High level of uric acid (hyperuricemia) forms needle shape-monosodium urate (MSU) crystals (tophi) in the joints and surrounding tissue. These crystals produce inflammatory reaction by stimulating macrophages to produce reactive oxygen/nitrogen species and proinflammatory cytokines. The leaves of Hydrangea serrata (Thunb.) Ser., native to the Korean mountains (also known as “San-Soogook”) have been used for many inflammatory disorders in far-east countries including Korea. In the present study, the hot water extract of H. serrata leaves and two compounds, hydrangenol and phyllodulcin isolated from the leaves were tested on the hyperuricemia-related pathogenic processes. The results were as follows. H. serrata leaf extract was shown to contain high levels of polyphenol- and flavonoid-like substances. H. serrata leaf extract and its active components, hydrangenol and phyllodulcin significantly inhibited the activity of xanthine oxidase (a uric acid producer) and productions of nitrogen oxide (NO), IL-1β and TNF-α from the MSU-stimulated murine RAW 264.7 macrophages through inactivation of Iκ-Bα and mitogen-activated protein kinases possibly via removing the reactive oxygen species. These results indicate the H. serrata leaf extract, and the two components could suppress the production of uric acid and generation of inflammation mediators from macrophages, suggesting that Hydrangea serrata (Thunb.) Ser. is a candidate that can be used for treating or preventing the hyperuricemia-induced inflammatory conditions.

Due to the COVID-19 pandemic, the trend of beauty products is widening from “outer beauty (cosmetics)” to “inner beauty (supplements)” including more concerns about their weight management. In addition, the demand for naturally sourced raw ingredients continues to rise in response to the growing “transparency” trend. Hydrangea serrata is the natural herb derived from a native Korean plant and its leaf extract can be considered both “Halal” and “Vegan” because it only used hot water. COSMAXBIO conducted a clinical trial to confirm its efficacy in skin health improvement based on in vitro and in vivo study. By ingesting the H. serrata leaf extract, there was shown to be an increased collagen content in the dermis layer followed by improved skin elasticity and the appearance of wrinkles. Also, the study proved that by increasing the amount of hyaluronic acid and water contents, H. serrata leaf extract ameliorated skin hydration and texture. COSMAXBIO R&I team has also studied H. serrata leaves extract’s efficacy in terms of body fat reduction. It has been confirmed that H. serrata leaf extract induces weight loss by reducing the size of fat cells, suppressing fat accumulation in the liver, and regulating cholesterol and fat metabolism hormones in preclinical study. In addition, according to the results of a microbiome analysis in the high-fat diet animal test, H. serrata leaf extract has the potential to suppress firmicutes and proteobacteria while potentially proliferating bacteroidetes. Furthermore, the clinical trial for weight management proved that H. serrata leaves extract reduced body weight and body fat mass, especially visceral fat mass. Taken together, H. serrata leaf extract acts as an innovative solution with dual claims in skin health and body fat loss. It could be a leading inner beauty product that satisfies consumer’s needs, especially during the COVID-19 era.
