        Knockdown of hsa_circ_0001964 inhibits hepatocellular carcinoma cell proliferation by inactivating PI3K/AKT signaling pathway

        Background Circular RNAs (CircRNAs) play vital roles in the pathogenesis of hepatocellular carcinoma (HCC). However, the exact role and detailed mechanism of hsa_circ_0001964 during HCC carcinogenesis remains unknown. Objective To explore the expression, role and mechanism of hsa_circ_0001964 in HCC. Result The results indicated that the mRNA expression of hsa_circ_0001964 was upregulated in HCC cell lines and tissues, respectively, and hsa_circ_0001964 knockdown inhibited HCC cell proliferation in vitro. Western blot analysis showed that hsa_circ_0001964 knockdown suppressed the activation of the PI3K/AKT signaling pathway and its downstream factors (including c-Jun, c-Myc, and CCND1). Conclusion The results of the present study suggested that hsa_circ_0001964 may correlate with HBV infection, and it promotes HCC carcinogenesis via the PI3K/AKT signaling pathway. The findings of the present study provide important insights into the molecular mechanisms of HCC initiation, which may help to develop new treatment strategies against HCC.

        LncRNA H19/miR-29b-3p/PGRN Axis Promoted Epithelial-Mesenchymal Transition of Colorectal Cancer Cells by Acting on Wnt Signaling

        This investigation was aimed at working out the combined role of lncRNA H19, miR-29b and Wnt signaling in the development of colorectal cancer (CRC). In the aggregate, 185 CRC tissues and corresponding para-carcinoma tissues were gathered. The human CRC cell lines (i.e. HT29, HCT116, SW480 and SW620) and normal colorectal mucosa cell line (NCM460) were also purchased. Si-H19, si-NC, miR-29b-3p mimics, miR-29b-3p inhibitor, si-PGRN and negative control (NC) were, respectively, transfected into the CRC cells. Luciferase reporter plasmids were prepared to evaluate the transduction activity of $Wnt/{\beta}-catenin$ signaling pathway, and dual-luciferase reporter gene assay was arranged to confirm the targeted relationship between H19 and miR-29b-3p, as well as between miR-29b-3p and PGRN. Finally, the proliferative and invasive capacities of CRC cells were appraised through transwell, MTT and scratch assays. As a result, overexpressed H19 and down-expressed miR-29b-3p displayed close associations with the CRC patients' poor prognosis (P < 0.05). Besides, transfection with si-H19, miR-29b-3p mimic or si-PGRN were correlated with elevated E-cadherin expression, decreased snail and vimentin expressions, as well as less-motivated cell proliferation and cell metastasis (P < 0.05). Moreover, H19 was verified to directly target miR-29b-3p based on the luciferase reporter gene assay (P < 0.05), and miR-29b-3p also bound to PGRN in a direct manner (P < 0.05). Finally, addition of LiCl ($Wnt/{\beta}-catenin$ pathway activator) or XAV93920 ($Wnt/{\beta}-catenin$ pathway inhibitor) would cause remarkably altered E-cadherin, c-Myc, vimentin and snail expressions, as well as significantly changed transcriptional activity of ${\beta}-catenin/Tcf$ reporter plasmid (P < 0.05). In conclusion, the lncRNA H19/miR-29b-3p/PGRN/Wnt axis counted a great deal for seeking appropriate diagnostic biomarkers and treatment targets for CRC.

        Initial fault time estimation of rolling element bearing by backtracking strategy, improved VMD and infogram

        Rolling bearing failure is widely regarded as a failure form of industrial machines. Owing to the poor operating circumstance with the stochastic contact between rolling elements, the performance of the bearing will deteriorate over time and cause a cascade breakdown in the mechanical system. Early fault detection has been found to be an effective strategy to avoid economic loss. Therefore, an integration method for fault diagnosis that combines backtracking strategy, improved variational mode decomposition (VMD), and infogram is proposed to tackle the challenge of the early feature extraction from the heavy noisy non-stationary signal. The backtracking strategy is adopted to track the data sample points earlier than the fault threshold determined based on the kurtosis index. The optimum parameters α and K of VMD are acquired through the particle swarm optimization (PSO) algorithm. In this way, the more accurate intrinsic mode functions (IMFs) can be gained by the improved VMD. The optimum IMFs are acquired according to the maximum values of kurtosis and correlation coefficients, and these IMFs can be reconstructed into the noise reduction signal. Since envelope analysis requires the selection of the appropriate central frequency and bandwidth, infogram is utilized to select the values of them. A simulated case is applied to demonstrate the validation of the proposed method. And to further illustrate its practicality, it is employed to perform early fault diagnosis for an experimental case. According to the diagnosis results, the proposed method has conspicuous superiority over the other existing technologies for estimating incipient fault time of the bearing.

        Overview of CSNS tantalum cladded tungsten solid Target-1 and Target-2

        A solid tungsten target was used at the China Spallation Neutron Source (CSNS) with 100 kW protonbeam power. To improve the lifetime, hot isostatic pressing (HIP) process was selected to bond tantalumcladding with tungsten plates. Radioactive isotope 182Ta, an activation product of tantalum, was found inthe cooling water after a period of operation, however, no radioactive isotopes of 187W was found, whichshows the tantalum layer remained mostly intact. The CSNS Target-1 had been operating safely for threeyears and was replaced by Target-2 in August 2020

        Spreading behaviors of high-viscous nanofluid droplets impact on solid surfaces

        In this work, the impact dynamics of high-viscous nanofluid droplets onto a solid surface has been investigated experimentally by means of high-speed camera visualization technique. We dispersed various nanoparticles (multiwall carbon nanotube (MWCNT), nano-graphene, and nano-graphite powder) into highviscous base fluid (epoxy resin) to obtain the stable and homogenous nanofluids without surfactant additives. The well dispersed nanofluids show different degree of shear-thinning behaviors, and the shear-thinning properties of those fluids have been characterized by the power-law rheology model. The dynamic contact angle (DCA), transient dimensionless height, and transient contacting factor along with the spreading time under different Weber numbers (We) have been investigated. The results show that the nanofluid with a lower shear viscosity over the entire range of the shear rates results in larger variations of the contacting factor and the dimensionless height. The effect of surface wettability on droplet impact behaviors is more significant for the fluid with higher shear viscosity and less shear-thinning degree during the receding phase. The latter spreading and receding motions of the droplet with higher shear viscosity and shearthinning degree are suppressed significantly, regardless of the Weber numbers in current study. Finally, a model based on experimental data has been proposed to predict the maximum spreading factor of high-viscous droplet impact on solid surface.

        Study on tool wear for efficient grooving blisk with disc milling cutter

        Tool wear has an important influence on machining quality, cost, cutting force and heat. Grooving blisk with disc milling cutter is 3-4 times more efficient than plunge milling and side milling, but the tool wear is serious because of the large milling force and high milling temperature during the machining process. In the study, the damage and wear morphology of the tool was observed by focused ion/double beam system; the wear mechanism was studied by energy spectrum analysis; the tool life curve was drawn by the increase of flank wear. The experimental results show that the main wear and damage morphologies of disc milling cutter are chip sticking to the tool, chip accumulation, edge collapse, peeling, crack, plastic deformation and boundary wear; the main wear mechanism are abrasive wear, adhesive wear, oxidation wear and diffusion wear; the wear rate of the left blade is the fastest, followed by the right blade, and the wear rate of the middle blade is the slowest. These results can provide guidance for iimproving tool life, optimizing tool and mproving machining processes.
