        In this study, authors investigated the bathroom soap development using plum(Prunus salicina Lindl. cv. Soldam) extracts and studied the effects and safety of a plum soap on acne patients. For obtaining antibacterial material from dried plum, the serial extraction using organic solvent was carried out and gained of 0.2% hexane, 0.2% chloroform, 0.23% ethyl acetate, 9.8% butanol and 2.1% water extracts. The main bioactive contents of the soap were consisted of 0.006% plum extract, 0.1% glycyrrhiza extract, and 0.15% tocopheryl acetate. A clinical study of soap containg 0.006% plum extracts was carried out on three groups of people: a treatment and 2 control groups on acne patients. Efficacy and safety were assessed at baseline and at one month later. The group applving the soạp with plum extract felt better than the one applying the 2 control group. Some patients developed mild and transient local side effects. The plum soap didn't change at 40℃ and was 130 hardness. In conclusion, cleansing three times a day with a facial soap consisting 0.006% ethyl acetate extract of plum was found to be effective and safe for patients suffering from acne vulgaris

        The dietary fiber(GLD-06), β-glucanic polysaccharide, from the mycelium of Ganoderma lucidum was fractionated by the sequential use of salts and hot extraction and obtained 0.35% of yield from the mycelial culture medium.

      • In vitro에서 B-glucan성 다당류들에 의해 활성화된 흰쥐 간내 Kupffer 세포에서의 NO, TNF-a 및 TGF-3의 형성

        Various B-glucans were evaluated for its ability to formation of nitric oxide (NO), tumor necrosis factor-a (TNF-a) and transforming growth factor(TGF-B) from rat kupffer cell in vitro. Hepatic macrophages activated by glucan did not significantly elevated concentration of TGF-B in the medium, but significantly elevated NO and TNF-a concentration in culture medium. Comparing the activity using structurally related glucans obtained from microorganisms, highly branched glucans, Ganoderan isolated from G. lucidum IY009 and zymosan from Saccharomyces cerevisiae, significantly increased TNF-a and NO production Ganoderan(GAN), a particulate 8-1.3-polyglucose immunomodulator from fungal mycelium, enhances 4-fold production of NO and 19-fold formation of TNF compared to control. The GAN-activated kupffer cells secrete 14.94M/C(p<0.01) of NO and 2619.5 pg/ml (p<0.01) of TNF-a after 38 hrs. Large molecular weight GAN(approximately 10Kd-20Kd) prepared by NaOH extraction method showed stronger stimulating effect. These facts suggested the glucans showed stimulating effect of TNF-a and NO production from kupffer cell in vitro that this effect was related to the degree of branching and molecular weight. The results suggest that B-glucan related-polysaccharide may be of value in combination with other therapies in the treatment of immunodeficiencies, cancer, infections, autoimmune disease and hepatic disease.

      • Ganoderma lucidum의 세포외성 다당류의 생리활성

        Antitumor polysaccharides were known to activate complement system and incease pecific serum proteins in mouse, and some researcher reported that antitumor activity of polysaccharides might be correlated and macrophage. The b-glucan of G. ucidum induces potent antitumor evaluate the bioactivities of extracellular polysaccharide obtaioned from the mycelia of G. lucidum. The water-insoluble rome (EXO-WI) showed the highest antitumor activity(92.1% inhibition rate) in carcoma- bearing mice, whereas, have relatively little activites at anticomplementary activity. In examination of macrophage activation, Raw 264.7 cells activated with EXO-WI incrased 3.5-fold(24 hrs) formation of nitric oxide levels and about 4-folds production of TNF – a compared with a control with medium only. EXO-WI was composed with exclusively b-D-glucose and 14.7% protein moiety. These results indicate that the EXO-WI purified from myelia of G. lucidum may be of a useful immunopotentiator in the treatment of cancer and immune disease.

        The extract of Basidiomycetes induces potent antitumor immunity in tumor-bearing mice. The study was set up to elucidate the anti-cancer effects of various mushroom extract against human cancer cell lines. The mushroom extract was prepared from the fruit body of many types of Basidiomycetes: Poria cocos, Ganoderma lucidum, Ganoderma oregonence. Ganoderma applanatum, Ganoderma tusgae. Fomitella fraxinea. Laetiporus sulphureus, Phellinus lintenus, Agaricus brazei. Most extracts showed cytotoxicity on human cancer cell line: hepatocytoma (SNU 387), human colon carcinoma (SNU C4)and human gastric adenocarcinoma (SNU-1). The highest anticancer effect(83.1% cytotoxicity) of tested samples occurs with Ganoderma applanatum extract on human hepatocytoma SNU 387 cell line. In conclusion, the low molecular extract(<10,000 M. W) of Basidiomycetes hot extract was higher anticancer effect than that the high molecular()10.000 M. W) extract on A549, SNU-1, SNU C4. and SNU 387 human cancer cell line.

      • 영지버섯(Ganoderma lucidum)의 균사체 생산에 관하여

        약효가 다양한 영지버섯의 균사체 생산에 최적인 배지를 조성코저 몇가지 실험을 통하여 아래와 같은 결과를 얻었다. 1.영지버섯을 2% soluble starch, 0.5% yeast extract, 0.5% KH₂PO₄, 0.01% MgSO₄.7H₂O그리고 0.001% K₂HPO₄로 조성한 배지에서 27℃로 7인간 진탕배양(120 rpm)했을때 16.38mg/ml로 최대의 균사체를 얻었다. 2.최적 배지내에서 균주의 배양 4일까지는 탄수화물을 서서히 감소하고, 환원당을 급격히 증가하였다. 그러나 배양4일 이후부터는 탄수화물과 환원당 모두 급격히 감소하였다. 3.최적 배지내에서 pH는 초기에 5.5에서 배양 3일째에 4.0으로 떨어져 배양 7일까지 지속되었다. In order to increase the mycelial production of Ganodermalucidum which exhibits various phamacological effects on mammalian, some ingredients of medium were investigated. 1.The effective culture medium for the mycelial production by the culture contained 2% soluble starch, 0.5% yeast extract, 0.5% KH₂PO₄, 0.01% MgSO₄.7H₂O and 0.001% K₂HPO₄ 2.The maximum yield of the mycelium of the culture was 16.38mg/ml when it was grown in the effective medium with rotary shaking (120 rpm) at 27℃ for 7 days. 3.During the culture of the strain in the effective medium, total carbohydrate in medium was decreased with the culture time but reducing sugar rapidly increased by the three days of culture time; it was decreased three days later. 4.The pH in the effective medium was decreased from 5.5 at the initial culture time to 4.0 on the third day but was maintained till the seventh day since then.

      • Bacillus thuringiensis var. israelensis의 포자와 crystal생성에 관하여

        Bacillus thuringiensis var. israelensis(B.t.i)의 내생포자와 crystal의 생성을 높이기 위하여 몇 가지 배양조건들을 조사하였다. B.t.i를 1.5% soluble starch, 0.5% glucose, 1.0% yeast extract, 0.2% K₂HPO₄, 0.01% MgSO₄.7H₂O, 0.02% CaCl₂, 0.002% FeSO₄.7H₂O, 그리고 0.002% MnSO₄.4H₂O로 구성되는 배지(pH7.0)에서 30C로 진탕배양(200rpm)했을때 배양 32시간에 균주의 생장이 최대가 되었다. 배양 72시간에 수획한 포자와 crystal복합체의 농도가 0.05㎍/㎖일때 Aedes togoi유충은 12% 죽었고 0.01㎍/㎖농도일 때 Culex pipiens 유충은 15% 죽었다. 한편 포자와 crystal복합체의 농도가 0.5㎍/㎖일때 Aedes togoi와 Culex pipiens유충은 모두 죽었다. In order to increase the production of spore-crystal complex of Bacillus thuringiensis var. israelensis which is toxic to mosquito larvae, some culture conditions of the culture were investigated. 1. The effective culture medium for the production of spore by the culture contained 1.5% soluble starch, 0.5% glucose, 1.0% yeast extract, 0.2% K₂HPO₄, 0.01% MgSO₄.7H₂O, 0.02% CaCl₂, 0.002% FeSO₄.7H₂O and 0.002% MnSO₄.4H₂O. 2.The population density of the culture was at a maximum after 32 hours of incubation when it was grown in the effective medium at 30C with rotary shaking. 3.Spore-crystal complex of B. thuringiensis var. israelensis which was grown in the effective medium exhibited toxicity to Aedes togoi larvae at 0.05㎍/㎖ and Culex pipiens larvae at a concentration 0.01㎍/㎖ respectively. One hundred percent of Aedes togoi and Culex pipiens larvae were killed at a dosage rate of 0.5㎍/㎖

      • 영지 (Ganoderma lucidum) 자실체 다당류의 분자량에 따른 생리활성

        The study was conducted to investigate the biological activities of an protein-bound polysaccharide (GLP) isolated from the fruit body of Ganoderma lucidum. GLP was extracted by hot water and then it was purified high (GLP-HM) and low (GLP-LM) molecular weight fraction by ethanol precipitation. Both of GLP-HM and GLP-LM at doses of 20 mg/kg in vivo had the ability to inhibit the tumor growth of Sarcoma 180 tumor-bearing mice. Inhibition rates of tumor (%) were inhibited 60.6% and 38.7% by GLP-HM and GLP-LM, respectively. In assay of anti-complementary activity in vitro, the GLP-HM were higher than that of the GLP-LM polysaccharide. And also, the GLP-HM enhanced nitric oxide production of mouse bone marrow cells. When the GLP-HM was injected intraperitoneally in tumor-bearing mouse at doses of 20 mg/kg, the concentration of TNF-α in vivo was 140.9 pg/ml in the serum, 269.6 pg/ml in sarcoma tumor mass and 879.4 pg/ml in liver. This study shows that the high molecular polysaccharide from mushroom can enhance more the bioactivities than that of the low molecular polysaccharide.
