A serious of test were conducted to examine the effects of the reinforcement length to the geosynthetic reinforced soil (GRS) wall models. The strength for the GRS wall with increased reinforcement length was improved. The failure in GRS wall models shows ductile behavior however the failure in unreinforced wall model shows inelastic behavior.
Expression of c-fos, c-Ha-ras and Laminin in the Human Stomach and Colorectal Carcinoma Cell Lines

한국인 위장관 종양에서 확립된 3종류의 위암 세포주(SNU-1, 5, 16)와 7종류의 대장암 세포주 (SNU-61, 67, 70, C1, C2A, C4, C5)를 대상으로 하여 암유전자 및 라미닌 유전자의 발현양상과 종양전이 능력과의 상관관계를 조사하였다. c-fos 암유전자의 경우 일차종양에서 확립된 SNU-C4와 C5에서 발현이 높게 나타나고, c-Haras 암유전자인 경우는 전이 부위에서 확립된 SNU-61 세포주에서 발현이 높으므로 전이성이 낮은 세포주에서 c-fos의 발현이 높고, 전이성이 높은 세포주에서는 c-Ha-ras의 발현이 높다는 종래의 실험결과와 어느 정도 일치함을 나타내었다. 그러나 라미니 유전자인 경우는 SNU-5, C2A, C5 등의 세포주에서 비교적 높게 발현이 되었으나 다른 세포주에서도 고르게 발현이 되고 있으므로 라미닌 유전자의 발현과 종양전이 능력과의 상관성을 SNU 세포주에서는 얻기가 어려운 것으로 추정이 된다. We have investigated a correlation between oncogene and laminin expression and metastatic aggressiveness with three stomach (SNU-1, 5, and 16) and seven colon (SNU-61, 67, 70, C1, C2A, C4, and C5) carcinoma cell lines which were estabilished from primary and metastatic tumor sites. According to Northern blot analysis and immunofluorescence staining, the expression of c fos was elevated in SNU-C4 (primary, colon) and SNU-C5 (primary, colon). The expression of c-Ha-ras was increased in SNU-61 (metastatic, colon). In malignant progression, ras oncogene expression was believed to be responsible for tumor metastasis, whereas the mRNA of c-fos was expressed mainly in the low metastatic clones. In this regard, our results were somewhat in accordance with previous reports. On the other hand, laminin mRNA was detected in all cell lines tested, and among them higher level of laminin mRNA expression was observed in SNU-5 (metastatic, stomach), SNU-C2A (primary, colon) and SNU-C5 (primary, colon). This result suggests that the tumor metastatic behavior can not be simplified to laminin gene expression and the correlation between tumor metastatic aggressiveness and laminin gene expression is more complex.

We obtained the total protein antibodies of Saccharomyces cerevisiae KCTC 1720 and Escherichia coli K-12 from the rabbit and the guinea pig to determine the host-derived proteins which may be remained in biotechnological products. The protein concentration of rabbit antibodies was 4.05 mg/ml in the case of yeast, 7.14 mg/ml in the case of E. coli and that of guinea pig antibodies was 1.90 mg/ml in the case of yeast, 7.17 mg/ml in the case of E. coli, respectively. To determine remained host-derived proteins in biotechnological products which produced by the hosts, S. cerevisiae or E. coli, we used a sandwich enzyme linked immunosorbent assay method in 96 well microplate. When the method applied to determine the remained host-derived proteins in commercial biotechnological products, it detected less than 3.5 ng/vial in human growth hormone, less than 1 ng/vial in hepatitis B vaccine and interferon-γ and 2∼23 ng/vial in interferon-α. The method can be used to determine the remained host-derived protein in biotechnological products.

Laminin, one of major constituents of the basement membrane, is a multisubunit glycoprotein with a cross-like shape and consists of three polypeptides of molecular weights 450kd (A chain), 240kd (B1 chain) and 230kd (B2 chain). There is experimental evidence that laminin is involved in many cellular processes, such as embryonic differentiation, morphogenesis, cell migration, tumor metastasis, and neurite growth and regeneration. To sutdy the regulation of laminin gene expression at the molecular level, we cloned almost full-length laminin B1 cDNA from mouse F9 teratocarcinoma stem cell treated with retinoic acid. The cloned laminin B1 cDNA was characterized by restriction analysis. The 5' end of the cDNA was isolated and sequenced by M13 DNA sequencing method. Then, the 5' end fragment of laminin B1 cDNA was inserted into plasmid pUC9 to use as a marker probe for the regulation of laminin gene expression in normal tissues and abnormal metastatic tumors.
인간 대장암 세포주에서 chromogranin A 발현의 증가

Chromogranin A(CgA) appears to be a major secretory protein in the parathyroid secretory granules and its distribution selves as a marker for neuroendocrine lineage differentiation. The amino acid sequence of bovine CgA shows a high degree of homology to pancreastatin, an isolated porcine peptide that inhibits glucose-induced insulin secretion. Three stomach and seven colon carcinoma cell lines established from Korean patients were used in this study. We tried to classify these cell lines into neuroendocrine lineage by detecting the expression of CgA mRNA. According to the results of Northern blot, the levels of CgA mRNA were highly elevated in two colorectal carcinoma cell lines such as SNU-61 and SNU-C4. To detect whether CgA mRNA expression was due to gene amplification or gene rearrangement, Southern blot analysis was performed. One major band was detected in all cell lines, however, CgA DNA in SNU-61 and SNU-C4 was not amplified as compared to the other cell lines. From the above result, we concluded that the elevation of CgA mRNA expression was not due to gene amplification or gene rearrangement, hence, its expression might be transcriptionally or post-transcriptionally increased.
