Extracellular agarase produced by the Pseudoalteromonas strain JYBCL 1 is used in a variety of applications in the biotechnology, pharmaceutical, cosmetic,and food industries. The optimization of culture conditions for agarase-producing microbes and agarase activity is thus an important consideration in many industrial applications. In this study, the optimum medium composition and culture conditions for the JYBCL 1 strain were determined using the “one factor at a time” (OFAT) method and a Plackett-Burman design. Optimal cell growth was obtained at a temperature of 25ºC and when 10 g/L tryptone was present in the culture medium. Optimal agarase activity occurred at a temperature of 40ºC and at pH 6. The presence of carbonyl groups in the extracellular agarase hydrolysis products was verified using FT-IR. LC-MS identified the hydrolyzates as neoagarohexaose, neoagarotetraose,and neoagarobiose. The extracellular agarase produced by the JYBCL 1 strain used in this study was identified as β-agarase by 13C-NMR spectroscopy.
8명의 한국인 남자 학생들에서 Sodium Valproate의 생체이용률 연구를 위한 HPLC 분석 방법의 개발

Sodium Valproate levels are frequently monitored in clinical laboratories to assess compliance in epileptic patients and to optimize theraphy. In this study, a simple and accurate high performance liquid chromato-graphic method for valproic acid in plasma was developed to be applicable for the studies on the bioavailability of sodium valproate tablets and the therpeutic drug monitoring of plasma valproic acid level. All plasma samples were added with an internal standard, zinc sulfate solution, and acetonitrile, followed by centrifugation. The supernatants were added with 4-bromophenacyl bromide and were heated at 70℃ for 15min. The derivatization products were extracted with hexane, and the hexane extraction solutions were evaporated to dryness and redissolved in mobile phase. The solution was analyzed using HPLC system. The inter-Day and the intra-Day coefficients of variation ranged from 1.8% to 4.9%, and 3.0% to 4.2%. The simple and accurate HPLC method of analysis of plasma valproic acid seems to be adequate for the studies of bioavailability of sodium valproate tablets and therapeutic drug monitoring of plasma valproic acid level.

Reactive oxygen species (ROS) have been implicated in the path-ogenesis of various diseases, and play an important role in the skeletal muscle damage through strenuous exercise or its aging. To investigate whether H₂O₂, as main precursor of highly reactive free radicals, induces apoptosis in mouse skeletal muscle cells C2C12, 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide(MTT) assay, terminal deoxynuclotidyl transferase -mediated dUTP nick end-labeling (TUNED assay, 4,6-diamidino-2- phenylindole (DAPI) staining, DNA fragmentation assay, reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis were performed. Through morphological and biochemical analyses, it was demonstrated that C2C12 cells treated with H₂O₂ exhibit classical apoptotic features. In addition, it was shown that H₂O₂ induces increases in the levels of bax, and a decrease in the bcl-2 expression. Based on the results, H₂O₂ appears to activate specific intracellular death-related pathways and the induction of apoptosis in skeletal muscle cells.
Flocculation과 Dissolved Air Floatation을 이용한 미세조류 수확 최적화

The harvesting of microalgae is a critical step that precedes biodiesel conversion. The most widely used harvesting technology is flocculation and floatation. In this study, the efficiency of the flocculants aluminum sulfate and poly aluminum chloride were evaluated for harvesting the alga Dunaliella tertiolecta in conjunction with dissolved air floatation. Using the jar test the optimum concentration range for aluminum sulfate was 1.0~1.5 g/L and for poly aluminium chloride, 1.5~2.0 g/L. The degree of coagulation was visualized by microscopy. Further analysis in combination with dissolved air floatation showed that the optimal concentration for aluminum sulfate was 1.1 g/L and for poly aluminum chloride, 1.6 g/L.
