      • 麥酒麥의 (1-3,1-4)-β-glucanase 遺傳子의 構造와 發現 樣狀

        禾穀類 種子 發芽時 호분층과 ??組織의 細胞壁에 多量 含有되어 있는 (1-3, 1-4)-β-glucan을 分解하여 種子의 發芽에 必要한 加水分解酵素의 分泌 및 擴散 浸透를 돕는다. 本 硏究에서는 國內 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子의 構造 및 發現 에 대한 基礎 情報를 얻기 위하여 生化學的 및 分子生物學的 方法을 利用하여 發芽種子 및 어린 잎에서의 酵素活性과 遺傳子의 發芽樣狀 그리고 遺傳子의 構造 等을 調査하였다. 麥酒麥 發芽種子中 (1-3, 1-4)-β-glucanase 活性은 發芽 7日째에 제일 높았다. (1-3, 1-4)-β-glucanase의 活性測定에 使用한 (1-3, 1-4)-β-glucan이 (1-4)-β-glucanase에 의해 分解될 可能性이 있으므로 (1-4)-β-glucanase의 活性도 測定하였는데 (1-4)-β-glucanase의 活性이 매우 낮게 나타나서(1-4)-β-glucanase가 (1-3, 1-4)-β-glucanase을 分解하여 (1-3, 1-4)-β-glucanse의 活性에 影響을 미쳤을 可能性이 매우 낮았다. 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子 構造를 PCR을 利用하여 調査하였던바 國內 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子 構造는 外國의 다른 品種들이나 귀리의 遺傳子 構造와 대단히 類似하였다. 酵素活性 測定과 귀리의 (1-3, 1-4)-β-glucanase cDNA clone을 ?識子로 利用한 보리의 mRNA 分析에 의해서 發芽傳子 및 어린 잎에서 (1-3, 1-4)-β-glucanase가 種子發芽 以外에도 生育中인 組織細胞의 伸張과 發育 및 β-glucan 再利用에 泌要한 β-glucan 代謝에 關與하고 있음을 示唆하는 結果라 생각된다. (1-3, 1-4)-β-glucans hydrolysis of cell wall (1-3, 1-4)-β-glucanas accompanies degradation of endosperm cell walls during cereal seed germination. To investigate (1-3, 1-4)-β-glucanase expression pattern in Korean malting barley, (1-3, 1-4)-β-glucanase activity, gene structure and mRNA expression patterns werre examined. (1-3, 1-4)-β-glucanase activity increased as germination proceeded and the highest enzyme activity was detected in seeds germinated for 7 days. Because (1-4)-β-glucanase have a potential to degrade (1-3, 1-4)-β-glucans, (1-4)-β-glucanase activity was also determined. (1-4)-β-glucanase activity, however, was very low minimizing possibility of (1-4)-β-glucanase action on (1-3, 1-4)-β-glucan substrate. PCR technique was employed to investigate (1-3, 1-4)-β-glucanase gene structure. The amplification product of the expected size from the targeted sites of Dusan #29 indicated that (1-3, 1-4)-β-glucanase systems were conserved in different barley varieties and even in oats. (1-3, 1-4)-β-glucanase activity and mRNAs were detected in young leaves as well as in germinating seeds. This may suggest that (1-3, 1-4)-β-glucanase are also involved in β-glucan metabolism required for cell elongation and β-glucan reutilization during leaf growth and developmetn.

        우리나라 산업의 정밀측정기술 수준분석

        The precision measurement technology is the infrastructure of industry technology and thus it is the critical element in the quality and hi-tech products. Nevertheless, the precision measurement technology in Korean industry is the lower level of the advanced state. In this study, we survey the demand of precision measurement technology including instrument and technologies in Korean industry, and provide the objective data on the level of precision measurement technology based on the report "A Study on the Current Status of Precision Measurement in Korea(KRISS, 2001)" . We summarize contents on the precision measurement technology in Korean industry, provide the assessment model for the level of precision measurement technology, and analyze the results of these. The level of the precision measurement technology of National Standards Institution, MISS, approaches those of the advanced state, but levels of the precision measurement technology in Korean industry are 2-3 years behind levels of National Standards Institution. Analysis model and results are analyzed for the important index to compare the levels of the precision measurement technology in Korean industry. Therefore, it is expected to provide the development for precision measurement technology and improve the system of precision measuring instruments, level of precision and accuracy, calibration, technical manpower for measurement etc. in Korean industry.

      • PCR-based allele specific markers for bacterial blight resistance gene in rice.

        The use of functional markers, it is expected to make direct identification about genetic diversity at DNA level and overcome the problem of recombination /linkage. These markers can be used to identify interesting alleles in a breeding program and indirectly select for the trait, saving money, time and labor. Bacterial blight of rice caused by Xanthomonas oryzaepv. Oryzae is a destructive disease in rice production worldwide. No bactericide is effective to control the bacterial blight disease yet. Xa3, which is a gene conferring resistance to BB of the rice plant has been previously characterized by map-based cloning. We have cloned and sequenced the Xa3/xa3 gene in Korean cultivar, Hwayoung, Ilmi and Goun with gene specific primers. Our work detected polymorphisms and PCR-based allele specific SNP markers were developed. Susceptible or resistant individuals from an F2 population developed from across between Milyang244 and Ilmi, Korean germplasms and near isogenic lines carrying BB resistance genes were screened with allele specific markers. We found that the genotype completely matched their phenotype to BB using ASP-primers. These markers could be effective to marker-assisted selection for the Xa3 gene in rice breeding programs.

        Enhanced antidiabetic efficacy and safety of compound K/β-cyclodextrin inclusion complex in zebrafish

        Background: 20(S)-Protopanaxadiol 20-O-D-glucopyranoside, also called compound K (CK), exerts antidiabetic effects that are mediated by insulin secretion through adenosine triphosphate (ATP)-sensitive potassium ($K_{ATP}$) channels in pancreatic ${\beta}$-cells. However, the antidiabetic effects of CK may be limited because of its low bioavailability. Methods: In this study, we aimed to enhance the antidiabetic activity and lower the toxicity of CK by including it with ${\beta}$-cyclodextrin (CD) (CD-CK), and to determine whether the CD-CK compound enhanced pancreatic islet recovery, compared to CK alone, in an alloxan-induced diabetic zebrafish model. Furthermore, we confirmed the toxicity of CD-CK relative to CK alone by morphological changes, mitochondrial damage, and TdT-UTP nick end labeling (TUNEL) assays, and determined the ratio between the toxic and therapeutic dose for both compounds to verify the relative safety of CK and CD-CK. Results: The CD-CK conjugate ($EC_{50}=2.158{\mu}M$) enhanced the recovery of pancreatic islets, compared to CK alone ($EC_{50}=7.221{\mu}M$), as assessed in alloxan-induced diabetic zebrafish larvae. In addition, CD-CK ($LC_{50} =20.68{\mu}M$) was less toxic than CK alone ($LC_{50}=14.24{\mu}M$). The therapeutic index of CK and CD-CK was 1.98 and 9.58, respectively. Conclusion: The CD-CK inclusion complex enhanced the recovery of damaged pancreatic islets in diabetic zebrafish. The CD-CK inclusion complex has potential as an effective antidiabetic efficacy with lower toxicity.

        Synergistic Potentials of Coffee on Injured Pancreatic Islets and Insulin Action via K<sub>ATP</sub> Channel Blocking in Zebrafish

        <P>Pancreatic islets (PIs) are damaged under diabetic conditions, resulting in decreased PI size. This study examined the regenerative effects of coffee and its components (caffeine, CFI; trigonelline, TRG; chlorogenic acid, CGA) on zebrafish larval PIs and β-cells damaged by administration of alloxan (AX). In addition, the influence of coffee and its active components on K<SUB>ATP</SUB> channels was investigated using diazoxide (DZ) as a K<SUB>ATP</SUB> channel activator. PI size and fluorescence intensity were significantly increased in the coffee-treated group relative to the no-treatment group (<I>P</I> < 0.0001). In addition, coffee exerted significant regenerative effects on pancreatic β-cells (<I>p</I> = 0.006). Treatment with TRG and CGA rescued PI damage, and the combination of TRG/CGA had a synergistic effect. In conclusion, the results indicate that coffee has beneficial effects on AX-damaged PIs and may also be useful as a blocker of pancreatic β-cell K<SUP>+</SUP> channels.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jafcau/2015/jafcau.2015.63.issue-23/acs.jafc.5b00027/production/images/medium/jf-2015-000272_0001.gif'></P>

      • Association of Herpesvirus Saimiri Tip with Lipid Raft Is Essential for Downregulation of T-Cell Receptor and CD4 Coreceptor

        <B>ABSTRACT</B><P>Lipid rafts are membrane microdomains that are proposed to function as platforms for both receptor signaling and trafficking. Our previous studies have demonstrated that Tip of herpesvirus saimiri (HVS), which is a T-lymphotropic tumor virus, is constitutively targeted to lipid rafts and interacts with cellular Lck tyrosine kinase and p80 WD repeat-containing endosomal protein. Through the interactions with Lck and p80, HVS Tip modulates diverse T-cell functions, which leads to the downregulation of T-cell receptor (TCR) and CD4 coreceptor surface expression, the inhibition of TCR signal transduction, and the activation of STAT3 transcription factor. In this study, we investigated the functional significance of Tip association with lipid rafts. We found that Tip expression remarkably increased lipid raft fractions in human T cells by enhancing the recruitment of lipid raft-resident proteins. Genetic analysis showed that the carboxyl-terminal transmembrane, but not p80 and Lck interaction, of Tip was required for the lipid raft localization and that lipid raft localization of Tip was necessary for the efficient downregulation of TCR and CD4 surface expression. Correlated with this, treatment with Filipin III, a lipid raft-disrupting agent, effectively reversed the downregulation of CD3 and CD4 surface expression induced by Tip. On the other hand, Tip mutants that were no longer present in lipid rafts were still capable of inhibiting TCR signaling and activating STAT3 transcription factor activity as efficiently as wild-type (wt) Tip. These results indicate that the association of Tip with lipid rafts is essential for the downregulation of TCR and CD4 surface expression but not for the inhibition of TCR signal transduction and the activation of STAT3 transcription factor. These results also suggest that the signaling and targeting activities of HVS Tip rely on functionally and genetically separable mechanisms, which may independently modulate T-cell function for viral persistence or pathogenesis.</P>

        본 연구에서는 우리나라 산업체와 관련기관의 측정기술, 기기 및 인력의 수요를 각 분야별로 조사하고, 이와 함께 국가표준 및 교정제도에 대한 의견 및 애로사항 등을 종합적으로 고려하여 정밀측정기술의 발전방안을 제시하였다. 본 연구에서 제시된 결과는 앞으로 산업체의 각 분약에 걸쳐 정말측정기술에 대한 인식의 변화와 정밀측정기술 인력의 양성 및 시설·설비투자의 활성화에 기여할 것으로 기대한다. In this study, we summarise the status of demand and contents on the precision measurement technology. And we provide the Enhancement scheme of the precision measurement technology in Korea through findings on national standards and calibration system. Results of this report is respected to provide the improvement for precision measurement technology and to advance system of precision measuring instruments, the level of precision and accuracy, the system calibration, the quality of the technical manpower for measurement etc. in korea industry.

        유아과학 교수자료 수집을 위한 한국 전래 그림동화의 내용분석

        This study investigated illustrated Koran fairy tales as material for teaching kindergarten science. The 64 books selected for this study were analyzed by the standards set forth in the Exploring Life segment of the 5th Revision of Kindergarten Teaching Procedures (1993). The story-structure strategy was based on the structural pattern suggested by Moen(1991). Results showed that the structural elements of the illustrated fairy tales could be classified into 7 categories and 8 types of content patterns. After validating the adaptability of the fairy tales for use in teaching Kindergarten science, the books were cataloged by type and category.
