Proceedings of international Symposium on East-West Medicine, Seoul. 44-64, 1999.-Cordyceps sinensis is a major parasitic fungus on larva of Lepidoptera. It is one of the well know fungi used in traditional Chinese medicine for treatment asthma, lung and kideney diseases. Many fungi belonging to the genus Cordyceps have been demonstrated to produce natural products with various biological activities. Reported biological activities of genus cordyceps include:(a)inhibition of DNA and RNA synthesis;(b)enhancement of cell differentiation;(c)restruction of cytoskeleton;(d)inhibition of protein kinase activity;(e)antitumor activity on bladder, colon, lung carcinoma as well as fibroblastoma;(f)inhibition of the infection and revers transciptase activity of human immunodeficiency virus type 1; and(g) inhibition of methylation of nucleic acid. Thus, it is suggested that the extracts of Cordyceps sp. Contain antitumor, antiviral, and many other biological activities. In our previous studies, we proved that C. sinensis contains the antitumor and immunomodulatory agents. In the present studies, effects of C. sinensis on bronchoalveolar lavage fluids(BAL)cells were demonstrated. The crude methanolic extracts of C. sinensis were fractionated into 15 fractions by silica gel column chromatography. Effects of various fraction on BAL cells proliferation and lipopolysaccharide(LPS) stimulated interleukin-1β(IL-8),platelet activating factor(PAF), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α),and interleukin-8(IL-8) production on BAL cells were determined. The result indicated that the CS-19-22 fraction suppressed BAL cells proliferation activated by LPS. The median inhibitory concentration(IC50) was 6㎍/ml. The CS-19-22 fraction also decreased IL-1β,PAF,IL-6, TNF-αand IL-8 production.The results of revers transcription-polymerase chain reaction(RT-PCR) demonstrated that the CS-19-22 fraction did not affect IL-1β,PAF,IL-6, TNF-α,and IL-8 mRNAs expression in BAL cells activated by LPS. By contrast, it inhibited the IL-10 mRNA expression but enhanced IFN-γand IL-12 mRNAs expression in activated BAL cells. Moreover, the CS-19-22 fraction blocked PAF-induced platelet aggregation. It is unlikely that cytotoxicity was involved. Because no cell deths were observable. We hypothesize that inhibitory mechanisms of CS-19-22 on BAL cells proliferation may be related to the impairments of gene expression and production of cytokines in BAL cells. The explanation of C. sinensis antiasthma function may involved the following(1)C.sinenesis contained immunomodulatory agents that stimulated IFN-γand IL-12 expression in TH1cells.(2)Enhanced secretion of IFN-γand IL-12 will inhibit the Tγcells immune responses Ex. Decreasing of IL-6 and IL-10 production.(3) The impairments of cytokines production in TH2 cells suppressed th BAL cells proliferation then more cytokines production Ex.IL-1β,PAF,IL-6,TNF-αand IL-8 in activated BAL cells were decreased. (4)The suppression of TH2cells immune responses will inhibit the growth and differentiation of B cells then IgE production will be decreased. (5)Reduced production of IgE will decreased asthma attack occurring in individuals. However, the results of the present study suggested that C. sinensis may also have acted to treat asthma in part by inhibiting BAL cells proliferation and cytokine gene expression and production. Future, experiments with treatment of animals with asthma with C. sinensis will be necessary to defined whether C. sinensis can reduce asthma injury. Plans are underway for the isolation of pure principal growth inhibitors and their mechanisms of action are subjected for further study.

      • Evaluation of sewage sludge incineration ash as a potential land reclamation material

        <P><B>Abstract</B></P> <P>This study evaluated the potential of utilising sewage sludge incineration ash as a land reclamation material. Toxicity assessment of the leachate of the ash was carried out for both terrestrial and marine organisms. Both the fruit fly <I>Drosophila melanogaster</I> and barnacle <I>Amphibalanus amphitrite</I> showed that both bottom and fly ash leached at liquid-to-solid (L/S) ratio 5 did not substantially affect viabilities. The leachate carried out at L/S 10 was compared to the European Waste Acceptance Criteria and the sewage sludge ashes could be classified as non-hazardous waste. The geotechnical properties of the sewage sludge ash were studied and compared to sand, a conventional land reclamation material, for further evaluation of its potential as a land reclamation material. It was found from direct shear test that both bottom and fly ashes displayed similar and comparable shear strength to that of typical compacted sandy soil based on the range of internal friction angle obtained. However, the consolidation profile of bottom ash was significantly different from sand, while that of fly ash was more similar to sand. Our study showed that the sewage sludge ash has the potential to be used as a land reclamation material.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Toxicity of fresh & aged sewage sludge incineration fly & bottom ash was evaluated. </LI> <LI> Terrestrial (fruit fly) & marine organisms (barnacle) were used for toxicity study. </LI> <LI> Leachates of ashes had no adverse toxicity on both fruit fly and barnacle tested. </LI> <LI> Fly ash (compared to bottom ash) had more similar geotechnical properties to sand. </LI> <LI> This study showed the potential in using sewage sludge ashes as fill material. </LI> </UL> </P>

        Extracting parameters of TMD and primary structure from the combined system responses

        Tuned mass dampers (TMDs) have been a prevalent vibration control device for suppressing excessive vibration because of environmental loadings in contemporary tall buildings since the mid-1970s. A TMD must be tuned to the natural frequency of the primary structure to be effective. In practice, a TMD may be assembled in situ, simultaneously with the building construction. In such a situation, the respective dynamic properties of the TMD device and building cannot be identified to determine the tuning status of the TMD. For this purpose, a methodology was developed to obtain the parameters of the TMD and primary building on the basis of the eigenparameters of any two complex modes of the combined building–TMD system. The theory was derived in state-space to characterize the nonclassical damping feature of the system, and combined with a system identification technique to obtain the system eigenparameters using the acceleration measurements. The proposed procedure was first demonstrated using a numerical verification and then applied to real, experimental data of a large-scale building–TMD system. The results showed that the procedure is capable of identifying the respective parameters of the TMD and primary structure and is applicable in real implementations by using only the acceleration response measurements of the TMD and its located floor.

        Damage identification of isolators in base-isolated torsionally coupled buildings

        This paper deals with the damage assessment for isolators of base-isolated building systems considering the torsion-coupling (TC) effect by establishing damage indices. The damage indices can indicate the reduction in lateral stiffness of the isolator story as explicit formulas in terms of modal parameters. In addition, the damage location, expressed in terms of the estimated damage index and eccentricities before and after damage, is also presented. Numerical analysis shows that the proposed algorithms are applicable for general base-isolated multi-story TC buildings. A procedure from the analysis of seismic response to the implementation of damage indices is demonstrated by using a numerical case. A system identification technique is employed to extract modal parameters from seismic responses of a building. Results show that the proposed indices are capable of detecting the occurrence of damage and preliminarily estimating the location of damaged isolator.

        Viability and Functions of Alginate-microencapsulated Islets Isolated from Neonatal Pigs

        Patients with Type I diabetes mellitus have been treated with porcine insulin for several decades and pigs have recently been deemed an ideal source of microencapsulated islet cells for clinical xenotransplantation. In this study, neonatal pigs were anesthetized and sacrificed prior to a pancreatectomy. Islet cells were isolated from pancreas via collagenase digestion. Islet cells were separated and collected by hand under microscopic guidance. These cells were suspended in 1.4% sodium alginate solution and encapsulated by dropping them into 1.1% calcium chloride solution and in which the round gel in size was 250-400 ${\mu}m$ in diameter. Viability of the microencapsulated islet cells cultured in medium at $37^{\circ}C$ was assessed by MTT assay. Furthermore, insulin released in response to glucose challenge was investigated using an enzyme-linked immunosorbent assay. Secretion of insulin was low in response to the basal glucose solution (4.4 mM) in medium and was significantly higher in response to the high glucose solution (16.7 mM). The viability of microencapsulated islet cells did not differ significantly over a period of 7 days; that is, the increasing pattern of insulin concentration in the culture medium after glucose stimulation interval day was similar throughout the 7 days cultivation. In summary, experimental evidences indicated that the effects of alginate-microencapsulation prolonged survival of the neonatal porcine islets in vitro cultures and the insulin response to glucose of the islets was maintained.

        A novel illumination system design for application in the integrated screen 3D display

        A mini-projector prototype employing a LED light source, a nontelecentric structure, and an LCOS panel for application in the integrated 3D display was fabricated. A seamless image was obtained by tilting an array of mini-projectors. Seamless quality was created by the excellent uniformity of the projection intensity on the mini-projector's screen, which was simulated as 98.34%. Great uniformity can be realized by optimizing the design of the light source and the optics configuration, which is the key to such realization.

        Ischemic postconditioning protects cardiomyocytes against ischemia/reperfusion injury by inducing MIP2

        Cardiomyocytes can resist ischemia/reperfusion (I/R)injury through ischemic postconditioning (IPoC)which is repetitive ischemia induced during the onset of reperfusion. Myocardial ischemic preconditioning up-regulated protein 2 (MIP2) is a member of the WD-40family proteins, we previously showed that MIP2 was up-regulated during ischemic preconditioning (IPC). As IPC and IPoC engaged similar molecular mechanisms in cardioprotection, this study aimed to elucidate whether MIP2 was up-regulated during IPoC and contributed to IPoC-mediated protection against I/R injury. The experiment was conducted on two models,an in vivo open chest rat coronary artery occlusion model and an in vitro model with H9c2 myogenic cells. In both models, 3 groups were constituted and randomly designated as the sham, I/R and IPoC/hypoxia postconditioning (HPoC) groups. In the IPoC group, after 45 min of ischemia, hearts were allowed three cycles of reperfusion/ischemia phases (each of 30 s duration)followed by reperfusion. In the HPoC group, after 6 h of hypoxia, H9c2 cells were subjected to three cycles of 10 minute reoxygenation and 10 minute hypoxia followed by reoxygenation. IPoC significantly reduced the infarct size, plasma level of Lactate dehydrogenase and creatine kinase MB in rats. 12 h after the reperfusion,MIP2 mRNA levels in the IPoC group were 10 folds that of the sham group and 1.4 folds that of the I/R group. Increased expression of MIP2 mRNA and attenuation of apoptosis were similarly observed in the HPoC group in the in vitro model. These effects were blunted by transfection with MIP2 siRNA in the H9c2cells. This study demonstrated that IPoC induced protection was associated with increased expression of MIP2. Both MIP2 overexpression and MIP2 suppression can influence the IPoC induced protection.
