KR 31378, a Potent Antioxidant, Inhibits Apoptotic Death of A7r5 Cells

This work describes the pharmacological inhibition by KR 31378 and its acetyl metabolite, KR 31612, of the apoptotic cell death induced by $H_2O_2$ in the A7r5 cells. Exposure of A7r5 cells to $H_2O_2$ (0.5 mM) induced a concentration-dependent cytotoxicity in association with oligonucleosomal DNA fragmentation. $H_2O_2-induced$ cell death was potently suppressed by KR 31378, KR 31612, ${\alpha}-tocopherol$ or trolox. Additionally, the apoptotic death of A7r5 cells (DNA ladders on electrophoresis) was also strongly suppressed by KR 31378 and KR 31612, but to a less degree by ${\alpha}-tocopherol$ and trolox. As a mechanistic study, incubation with $H_2O_2$ markedly showed a decreased Bcl-2 level and, in contrast, increased Bax protein and cytochrome C release, which were significantly and concentration-dependently reversed by KR 31378 and KR 31612 as well as by ${\alpha}-tocopherol$ and trolox. KR 31378 and ${\alpha}-tocopherol$ significantly reduced lipid peroxidation in accordance with reduced intracellular ROS and peroxyl radical. These results suggest that KR 31378 has a therapeutic potential against the apoptotic injury via mediation of anti- oxidative stress.
Fabrication Technology of the Focusing Grating Coupler using Single-step Electron Beam Lithography

A focusing grating coupler (FGC) was not fabricated by the 'Continuous Path Control'writing strategy but by an electron-beam lithography system of more general exposure mode, which matches not only the address grid with the grating period but also an integer multiple of the address grid resolution (5 nm). To more simplify the fabrication, we are able to reduce a process step without large decrease of pattern quality by excluding a conducting material or layer such as metal (Al, Cr, Au), which are deposited on top or bottom of an e-beam resist to prevent charge build-up during e-beam exposure. A grating pitch period and an aperture feature size of the FGC designed and fabricated by e-beam lithography and reactive ion etching were ranged over 384.3 nm to 448.2 nm, and 0.5 $\times$ 0.5 mm$^2$area, respectively. This fabrication method presented will reduce processing time and improve the grating quality by means of a consideration of the address grid resolution, grating direction, pitch size and shapes when exposing. Here our investigations concentrate on the design and efficient fabrication results of the FGC for coupling from slab waveguide to a spot in free space.

The role of polyamines in skeletal myoblast differentiation was investigated using the polyamine metabolic inhibitor methylglyoxal bis(guanylhydrazone)(MGBG). Concentrations of intracellular free spemnidine and spemline increased 2 to 2.5-fold at the onset of myoblast fusion. The synthesis of actin, and creative kinase activity both dramatically increased during myotube formation. However, MGBG at a concentration of 0.5 mM not only abolished the increase of intracellular free polyamines, but also reduced cell fusion to almost half the level of untreated cells, without noticeable morphological alteration. The production of actin, and creative kinase activity were almost completely abolished by MGBG. The inhibition of myoblast fusion by MGBG was partially recovered with 0.1 mM of spermidine or spermine added externally. Results indicate that polyamines are necessary for normal myoblast differentiation. Since the first indication of myoblast differentiation is alignment of muscle cells and membrane fusion of adjacent cells, and since polyamine depletion completely inhibited the synthesis of acttn, which might be associated with membranes, polyamine might be involved in myoblast differentiation through membrane reorganization events.

In estrogen-dependent MCF-7 human breast cancer cells, $E_2$ at 10 nM stimulated cell proliferation to over 200% compared to the untreated control. EGF and TGF${\alpha}$, which are known as the autocrine/paracrine growth factors induced by $E_2$, also directly stimulated the cell growth in almost as the same extent as $E_2$. DFMO which is the specific inhibitor of ODC could inhibit cell growth even at as low as 0.5 mM. In the treatment with 1 mM DFMO for 4 days, the cell growth was inhibited to 38% of the control. HO-TAM at 1 ${\mu}$M could inhibit the proliferation of MCF-7 cells to 19% of the control. Those inhibitory effects were also found in the cells stimulated with $E_2$, EGF, and TGF${\alpha}$. The inhibitory effects were found even in 2 days of treatment. However, $E_2$, EGF, and TGF${\alpha}$ did not give any effect in the protein synthesis. Neither DFMO or HO-TAM gave any effect on the total protein synthesis. But the pattern of protein secretion was noticeably influenced by the growth stimulants or inhibitors. Proteins of 160, 52, 42, 36, and 32 kDa belonged to the major secretory proteins. Especially, 42 and 36 kDa proteins were most significantly influenced by the treatment of $E_2$, EGF, or TGF$\alpha$. DFMO and HO-TAM inhibited the secretion of these major proteins.
Drosophila p53 Upregulates VDAC Gene Expression That Is Linked to Apoptosis

p53 is a representative tumor suppressor whose dysfunction is a major cause of human cancer syndrome. Upon activation by various cellular stresses, p53 transactivates downstream target genes as a sequence-specific transcription factor, regulating cell cycle and apoptosis. The signaling pathway leading to apoptosis via the mitochondria is triggered by binding of the Bcl-2 family such as Bcl-2, Bax and Bak to voltage dependent anion channel (VDAC), and this interaction forms large pore in the mitochondria membrane and triggers cytochrome c release. In this study, we investigated the effect of Drosophila p53 (dp53) on VDAC gene expression. Our results indicate that overexpression of dp53 under specific GALA drivers induces severe morphological and developmental defects and dp53-mediated apoptosis may occur via VDAC gene upregulation.

스포츠 캐스팅을 할 때는 스포츠 장면에 대한 상황 정보, 선수 정보 그리고 과거 지식을 기반으로 현재 상황에 대한 이해와 추론이 필요하다. 본 논문에서는 장면 분류 모델, 선수 검출 모델 그리고 선수의 행동 인식 모델을 학습하여 스포츠 영상에 대한 정보를 얻고 과거 데이터를 지식화 해서 저장 해놓은 온톨로지를 이용하여 현재 상황에 대한 이해와 추론을 하는 방법을 소개한다. 총 3가지 종류의 캐스팅을 생성한다. 실시간 웹 데이터로부터 지식화 하여 캐스팅을 생성하고, 13개의 장면을 분류하여 온톨로지와 결합하여 캐스팅을 생성한다. 그리고 선수의 포지션과 8개의 행동을 인식하여 온톨로지와 결합하여 캐스팅을 생성한다. 모든 데이터는 2018년 4월 1일부터 2018년 4월 14일까지 있었던 KBO 경기를 직접 labeling 하여 모델을 학습하였다. Sports broadcasting requires understanding and reasoning of a current situation based on information regarding sports scenes, players, and past knowledge. In this paper, we introduced how scene classifier, player detector, motion recognizer could be used to obtain information on sports images and understand current situations. We created three types of commentaries. One was from web data, another was from 13 scenes with scene classifier, and the other was generated by the position of the players, eight motions, and the ontology. Data from the KBO (Korea Baseball Organization League) games from April 1, 2018, to April 14, 2018, were directly labeled to learn the model.
Effect of Selected Persimmon Leaf Components against Sarcoma 180 Induced Tumor in Mice

Sarcoma 180 복수형 종양세포를 생쥐의 왼쪽 서혜부에 이식한 후 감잎에서 분리한 tannin, chloroform fraction 추출물 및 2, 4-decadienal을 복강 내에 각각 단독투여하여 그 효과를 형태학적으로 검토하였다. 이식된 sarcoma 180 육종에서 암화의 진행을 지연시키는데는 tannin 투여시에 가장 효과적이었고 chloroform fraction 추출물 및 2, 4-decadienal 투여시에는 형태적으로 간주할 수 있는 지연효과는 없는 것으로 나타났다. 감잎 tannin은 종양세포를 선택적으로 파괴하여 종양의 증식을 억제하는 것이 아니라 일종의 혐기상태의 세포 환경에서 종양을 괴사과정으로 유도하지않고, 발암성을 가지는 단백질과 같은 물질의 합성을 저해하거나 또는 자식작용을 가진 과립들로 하여금 이들을 소화시켜 이식된 종양세포를 악성종양으로 유도하는 과정을 지연시키는 효과를 나타내었다. Antitumor activities of tannin extract and chloroform fraction extract from the persimmon leaves, and 2, 4-decadienal identified as an antimutagenic compound in persimmon leaves were examined in sarcoma 180 implanted tumor in mice by using both light and transmission electron microscopes. Among them, tannin extracted from the persimmon leaves delayed the progression of malignant tumor but the other two did not show any noticeable effect. The antitumorigenic activity of tannin extract might not come from the selective cytotoxicity against tumor cells, but might be, in an anaerobic environment, from the inhibitory action against oncogenic protein synthesis or from the proteolysis of the pre-formed oncogenic proteins by autophagocytic granules. Therefore, the tannin from persimmon leaves might protect cells from fast progression of malignant tumorigenesis.
