<P>We explored a very interesting gold nanoparticle system—pegylated gold in colloidal solution—and analyzed its uptake by mice colorectal adenocarcinoma CT26 tumor cells and the impact on the cell’s response to x-ray irradiation. We found that exposure to polyethylene glycol (PEG) modified (‘pegylated’) 4.7 ± 2.6 nm gold nanoparticles synthesized by a novel synchrotron-based method enhances the response of CT26 cells to x-ray irradiation. Transmission electron microscopy (TEM) and confocal microscopy revealed that substantial amounts of such nanoparticles are taken up and absorbed by the cells and this conclusion is supported by quantitative induced coupled plasma (ICP) results. Standard tests indicated that the internalized particles are highly biocompatible but strongly enhance the cell damage induced by x-ray irradiation. Synchrotron radiation Fourier transform infrared (SR-FTIR) spectromicroscopy analyzed the chemical aspects of this phenomenon: the appearance of C = O stretching bond spectral features could be used as a marker for cell damage and confirmed the enhancement of the radiation-induced toxicity for cells.</P>
Monacolin K affects lipid metabolism through SIRT1/AMPK pathway in HepG2 cells

Monacolin K is the secondary metabolite isolatedfrom Monascus spp. It is the natural form of lovastatin,which is clinically used to reduce the synthesis of cholesterolby inhibiting 3-hydroxy-3-methylglutaryl coenzyme Areductase. In the present study, monacolin K increased proteinexpression of SIRT1 and phosphorylation level of AMPactivatedprotein kinase (AMPK) in HepG2 cells. Throughactivation of SIRT1/AMPK pathway, monacolin Kincreased phosphorylation of acetyl CoA carboxylase andcaused nuclear translocation of forkhead box O1. The westernblotting results showed that monacolin K increasedexpression of adipose triglyceride lipase but decreasedabundances of fatty acid synthase (FAS) and sterol regulatory element-binding protein 1 (SREBP1). MonacolinK also decreased the intracellular accumulation of lipids asdemonstrated by Oil Red O staining. In addition, theimmunostaining showed that monacolin K prevented thenuclear translocation of SREBP1, indicating the associationwith down-regulation of FAS. All the demonstrated effectsof monacolin K were counteracted by nicotinamide orcompound C, the inhibitors of SIRT1 orAMPK. In summary,monacolin K reduces the lipid content through SIRT1/AMPK pathway in HepG2 cells, which promotes catabolismand inhibits anabolism of lipid.
IgE-Binding Epitope Mapping and Tissue Localization of the Major American Cockroach Allergen Per a 2

Purpose: Cockroaches are the second leading allergen in Taiwan. Sensitization to Per a 2, the major American cockroach allergen, correlates with clinical severity among patients with airway allergy, but there is limited information on IgE epitopes and tissue localization of Per a 2. This study aimed to identify Per a 2 linear IgE-binding epitopes and its distribution in the body of a cockroach. Methods: The cDNA of Per a 2 was used as a template and combined with oligonucleotide primers specific to the target areas with appropriate restriction enzyme sites. Eleven overlapping fragments of Per a 2 covering the whole allergen molecule, except 20 residues of signal peptide, were generated by PCR. Mature Per a 2 and overlapping deletion mutants were affinity-purified and assayed for IgE reactivity by immunoblotting. Three synthetic peptides comprising the B cell epitopes were evaluated by direct binding ELISA. Rabbit anti-Per a 2 antibody was used for immunohistochemistry. Results: Human linear IgE-binding epitopes of Per a 2 were located at the amino acid sequences 57-86, 200-211, and 299-309. There was positive IgE binding to 10 tested Per a 2-allergic sera in 3 synthetic peptides, but none in the controls. Immunostaining revealed that Per a 2 was localized partly in the mouth and midgut of the cockroach, with the most intense staining observed in the hindgut, suggesting that the Per a 2 allergen might be excreted through the feces. Conclusions: Information on the IgE-binding epitope of Per a 2 may be used for designing more specific diagnostic and therapeutic approaches to cockroach allergy.

This work provides comprehensive comparisons of the device characteristics, including the voltage gain, power performance, linearity and noise characteristics, for InAlAs/InGaAs/GaAs metamorphic high electron mobility transistors with respect to different indium contents in the InGaAs channel. Though the device with a high In-ratio InGaAs channel usually demonstrated improved high-frequency characteristics, the kink effects in their narrow-gap channel materials were found to seriously degrade the device performance. On the other hand, the low In-ratio channel device had characteristics advantageous for high-gain and high-linearity applications while the device with a compromised channel composition design was suitable for high-power circuit applications. 왕불유행, 목통, 통초가 산후 생쥐의 유즙분비량과 유즙분비 관련 인자에 미치는 영향

Objectives: The purpose of this study is to investigate the effect of Melandrii Herba (MH), Akebia Quinata Decaisne (AQ), and Tetrapanax Papyriferus (TP) on milk secretion and aquaporin (AQP) expression in lactating mice. Methods: For the experiment, the mice were divided into three groups, which were orally administered MH (2,720 mg/kg), TP (400 mg/kg) and AQ (2,800 mg/kg) extracts respectively for 3 weeks from Day 1 after the birth, compared with the control group (C group), which was administered distilled water. A group consisted of six infantile mice per postpartum mouse. For comparison with the C group, non-pregnant SKH-1 mice were used as the virgin group. Results: 1. When it comes to the immunohistochemical staining for prolactin receptors in the mammary glands, the AQ and MH groups showed a strong immune response to the secretory epithelial cells constituting the mammary alveoli, while the TP group represented a weaker immune response. 2. In the immunohistochemical staining for AQP in the mammary glands, AQP1 showed a strong immune response in the walls of capillaries and venules around the mammary alveoli, and AQP3 in the epithelial cells constituting the mammary alveoli, and AQP5 in some tissues between the mammary alveoli. AQP1 was expressed in the order of TP group>AQ group=C group>MH group, and AQP3 was MH group and AQ group>TP group=C group, and AQP5 was MH group>C group>AQ group and TP group. 3. In the Western blot, AQP1 was expressed in the order of TP group>AQ group>C group>MH group, and AQP3 was MH group>AQ group>C group>TP roup, and AQP5 was MH group>TP Group>C group>AQ group. All of AQP1, 3, 5 expression were significantly higher in the C group than in the Virgin group. Conclusions: The administration of Akebia Quinata Decaisne, Tetrapanax Papyriferus and Melandrii Herba have the effect of improving prolactin levels in postpartum mice and increasing the expression of prolactin receptor and AQPs in the mammary glands, suggesting that lactation might be enhanced by the development of the mammary glands.
Constructing a Conversation Key in Three-Party Communications Environment

This article proposes an efficient, less communication rounds, three-party encrypted key exchange protocol to achieve the authentication requirement. The protocol is provided with (1) no asymmetric encryption algorithm which is adopted to reduce the costs (such as any public-key infrastructure); (2) using pre-shared key to prevent adversaries that masquerade as legal users after guessing attacks; (3) avoiding the variant man-in-the-middle attacks on Diffie-Hellman based protocols; (4) achieving mutual authentication. With these four features, the proposed protocol is suitable for being applied for establishing secure channels between two clients, which are supported with the same trusted server.
