      • C. elegans as a model organism to study animal Behavior

        Ca2+ / calmodulin-dependent calcineurin has been shown to have important roles in various Ca2+ signaling pathways. We have previously reported that cnb-1 mutants, null mutants of a regulatory B subunit, displayed pleiotropic defects including uncoordinated movement and delayed egg laying in C. elegans. Interestingly, gain-of-function mutants of a catalytic A subunit showed exactly opposite phenotypes to those of cnb-1 mutants providing an excellent genetic model to define calcium-mediated signaling pathway at the organism level. Furthermore, calcineurin is also important for normal cuticle formation, whichis required for maintenance of normal body size in C. elegans. Genetic interactions between tax-6 and several mutants including egl-30 and egl-10 which are known to be involved in Gprotein signaling pathway suggest that calcineurin indeed regulates locomotion and serotonin-mediated egg laying through goa-1(Goα) and egl-30(Gqα). Our results indicate that, along with CaMKII, calcineurin regulates G-protein-coupled phosphorylation signaling pathways in C. elegans. The enteric muscle contraction (EMC) is the last step of the defecation behavior which occurs every 50 seconds in C. elegans. This EMC is regulated by intestinal and anal depressor muscles, which are innervatedby GABA motor neurons. Our data show that calcineurin (tax-6) is expressed in intestinal muscle and anal depressor muscle, and the gain-of-function mutant of calcineurin, tax-6(jh107), shows defects in enteric muscle contractions. In addition, exogenous GABA failed to rescue the EMC defects of tax-6(jh107) indicating that calcineurin functions in postsynaptic muscles rather than presynaptic neurons regulating EMC, which could be rescued by exogenous GABA treatment. Genetic epistasis with exp -1 mutant, which encodes an excitatory GABA receptor, suggests that calcineurin functions as a negative regulator of exp-1 in excitatory GABA signaling.

        In the 21st century, in prospect that cultural creativity will be the core of the development of a country, cultural administration is getting more important. Following this tendency, a great deal of effort is being made for the public relation and strategies concerned with culture and art and also the concern and the rate of participation of people is increasing. Recently, though the social and cultural extents of dance, which is one of the genres of art and culture, have got bigger and more diverse, too, dance is not known well compared with the other kinds of art. It is because the qualitative growth did not go together with the expansion in quantity, and especially, it is due to not so much the creativeness of dance as the defect found in administration or execution that lays the decline of reality. Therefore, in this situation that a solution which prepares the future in which the importance of the cultural administration is embossed and innovation over the base of administration of dance are desperately needed, I attempted to investigate the changing process of the administration of dance-how dance has been reflected in policy as a field that had cultural value until now- and to analyze the present condition of the field of dance and finally tried to find the problems of it expecting that this paper could be used as a guide for the dance cultural policy.

        연천 지역사회를 대상으로 하는 당뇨병 환자에서의 사망률 조사

        연구배경:1993년에 경기도 연천 지역에서 당뇨병의 유별률과 발생률을 산출하고자 시행된 한 단면연구의 대상자들을 추적 조사하고, 그 대상자들의 사망자료를 분석하여 당뇨병 환자의 사망률과 당뇨병 환자에서 흡연, 고혈압, 비만 등 여러 위험요인이 사망에 미치는 영향을 조사하고자 하였다. 대상 및 방법:1993년도에 수행되었던 제1차 당뇨병 유병율 조사 참여하였던 2,520명을 대상으로 하여 1993년 3월부터 1998년 12월까지 통계청의 사망자료를 확보하고 코호트 자료에 사망자료를 연계 결합함으로써 코호트 내에서의 사망례와 사망일을 확인하였다. 2,520명중 생화확적 검사 미실시, 설문조사 미실시, 주미등록번호 기재의 오류 등으로 2,305명만이 연구대상에 포함되었으며 사고와 중독으로 사망한 8명은 자료 분석 시 제외되어 2,297명이 최종적으로 분석 대상에 포함되었다. 결과:1. 코호트 구성원 2,297중 사망자는 6년 동안 남자 56명(5.5%), 여자 31명(2.4%)로 총 87(3.7%)이었다. 2. 비당뇨군 대 당뇨군의 전체 사망률에 대한 상대 위험도는 여러 위험요인들(연령, 성별, 수축기 혈압, 혈중 총 콜레스테롤, 흡연, 그리고 BMI)을 보정한 후 2.03이었다. 3. 비 당뇨군 대 당뇨군의 상대위험도는 남자에서는 40∼49세 사이에서, 여자에서는 60∼69세 사이에서 가장 크게 나타났으며, 나이가 증가함에 따라 감소하는 양상을 보여 주었다. 4. 흡연, 혈중 총 콜레스테롤, 수축기 혈압 및 BMI에 대하여 여러 군으로 나누어 당뇨군과 비당뇨군에서 사망률의 차이를 보면 당뇨군에서는 흡연, 혈중 총 콜레스테롤은 위험요인이 증가할수록 사망률도 증가하였다(p value<0.05). 그러나 수축기 혈압 및 BMI는 뚜렷한 연관성을 보이지 않았다. 5. 고혈압, 고지혈증, 비만 흡연 등의 위험요인이 증가함에 따른 사망률의 증가가 비당뇨군에 비해 당뇨군에서 더욱 크게 나타났다. 결론:이전의 대부분의 연구들이 당뇨의 유병률과 당뇨의 발생위험요인에 대한 단면적인 연구들이었으나 이 연구는 2.297명을 대상으로 6년 동안 추적 관찰하여 전향적으로 진행된 코호트 연구로서 우리나라에서 당뇨병 환자의 사망률을 조사한 최초의 연구이다. 다만 연구 대상자의 수가 적고 추적기간이 짧아 여러 위험요인이 사망률에 미치는 효과를 파악하기에는 부족하여 향후 추가적인 추적조사가 필요할 것으로 생각된다. Background : This study is designed to estimate the mortality rate and to assess the relation between other risk factors for mortality and death in adults with diabetes by analysis of death certificates in Yonchon cohort population. Methods : A population-based cross-sectional study was conducted in 1993 in Yonchon country to estimate the prevalence and risk factors of diabetes. This study population consists of respondents (2,463 persons) from Yonchon study and followed for 6 years from 1993 to 1998. Status of death and causes of death were determined from death certificates from National Statistical Office. Results : During the 6 year follow-up, 18 deaths (10%) occurred in the 184 persons with diabetes and 69 deaths (3.3%) occurred in the 2,113 persons without diabetes. After adjustment for multiple variable (age, sex, total cholesterol, systolic blood pressure, smoking and body mass index (BMI), mortality rate was significantly higher for diabetic adults than non-diabetic adults (RR, 2.03). The proportional hazard analysis for all-cause mortality in the 190 persons with diabetes showed that smoking, high total cholesterol, and high LDL-cholesterol were significantly associated with increased risk for mortality (p value < 0.05), but BMI, HDL-cholesterol, and high systolic blood pressure were not significantly associated with increased risk for mortality. Conclusions : This study was a prospective cohort study that followed 2,463 persons of Yonchon cohort for 6 years and showed that diabetic adults had higher mortality than non-diabetic adults. The strength of the association between risk factors and mortality was less clear because follow-up period was short and study population size was small, therefore further follow-up study are needed (J Kor Diabetes Asso 25:384~398, 2001).

        예쁜 꼬마 선충의 1 번 염색체에 위치하는 unc-29 유전자 좌위 주위의 jh 2 돌연변이의 유전학적 분석

        Six lethal mutations near the unc-29 locus on chromosome I of Caenorhabditis elegans have been isolated previously. Among these mutants, one particular mutant (jh2) showed interesting terminal defect, which suggest that embryos of this mutant suffer specific defects in posterior body parts during early development. As the first step to further characterize the mutation, we have performed genetic outcrossing using three genetic markers of dpy-5(e61), dpy-5(e61 ) unc-13(e1091), unc-13(e1091) lin-11(n566) in the gene cluster region near the unc-29 locus. Following the outcrosses. three-factor crosses were conducted to place the jh2 mutation on a precise genetic map near the unc-29 locus on chromosome I. The results of three-factor crosses have placed the jh2 mutation to the region between 4.41 and 4.54 map unit right from the center of chromosome I, which corresponds to approximately a 200kb region in the physical map.
