      • Proteus 균종에서 플라스미드 pBluescript의 전기적 재결합에 관한연구

        In order to genetic analysis of microorganisms, electroporation method was applied to induce electrotransformation of two strains of Proteus mirabilis (API code: 0536000 and 0737000). isolated from patients at Wonkwang University Hospital. In this study, we tried to evaluate the effect of parameters, ie amount of competent cell and plasmid DNA(pBluescript) on the survival of transformant and transformation efficiency and examine antibiotic susceptibility patterns and biochemical properties of tested strains before and after electroporation. The results were as follows: 1. The effect of amount of competent cells and plasmid DNA to transformation efficiency were different between strains. But the highest efficiency was noted at the amount of pBluescript 20ng. 2. In order to confirm the transformant, pBluescript before and after electroporation were isolated from colonies on the LB agar containing ampicillin and electrophoresed. Two plasmids showed same sized DNA. 3. Antibiotic susceptibility to ampicillin was changed from susceptible to resistant in both strains and to carbenicillin. changed from susceptible to intermediate.
