        Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

        High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.

        Maintaining Drosha expression with Cdk5 inhibitors as a potential therapeutic strategy for early intervention after TBI

        Traumatic brain injury (TBI) is a major cause of death and disability in adults. The pathological process of TBI involves a multifactorial cascade in which kinases have been proven contribute to interactions between relevant factors and amplification of signaling cascades. Cyclin-dependent kinase 5 (Cdk5) is a promising kinase that has been implicated in various brain disorders, including TBI. However, the mechanism by which Cdk5 induces neuronal damage remains unclear. Here, we show for the first time that Drosha, a key enzyme in microRNA biogenesis, is a pivotal substrate of abnormally activated Cdk5. Cdk5-mediated phosphorylation decreases Drosha expression and exacerbates nerve injury in TBI. We proved that maintaining Drosha expression via the administration of repurposed Cdk5 inhibitors that were previously studied in clinical trials is a promising approach for the early treatment of TBI. Together, our work identifies Drosha as a novel target for neuroprotective strategies after TBI and suggests Cdk5-mediated regulation of Drosha expression as a potential therapeutic strategy for early TBI intervention.

        Molecular mechanisms conferring asymmetrical cross-resistance between tebufenozide and abamectin in Plutella xylostella

        Based on the confirmation of asymmetrical cross-resistance between abamectin and tebufenozide in Plutella xylostella, the present work proved that the cytochrome P450 monooxygenase plays a decisive role in crossresistance, and the expression of various cytochrome P450 (CYP450) genes in different strains was surveyed to elucidate the molecular basis of the underlying mechanisms. Enzyme analysis showed the activity of cytochrome P450 monooxygenase was notable enhanced in the strains resistant to both tebufenozide (3.07-fold) and abamectin (3.37-fold), suggesting that the enhancement of cytochrome P450 monooxygenase is the main detoxification mechanism responsible for the cross-resistance. CYP4M7 (64.58-fold) and CYP6K1 (41.97-fold) had extremely high expression levels in the Teb-R strain, selected using tebufenozide, which was highly resistant to tebufenozide (RR 185.5) and moderately cross-resistant to abamectin (RR 41.0). When this strain was subjected to further selection using abamectin, the resultant Aba-R strain showed a higher expression of CYP6K1 (60.32-fold). However, the expression of CYP4M7 was reduced (10.62-fold). Correspondingly, the Aba-R strain became more resistant to abamectin (RR 593.8) and less resistant to tebufenozide (RR 28.0). Therefore, we concluded that the over expression of CYP4M7 was the main cause for tebufenozide resistance, and that CYP6K1 mainly conferred abamectin resistance. The asymmetrical cross-resistance occurred because tebufenozide selection not only enhanced the expression of CYP4M7, but also that of CYP6K1. This is the first report on the molecular mechanism of asymmetrical cross-resistance between insecticides.

        A Novel Parallel Precision Stage with Large Working Range Based on Structural Parameters of Flexible Hinges

        In order to solve the inherent contradiction of high precision and large working range in the traditional parallel positioning stage, a novel parallel precision stage with large working range based on optimized flexible hinge was studied. Firstly, a new structure parameter Ps was proposed, and its influence on the rotation stiffness was analyzed by comparing the commonly used flexible hinges. The factors that could enlarge the rotation ability were summarized, and the cylindrical flexible hinge with large working range was optimized. Then the cylindrical flexible hinge was used to constructed a novel parallel precision positioning stage, which could realize both of high resolution and large working range by taking use of the step and continuous motion of the piezoelectric actuators. The experiment results show that the resolution of translation is 0.09 μm, and the rotation resolution are 0.8 μrad, 0.9 μrad and 1.0 μrad, respectively. While the working range of the parallel stage could achieve 120 mm for translation, and 6.18°, 6.74° and 6.58°, respectively for each axis of rotation. All of the basic research contents is helpful for further study of the control laws of the compliant parallel mechanism.

        The CKLF-like MARVEL transmembrane domain-containing family (CMTM) is a novel family of genes first reported at international level by Peking University Human Disease Gene Research Center. The gene products act between chemokines and the transmembrane-4 superfamily. Located in several human chromosomes, the CMTMs CKLF and CMTM1 to CMTM8 may be unregulated in tumors and act as potential tumor suppressor genes with important roles in the immune, male reproductive and hematopoietic systems. In-depth studies in recent years established a close relation between CMTMs and tumorigenesis and metastasis. The CMTM family has a significant clinical value in diagnosis and treatment of diseases linked to tumors and the immune system.

        Removal characteristics of Cd(II) ions from aqueous solution on ordered mesoporous carbon

        Ordered mesoporous carbon (CMK-3) was synthesized using SBA-15 mesoporous molecular sieve as a template and sucrose as carbon source. The materials were characterized by XRD, TEM and N2 physical adsorption technique. The resulting CMK-3 was used as adsorbent to remove Cd(II) ions from aqueous solution. The effect of pH, contact time and temperature on adsorption process was investigated in batch experiments. The results showed that the removal percentage could reach ca. 90% at the conditions of initial Cd(II) ions concentration of 20 mg/L, dose of 20mg, pH 6.5, contact time of 3h and 293K. Langmuir and Freundlich models were employed to describe the adsorption equilibrium. The kinetics data were described by the pseudo-first-order and pseudo-second-order models, respectively. The adsorption isotherm was well fitted to the Langmuir model, and the adsorption process was well described by the pseudo-second-order kinetic model.

      • Phase II Trial of Loubo<sup>®</sup> (Lobaplatin) and Pemetrexed for Patients with Metastatic Breast Cancer not Responding to Anthracycline or Taxanes

        Purpose: This phase II study was undertaken to determine the efficacy and safety of Loubo$^{(R)}$ (Lobaplatin) in combination with pemetrexed in treating patients with metastatic breast cancer who failed to respond to anthracycline or taxanes. Patients and Methods: Metastatic breast cancer cases who had previously received an anthracycline and a taxane in either adjuvant or metastatic settings, were enrolled. All patients were recruited from Jiangsu Cancer Hospital and Research Institute, and were treated with Loubo$^{(R)}$ (Lobaplatin) 35 $mg/m^2$ (intravenous; on day 1) and pemetrexed 500 $mg/m^2$ (intravenous; on day 1) every 21 days. Efficacy and side effects were evaluated after at least two cycles of chemotherapy. Results: All eligible 19 patients completed at least 2 cycles of chemotherapy with pemetrexed and lobaplatin, and were evaluable. Overall, 3 (15.8%) patients achieved partial response, 11 (57.9%) stable disease, 5 (26.3%) progression of disease, with no complete remission. Response rate was 15.8%, disease control rate was 42.1%. The median survival time was 10.3 months. Neutrophil suppression occurred in 36.8% of patients who had grade 2 toxicity, and 26.3% had grade 3, 26.4% had grade 4. Thrombocytopenia was encountered as follows: 21.1% grade 2, 15.8% grade 3 and 5.5% grade 4. Incidences of anemia were 10.5% in grade 2, 5.3% grade 3 and 0% grade 4. Only 5.3% of patients required packed red blood cell transfusion. Grade 3 digestive tract toxicity occurred in 5.5% of patients. Other toxicities included elevated transaminase,oral mucositis and skin rashes. Conclusions: The regimen of lobaplatin and pemetrexed is modestly active in metastatic breast cancer patients who failed anthracycline or taxanes, and the toxicity profile suggesting that the doses of chemotherapy should be further modified.

        스파이럴형 풍력터빈 블레이드의 설계 및 공력특성에 관한 연구

        This paper describes a new design of small-scale horizontal wind blade, called spiral wind turbine blade. Theoretical and numerical approaches on the prediction of aerodynamic performance of the blade have been conducted. A theoretical equation is successfully derived using the angular momentum equation to predict aerodynamic characteristics according to the design shape parameters of spiral blade. To be compared with the theoretical value, a numerical simulation using ANSYS CFX v12.1 is performed on the same design with the theoretical one. Large scale tip vortex is captured and graphically presented in this paper. The TSR-C<SUB>p</SUB> diagram shows a typical parabolic relation in which the maximum efficiency of the blade approximately 25% exists at TSR=2.5. The numerical simulation agrees well with that of the theoretical result except at the low rotational speed region of 0~20 rad/s.

        Infection Control and Management Strategy for COVID-19 in the Radiology Department: Focusing on Experiences from China

        Coronavirus disease 2019 (COVID-19) is a new infectious disease rapidly spreading around the world, raising global public health concerns. Radiological examinations play a crucial role in the early diagnosis and follow-up of COVID-19. Cross infection among patients and radiographers can occur in radiology departments due to the close and frequent contact of radiographers with confirmed or potentially infected patients in a relatively confined room during radiological workflow. This article outlines our experience in the emergency management procedure and infection control of the radiology department during the COVID-19 outbreak.

        This research describes a new design method of small-scale wind turbine blade, called Archimedes spiral wind turbine blade. Numerical approaches on the prediction of aerodynamic performance of the blade have been conducted. Before using the PIV experiment to measure the aerodynamic characteristics, a numerical simulation using ANSYS CFX is performed on the special design. Large scale tip vortex is captured and graphically addressed in this research. In order to proving the results of numerical simulation, Particle Image Velocimetry (PIV) has been used to measure the aerodynamic characteristics of the Archimedes spiral wind turbine blade. Detail velocity vector fields around the blade are obtained and compared to those predicted by the CFD analysis. Mean velocity profiles are in a good agreement between experiment and CFD, the trajectory and magnitude of tip vortices generated by the blade are identified from the PIV results.
