        Investigation of the Phenomenon of Propagated Sensation along the Channels in the Upper Limb Following Administration of Acupuncture and Mock Laser

        Background: Similar to De Qi psychophysical responses, propagated sensation along the channels (PSC) is considered an important phenomenon in traditional Chinese acupuncture. In acupuncture clinical trials, different acupuncture manipulation techniques are used to enhance the propagation of sensation along the channels to facilitate an optimum therapeutic result. Aim: To examine and compare the PSC reported by participants in a clinical trial following the administration of acupuncture and inactive mock laser. Methods: The study was embedded in a two-arm parallel design multicenter, randomized clinical trial, the Tennis Elbow AcupuncturedInternational StudydChina, Hong Kong, Australia, Italy (TEA IS CHAI). Needle sensations were measured using a validated instrument, the Massachusetts General Hospital Acupuncture Sensation Spreading Scale. Ninety-six participants with lateral elbow pain were randomly allocated into two groups in a 1:1 ratio; the acupuncture treatment group (n Z 47) and the mock laser control group (n Z 49). Participants in both groups received the intervention at two acupoints, LI10 and LI11, consisting of 2 minutes of either standardized needle manipulation or mock laser at each acupoint with a rest period between each intervention period. Data were collected immediately following the interventions at the first and the ninth session within the clinical trial. Results: Although participants in both groups perceived PSC radiating to similar sites along the upper limb, the frequency of the reported radiation sites among the two intervention groups for both radiation up the limb (p < 0.05) and radiation down the limb (p < 0.001) were statistically significantly different. Among the radiating sensation sites recorded within the two study groups, the sensations were reported as radiating a greater distance down the forearm to the wrist compared to up the arm. Evaluation of PSC across the four study sites revealed a statistically significant difference in frequency of the reported radiation down the limb sites in each study group and radiation up the limb sites only in control group only (p < 0.001). Conclusion: The findings of the study demonstrated that the PSC phenomenon is not just associated with needling but can be perceived when using a mock laser.         Association of FOXJ1 polymorphisms with systemic lupuserythematosus and rheumatoid arthritis in Korean population

        The forkhead-box J1 (FOXJ1) transcription factor could suppress a spontaneous activation of T cells and κBβ that results in re-pression of NF-κB activity. In Foxj1 deficiency mice, systemic autoimune inflamation is quite comon symptom. Therefore, deregulated Foxj1 is suposed to be associated with autoimmune diseases and/or other inflammatory diseases. Previously, we identified that polymorphisms of human FOXJ1 gene (g.460C>T, g.1805G>T and g.3375G>C) are associated with al-lergic rhinitis in a Korean population. In present study, we compared the genotype and allele frequencies of these SNPs between healthy controls and systemic lu-pus erythematosus (SLE) or rheumatoid arthritis (RA) patients. We also investigated the relationships be-twen each genotype and the expression levels of anti- nuclear antibodies in SLE patients, and rheumatoid patients. The frequencies of haplotypes constructed by these FOXJ1 SNPs were compared between con-trols and SLE (or RA) patients. The results of genotype and allele analysis showed that the prevalence of poly-morphism g.3375G>C was associated with the sus-ceptibility of SLE (P = 0.0072 and 0.0042, respectively). But no significant association was found with RA. In the haplotype analysis, however, the main CGG patients (P = 0.048).

      • Interleukin-27 polymorphisms are associated with inflammatory bowel diseases in a Korean population

        <P>Abstract</P><P>Background and Aims: </P><P>The cytokine interleukin (IL)-27 is composed of two subunits, Epstein–Barr virus-induced gene 3 (EBI3) and p28, and IL-27 is a novel IL-12 family member that mediates between the innate and adaptive immune systems. We previously identified four polymorphisms in the human <I>IL-27</I> gene and we suggested that the polymorphism of <I>IL-27</I> is associated with the susceptibility to asthma. IL-27 transcripts are significantly elevated in active Crohn's disease (CD) but not in ulcerative colitis (UC). To determine whether these <I>IL-27</I> single nucleotide polymorphisms are associated with the susceptibility to inflammatory bowel disease (IBD), the genotype and allelic frequencies of the <I>IL-27</I> polymorphisms were analyzed between the IBD patients and the healthy controls.</P><P>Methods: </P><P>Genotype analysis of the <I>IL-27</I> gene was performed by the single-base extension (SBE) method. The haplotype frequencies of <I>IL-27</I> for multiple loci were estimated using the expectation maximization (EM) algorithm.</P><P>Results: </P><P>The genotype frequencies of the g.-964A > G polymorphism in the IBD patients were significantly different from those of the healthy control group (<I>P</I> = 0.001). In both the UC and CD patients, the genotype frequencies of the g.-964A > G polymorphism were also significantly different from the frequencies of the healthy control group (<I>P</I> = 0.009). The frequencies of the AGT and GGT haplotypes were significantly different between the healthy control group and the IBD patient group (<I>P</I> = 0.00004 and 0.021, respectively).</P><P>Conclusion: </P><P>Our results suggest that the g.-964A > G polymorphism of the <I>IL-27</I> gene located on the <I>IBD1</I> locus might be associated with the susceptibility to IBD.</P>

        Foxl2 of the Hong Kong catfish (Clarias fuscus): cDNA cloning, tissue distribution and changes in gene expression towards methyltestosterone, estradiol and letrozole exposure of the fries during gonadal differentiation

        Winged helix/forkhead transcription factor gene 2 (Foxl2) plays a crucial role during early ovarian development in fish. Sex steroids and aromatase inhibitors can regulate gonadal differentiation in teleosts. To address the role of Foxl2 in gonadal differentiation, foxl2 was isolated and characterized from the Hong Kong catfish, Clarias fuscus. Tissue distribution analysis by quantitative real-time PCR (qPCR) showed that the foxl2 mRNA was highest in the ovaries; moderate in the female pituitary and hypothalamus; but weak in the forebrain, liver, testis and male pituitary, with a sexually dimorphic pattern in which there was higher expression overall in females versus males. Treatment of the fries during the period of gonadal differentiation [2–30 days post hatching (dph)] with 17amethyltestosterone (MT), an aromatase inhibitor (letrozole) and 17b-estradiol (E2) affected the expression of foxl2. The expression of foxl2 was highest before gonadal differentiation (12 dph), but decreased significantly after gonadal differentiation (18-30 dph). In addition, MT and letrozole were able to down-regulate foxl2, but E2 up-regulated foxl2 during gonadal morphological differentiation. These results indicate the significant roles of Foxl2 in the gonadal differentiation of C. fuscus.

        Metabolomics reveals abundant flavonoids in edible insect Antheraea pernyi

        The natural flavonoids in foods of plant origin have been well-characterized due to their beneficial biological properties. However, the information regarding the flavonoid compounds in edible insects remains severely limited. In the present study, we used a metabolomics approach to identify the flavonoid compounds in the Chinese oak silkworm, Antheraea pernyi Guérin-Méneville (Lepidoptera: Saturniidae), an traditional edible insect. Our study identified over 200 flavonoid metabolites in the larval midgut of A. pernyi with LC-ESI-MS/MS system. These flavonoid metabolites come from eight subclasses, including flavones (1 0 3), flavonols (34), flavonoids (28), flavanones (20), polyphenols (19), isoflavones (9), anthocyanins (9), and proanthocyanidins (4). The relative content of the flavones is the most abundant, with a value of 36.74% of the total. The top five flavonoid components in A. pernyi are hyperoside, isoquercitroside, tricin 7-O-hexoside, hesperetin 5-O-glucoside and protocatechuic acid, accounting for 51.17% of the total flavonoids. Hyperoside is the most abundant flavonoid compound (18.07% of the total) in A. pernyi. Our findings indicated targeted metabolomics is a useful approach to identify flavonoids in edible insects which contain abundant flavonoids than we already knew.

      • A Saliency Detection Method Based on Global Contrast

        To highlight the saliency object clearly from the foreground, we propose a saliency detection method based on global contrast with cluster. Due to the fact that background pixels usually have similar patches, we use cluster analysis to merge the background regions. By using mean shift filter, the background pixels with similar color level are clustered and the saliency calculation can be decreased a lot. In the method, we use the contrast of color feature with all the other pixels to compute the saliency map. A weight coefficient is utilized to improve the detection accuracy in global contrast differences evaluation. The results of extensive experiments on public dataset show that our method perform well and can highlight the salient object clearly against the other five state-of-the-art methods. Besides, we demonstrate that the applications in image segmentation and fusion with our saliency map can get satisfactory results.

      • Involvement of Cdc25c in Cell Cycle Alteration of a Radioresistant Lung Cancer Cell Line Established with Fractionated Ionizing Radiation

        Cancer patients often suffer from local tumor recurrence after radiation therapy. Cell cycling, an intricate sequence of events which guarantees high genomic fidelity, has been suggested to affect DNA damage responses and eventual radioresistant characteristics of cancer cells. Here, we established a radioresistant lung cancer cell line, A549R, by exposing the parental A549 cells to repeated ${\gamma}$-ray irradiation with a total dose of 60 Gy. The radiosensitivity of A549 and A549R was confirmed using colony formation assays. We then focused on examination of the cell cycle distribution between A549 and A549R and found that the proportion of cells in the radioresistant S phase increased, whereas that in the radiosensitive G1 phase decreased. When A549 and A549R cells were exposed to 4 Gy irradiation the total differences in cell cycle redistribution suggested that G2-M cell cycle arrest plays a predominant role in mediating radioresistance. In order to further explore the possible mechanisms behind the cell cycle related radioresistance, we examined the expression of Cdc25 proteins which orchestrate cell cycle transitions. The results showed that expression of Cdc25c increased accompanied by the decrease of Cdc25a and we proposed that the quantity of Cdc25c, rather than activated Cdc25c or Cdc25a, determines the radioresistance of cells.

        Analysis of the Relationship between MHC-DRB1 Gene Polymorphism and Hydatidosis in Kazakh Sheep

        The objective of this work was to analyze the relationship between ovine major histocompatibility complex (MHC) DRB1 gene polymorphism and genetic resistance to hydatidosis in Kazakh sheep. The Ovar (ovine MHC) class II DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of 702 Kazakh sheep, including 302 sheep with hydatidosis and 400 health controls. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique using five restriction enzymes, i.e., MvaI, HaeIII, SacI, SacII and Hin1I, yielding 14 alleles and 28 genotypes. Comparing the frequency of genotypes in hydatidosis sheep with the control group, it was found that the genotype frequencies of MvaIbc, Hin1Iab, SacIIab, HaeIIIde, HaeIIIdf and HaeIIIdd in control sheep were significantly (p<0.01) higher than in hydatidosis sheep, indicating that a significant correlation existed between these genotypes and resistance to hydatidosis. Genotype frequencies of MvaIbb, SacIIaa, Hin1Ibb and HaeIIIef in sheep with hydatidosis were extremely significantly (p<0.01) higher than in the control group, and the genotype frequency of HaeIIIab was significantly higher (p<0.05), indicating that a marked correlation existed between these genotypes and susceptibility to hydatidosis. By way of analyzing haplotype with these resistant genotypes, the hydatidosis resistant haplotype MvaIbc-SacIIab-Hin1Iab of Kazakh sheep was screened out, and then verified through artificial hydatid infection in sheep. The results indicated that the infection rate of sheep with the resistant haplotype of hydatidosis was significantly lower (p<0.01) than without this resistant haplotype. It showed that the genic haplotype MvaIbc-SacIIab-Hin1Iab of Ovar-DRB1 exon 2 was the resistant haplotype of hydatidosis in Kazakh sheep.

        Stanniocalcin-1 protects bovine intestinal epithelial cells from oxidative stress-induced damage

        Chronic enteritis can produce an excess of reactive oxygenspecies resulting in cellular damage. Stanniocalcin-1(STC-1)reportedly possesses anti-oxidative activity, the aim of thisstudy was to define more clearly the direct contribution ofSTC-1 to anti-oxidative stress in cattle. In this study, primaryintestinal epithelial cells (IECs) were exposed to hydrogenperoxide (H2O2) for different time intervals to mimic chronicenteritis-induced cellular damage. Prior to treatment with 200μM H2O2, the cells were transfected with a recombinantplasmid for 48 h to over-express STC-1. Acridine orange/ethidium bromide (AO/EB) double staining and trypan blueexclusion assays were then performed to measure cell viabilityand apoptosis of the cells, respectively. The expression of STC-1and apoptosis-related proteins in the cells was monitored byreal-time PCR and Western blotting. The results indicated thatboth STC-1 mRNA and protein expression levels positivelycorrelated with the duration of H2O2 treatment. H2O2 damagedthe bovine IECs in a time-dependent manner, and this effectwas attenuated by STC-1 over-expression. Furthermore, overexpressionof STC-1 up-regulated Bcl-2 protein expression andslightly down-regulated caspase-3 production in the damagedcells. Findings from this study suggested that STC-1 plays aprotective role in intestinal cells through an antioxidant mechanism.
