomycin C, components of the renal glomeruli of the mice are looked compact since they were filled with the widened the mesangium, and showed narrowing lumen of glomerular capillaries and of urinary spaces. The electron densities of the lamina rara interna and the lamina rara externa were similar to the density of the lamina densa and the expanded lamina rara interna were often seen. and the cisternae of the rough endoplasmic reticulum of the podocytes were slightly swollen, whereas flattened cisternae of the Golgi complexes were frequently observed. From the above results, it was suggested that although small amount of mitomycin C(400ug/Kg) may give damages on the fine structure of the renal glomeruli of the mice.

        김대건 (안드레아) 신부 얼굴복원

        본 작업은 서울대교구 명동성당에서 가툴릭의대에 우리나라 최초의 신부인 김대건 신부의 얼굴복원작 업을 의뢰해 1999년 9월부터 약15개월에 걸쳐 북원작업을 한 것으로 1971년 가툴릭의대 해부학교실에서 조사한 김대건 신부 머리뼈의 직접제측치와 사진 자료를 창조하여 제작하였다 작업의 과정은 머리뼈 계측값이 가장 비슷한 머리뼈를 찾아 복제를 하고 계측치에 맞게 수정한 후, 해부학 실습용 시체에서 직접계측하거나 초음파를 이용하여 간접계측을 한 한국인 얼굴의 표준두께를 적용하여 영굴을 복원하였다. 아울러 한국인에서 조사 한 머리뼈와 얼굴 각 부위의 연관관계자료를 토대로 유토를 이용한 복원작업을 시행하였으며 최종 결과물은 청동을 소재로 흉상을 만들었다. 이와 같은 작업은 법의학분야에서 신원미확인 머리뼈가 발견되었을 때 얼굴복원을 통해 신원을 확인하는 법 의인류학적 연구의 하나로, 한국인의 자료를 토대로 작업한 최초의 영굴복원이며 앞으로 신원확인을 위해 얼굴 을 복원하는 작업에 도움이 될 것으로 생각한다.
