인간의 말초혈액에서 염증성 사이토카인에 의한 Th17 세포의 분화에 미치는 영향

Objective: IL-17-producing T cells (Th17 cells) have been identified as a distinct lineage of CD4+ T helper cells in mice. Since this discovery, many efforts have been made to investigate the characteristics and the role of human Th17 cells and the factors involved in their differentiation. This study was undertaken to assess the effects of cytokines and stimulatory conditions on the differentiation of human CD4+ T cells into Th17 cells. Methods: Peripheral blood CD4+ T cells were isolated from healthy humans and then these cells were cultured with using various stimulatory conditions. The Th17 cells and regulatory T (Treg) cells were detected by flow cytometry (FACs). The related gene expressions of cytokines, transcription factors and chemokine receptors were determined by ELISA and RT-PCR. Results: In the presence of inflammatory cytokines, TNFa and IL-1b, the human CD4+ T cells rapidly produced IL-17 in response to anti-CD3/anti-CD28 stimulation, whereas, with anti-CD3/ anti-CD28 stimulation alone, the CD4+ T cells expressed low levels of IL-17. TNFa and IL-1b were also important inducers of IL-22 production. IL-6 and IL-23 up-regulated the RORgammat, CCR4 and CCR6 expressions in the human CD4+ T cells. In response to TGF-b and IL-2, the human CD4+ T cells were rapidly induced to express FoxP3, IL-10 and CCR7, as compared with anti-CD3/anti-CD28 stimulation alone. Conclusion: The effect of inflammatory cytokines on the differentiation of human Th17 cells may help us to understand their pathogenic role. Moreover, the differential expression of chemokine receptors and transcription factors of the subsets of CD4+ T cells with the different features of Th17 and Treg, may raise new issues concerning the pathogenesis of autoimmune inflammatory diseases.
영역별 투영 히스토그램 매칭 및 선형 회귀모델 기반의 차량 운행 영상의 안정화 기술 개발

본 논문에서는 블랙박스 혹은 운전석에 장착된 카메라로부터 얻어진 차량 영상에 대한 영역별 수직히스토그램 매칭 및 선형 회귀분석 모델(linear regression model)을 활용한 강건한 차량 운행 동영상의 안정화(video stabilization) 기법을 제안한다. 동영상 안정화 기법은 영상의 흔들림 보정뿐 아니라 동영상 내 강건한 특징점 추적 및 매칭을 위한 이전의 전처리 과정으로 적용된다. 일반적으로 촬영 과정에서 많은 떨림이 포함될 수 있는 야외 CCTV 영상이나 손으로 들고(hand-held) 촬영된 동영상에 대한 흔들림 보정 등에 적용되고 있으나 영상 내 특징점이 지속적으로 변하고 영상의 변화 정도가 매우 심한 차량 운행 동영상에서는 적용된 사례가 드물다. 본 연구에서는 일반적인 비디오 안정화 기술이 적용되기 어려운 차량 운행 동영상에 대하여 수직 투영 히스토그램 매칭 및 선형 회귀분석 모델 기반의 안정화 기법을 제안한다. 제안된 기법은 입력영상에 대한 영역별 수직 투영 히스토그램 매칭을 수행하고 선형 회귀모델을 통해 영상에 나타나는 수직 및 회전이동 변환을 선형 근사하여 시간 영역 상의 입력 영상에 대한 안정화를 달성한다. 제안 방법의 검증을 위해 블랙박스로 촬영된 실제 동영상에 동영상 안정화 기술을 적용하였으며, 운행 중 불규칙한 노면으로 인한 영상의 흔들림이 효과적으로 제거되는 것을 확인할 수 있었다.

Autoimmune arthritis, such as rheumatoid arthritis (RA), is a chronic inflammatory disorder that primarily affects the joints and then results in their progressive destruction. Effector Th cells have been classified as Th1 and Th2 subsets based on their cytokine expression profiles and immune regulatory function. Another subset of T cells termed Th17 was recendy discovered and known to selectively produce IL-17. Also, Th17 was shown to be generated by TGF${\beta}$ and IL-6 and maintained by IL-23. IL-17 is a proinflammatory cytokine that is considered to involve the development of various inflammatory autoimmune diseases such as RA, asthma, lupus, and allograft rejection. IL-17 is present in the sera, synovial fluids and synovial biopsies of most RA patient. IL-17 activates RA synovial fibroblasts to synthesize IL-6, IL-8 and VEGF via PI3K/Akt and NF-${\kappa}B$ dependent pathway. IL-17 increases IL-6 production, collagen destruction and collagen synthesis. In addition, it not only causes bone resorption but also increases osteoclastogenesis and fetal cartilage destruction. Inhibition of the IL-17 production may contribute a novel therapeutic approach along with potent anti-inflammatory effect and with less immunosuppressive effect on host defenses.
