      • ST6Gal-I Predicts Postoperative Clinical Outcome for Patients with Localized Clear-cell Renal Cell Carcinoma

        Hyperactivated ${\alpha}2$-6-sialylation on N-glycans due to overexpression of the Golgi enzyme ${\beta}$-galactoside: ${\alpha}2$-6-sialyltransferase (ST6Gal-I) often correlates with cancer progression, metastasis, and poor prognosis. This study was aimed to determine the association between ST6Gal-I expression and the risk of recurrence and survival of patients with localized clear-cell renal cell carcinoma (ccRCC) following surgery. We retrospectively enrolled 391 patients (265 in training cohort and 126 in validation cohort) with localized ccRCC underwent nephrectomy at a single center. Tissue microarrays were constructed for immunostaining of ST6Gal-I. Prognostic value and clinical outcomes were evaluated. High ST6Gal-I expression was associated with Fuhrman grade (p<0.001 and p=0.016, respectively) and the University of California Los-Angeles Integrated Staging System (UISS) score (p=0.004 and p=0.017, respectively) in both cohorts. Patients with high ST6Gal-I expression had significantly worse overall survival (OS) (p<0.001 and p<0.001, respectively) and recurrence free survival (RFS) (p<0.001 and p=0.002, respectively) than those with low expression in both cohorts. On multivariate analysis, ST6Gal-I expression remained associated with OS and RFS even after adjusting for the UISS score. Stratified analysis suggested that the association is more pronounced among patients with low and intermediate-risk disease defined by the UISS score. High ST6Gal-I expression is a potential independent adverse predictor of survival and recurrence in ccRCC patients, and the prognostic value is most prominent in those with low and intermediate-risk disease defined by the UISS score.

        A Study of the Content Validity of Reading Comprehension Tests in NMET II

Based on Bachman and Palmer’s (1996) framework of task characteristics, this paper aims to compare the reading comprehension tests of the 2013-2017 National Matriculation English Test II (NMET II) with the requirements of the New Curriculum Standards and Testing Syllabus, in particular, concerning the content validity of the tests in terms of the characteristics of input and expected responses. The implications of this study are discussed for test designers, teachers, and students. The results show that reading comprehension tests from 2013 to 2017 basically meet the requirements of the New Curriculum Standard and Testing Syllabus and have a high content validity. In terms of the characteristics of input, the length, reading speed, and readability of comprehension passages are appropriate for students; however, the distribution of topics and genres in each year is uneven. In terms of the characteristics of expected responses, the wordings of questions and the design of question items are reasonable. However, the response type is mainly restricted to selected response questions (multiple-choice), and the measure of reading skills coverage is unbalanced.

        De novo assembly of the transcriptome for Greenbug (Schizaphis graminum Rondani) and analysis on insecticide resistance-related genes

        The greenbug, Schizaphis graminum Rondani is a major pest species of wheat crops. In this study, a transcriptome sequencing, and the expression of the 12 genes related to insecticide resistance were conducted in S. graminum. The sequencing and subsequent bioinformatics analysis outputed 46,593 unigenes, among which 28,289 unigenes were annotated to corresponding functions by blasting with high homologous genes in database, giving annotation rate of 60.72%. To gain insight into the mechanism of insecticide resistance, the expression of the 12 genes related to insecticide resistance for S. graminum was investigated. The expression level of aminopeptidase N (AN), cytochrome P450 (CYP), acetylcholinesterase 1 (AC), catalase (CAT), cytochrome c oxidas (CCC), GABA receptor (GABA), glutathione S-transferase (GST) were highest in the apterous nymphs among different developmental stages; The expression level of AN, CBL, CYP, CA, SD, and GST were relatively more abundant in the abdomen compared to head and throax. The results could give out the key information about the relationship between the expression of these genes in different developmental stages, tissues, treatments and metabolism of insecticides. These genes that were co-up-regulated significantly in third instar nymphs of S. graminum induced by imidacloprid, were consistent with their putative involvement in insecticide resistance. This provides most comprehensive transcriptome data for S. graminum to further study and managenment.

        There is growing evidence that the coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is associated with increased risks of psychiatric sequelae. Depression, anxiety, cognitive impairments, sleep disturbance, and fatigue during and after the acute phase of COVID-19 are prevalent, long-lasting, and exerting negative consequences on well-being and imposing a huge burden on healthcare systems and society. This current review presented timely updates of clinical research findings, particularly focusing on the pathogenetic mechanisms underlying the neuropsychiatric sequelae, and identified potential key targets for developing effective treatment strategies for long COVID. In addition, we introduced the Formosa Long COVID Multicenter Study (FOCuS), which aims to apply the inflammation theory to the pathogenesis and the psychosocial and nutrition treatments of post-COVID depression and anxiety.

      • Towards Spammer Detection in Microblogging Platforms

        Spamming on microblogging platforms has been a critical issue in microblog-based applications, because spamming has a significant impact on information quality and credibility. In this paper, we characterize two types of spammers on microblogging platforms, namely advertised spammers and following spammers, and then present preliminary approaches to detect these spammers. We first use a real data set to characterize the features of AS and FS in terms of various aspects such as profile, behavior, and social relationship. Specially, we introduce a new feature named duplication for FS detection, which describes the duplicated behavior of users in sharing information on microblogging platforms. We present a content-sharing graph to model the relationship between users and microblogging contents, and propose an effective algorithm to calculate the duplication feature. We run several classification methods on the characterized features to test the effectiveness of the features in AS and FS detection. The results w.r.t. precision, recall, F-measure, and ROC suggest the effectiveness of our proposed features. In particular, the duplication feature is able to improve the effectiveness of FS detection.

        <P>HtrA2/Omi is a mammalian mitochondrial serine protease homologous to the E. coli HtrA/DegP gene products. Recently, HtrA2/Omi was found to have a dual role in mammalian cells, acting as an apoptosis-inducing protein and being involved in maintenance of mitochondrial homeostasis. By screening a human brain cDNA library with A beta peptide as bait in a yeast two-hybrid system, we identified HtrA2/Omi as a binding partner of A beta peptide. The interaction between A beta peptide and HtrA2/Omi was confirmed by an immunoblot binding assay. The possible involvement of HtrA2/Omi in A beta peptide metabolism was investigated. In vitro peptide cleavage assays showed that HtrA2/Omi did not directly promote the production of A beta peptide at the beta/gamma-secretase level, or the degradation of A beta peptide. However, overexpression of HtrA2/Omi in K269 cells decreased the production of A beta40 and A beta42 by up to 30%. These results rule out the involvement of HtrA2/Omi in the etiology of Alzheimer's disease. However, the fact that overexpression of HtrA2/Omi reduces the generation of A beta40 and A beta42 suggests that it may play some positive role in mammalian cells.</P>

        Research and Application of Fiber Bragg Grating Sensor in Geological Disaster Automation Monitoring

        In this paper, fiber grating technology is used for accurately measure stresses, temperature and displacement. The paper introduces the basic principles of FBG technology and the wavelength demodulation method of FBG sensors, and put fiber grating technology into practical engineering projects. The paper design a optical fiber grating real-time automation monitoring system to distributed measure stress, temperature and displacement in geological disaster body, to provide effective technical supports for geological disaster prevention and mitigation.

        Clusterin negatively modulates mechanical stress-mediated ligamentum flavum hypertrophy through TGF-β1 signaling

        Ligamentum flavum hypertrophy (LFH) is a major cause of lumbar spinal canal stenosis (LSCS). The pathomechanisms for LFH have not been fully elucidated. Isobaric tags for relative and absolute quantitation (iTRAQ) technology, proteomics assessments of human ligamentum flavum (LF), and successive assays were performed to explore the effect of clusterin (CLU) upregulation on LFH pathogenesis. LFH samples exhibited higher cell positive rates of the CLU, TGF-β1, α-SMA, ALK5 and p-SMAD3 proteins than non-LFH samples. Mechanical stress and TGF-β1 initiated CLU expression in LF cells. Notably, CLU inhibited the expression of mechanical stress-stimulated and TGF-β1-stimulated COL1A2 and α-SMA. Mechanistic studies showed that CLU inhibited mechanical stress-stimulated and TGF-β1-induced SMAD3 activities through suppression of the phosphorylation of SMAD3 and by inhibiting its nuclear translocation by competitively binding to ALK5. PRKD3 stabilized CLU protein by inhibiting lysosomal distribution and degradation of CLU. CLU attenuated mechanical stress-induced LFH in vivo. In summary, the findings showed that CLU attenuates mechanical stress-induced LFH by modulating the TGF-β1 pathways in vitro and in vivo. These findings imply that CLU is induced by mechanical stress and TGF-β1 and inhibits LF fibrotic responses via negative feedback regulation of the TGF-β1 pathway. These findings indicate that CLU is a potential treatment target for LFH.

        Today, the massive development and blossom of the culture and creative industry has been an important symbol of the social fast development and innovation. More and more international cities are improving the development of culture and creative industries in order to upgrade the city’s culture intention and promote the city’s brand image. In this article, it’s claimed that the Chengdu city aims at developing into an influential and internationalized metropolitan, whereas the culture and creative industry should be emphasized with individuation and hence, the city’s culture intention can be broaden, with improved culture image and the culture industry development. Meanwhile, the service platform for culture and creative industry in Chengdu is the gap point and bottle neck for the industry development for the time being. This article analyzes the current situation of Chengdu’s culture and creative industry development and proposes suggestions in regard of the providing sector cultivation, and further counter measures are suggested in terms of the information platform development for the culture and creative industry in Chengdu, including the major functional structure.

        In vitro stability evaluation of coated lipase

        Objective: The study was conducted to evaluate the stability of commercial coated lipase (CT-LIP) in vitro. Methods: The capsules were tested under different conditions with a range of temperature, pH, dry heat treatment and steaming treatment, simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) in this work, respectively. Free lipase (uncoated lipase, UC-LIP) was the control group. Lipase relative activities measured in various treatments were used as a reference frame to characterize the stability. Results: The lipase activities were decreased with increasing temperatures (p<0.05), and there was a markedly decline (p<0.01) in lipase comparative activities of UC-LIP at $80^{\circ}C$ compared with CT-LIP group. Higher relative activities of lipase were observed in CT-LIP group compared with the free one under acidic ambient (pH 3 to 7) and an alkaline medium (pH 8 to 12). Residual lipase activities of CT-LIP group were increased (p<0.05) by 5.67% and 35.60% in dry heat and hydrothermal treatments, respectively. The lipase relative activity profile of CT-LIP was raised at first and dropped subsequently (p<0.05) compared with constantly reduced tendency of UC-LIP exposed to both SGF and SIF. Conclusion: The results suggest that the CT-LIP possesses relatively higher stability in comparison with the UC-LIP in vitro. The CT-LIP could retain the potential property to provide sustained release of lipase and thus improved its bioavailability in the gastrointestinal tract.
