PURPOSE: Strengthening training is effective at improving the function of stroke patients. This study was conducted to compare the effects of exercise intensity on walking, balancing ability, and blood glucose in stroke patients with type 2 diabetes mellitus and to suggest more effective exercise intensity for stroke patients. METHODS: Twenty-two patients with type 2 diabetes mellitus were divided into an experimental group (n=11) and a control group (n=11). In the experimental group, a 40-70% strength of 1RM exercise program was applied for 40 minutes a day, three times a week for 4 weeks. The control group received a 50% intensity of 1RM for the same number and duration of interventions as the experimental group. RESULTS: Both the experimental group and the control group showed significant improvement in the gait function, balance ability, and blood glucose level after the intervention compared to before the intervention. In the FGA (functional gait assessment), the experimental group showed a 52.4% greater increase than the control group, and the fasting blood glucose level was 16.7% greater in the experimental group than the control group. CONCLUSION: The strengthening exercise program, which gradually increases exercise intensity, seems to be effective at improving the function of stroke patients with type 2 diabetes.
배양 인체피부섬유모세포에 있어서 알러지성 접촉피부염 유발제인 구리에 대한 쇠비름 추출물의 항산화 효과

To elucidate the cytotoxicity and antioxidative effect of Portulaca oleracea L. (PO) extract on copper (Cu), an allergic contact dermatitis(ACD)-induced agent, cell viability were analysed by XTT assay after the treatment of cupric sulfate (CuSO4) to cultured human skin fibroblasts (Detroit 551). And also, the effects of vitamin E and PO extract on CuSO4- induced cytotoxicity were measured after Detroit 551 cells were pretreated with vitamin E or PO extract for 2 hours. In the antioxidative effects of PO extract were measured by superoxide dismutase (SOD)-like activity, the inhibitory activity of lipid peroxidation (LP) and lactate dehydrogenase (LDH) activity. In this study, CuSO4 significantly decreased cell viability in dose-dependently compared with control, and the XTT50 value was calculated at 59.8μM of CuSO4. While, vitamin E effectively prevented CuSO4-induced cytotoxicity and also, PO extract significantly increased cell viability compared with CuSO4-treated group. In the antioxidative effect, PO extract showed the inhibitory activity of LP and the decrease of LDH activity as well as SOD-like activity. From these results, it is suggested that the PO extract effectively prevented the cytotoxicity induced by an ACD-induced agent, CuSO4 via antioxidative effect. Conclusively, plant extract such as PO may be a putative resources as an alternative antioxidant for the prevention or diminution of ACD induced by allergen of heavy metals correlated with oxidative stress.

본 연구는 미용제에서 정균제 기능을 가지고 있는 수은에 대한 세포독성을 산화적 손상 측면에서 조사하기 위하여 배양 인체피부흑색종세포 (SK-MEL-3)를 사용하여 HgCl2의 독성을 산화적 손상 측면에서 조사하고 이와 동시에 수은의 독성에 대한 CM 추출물의 영향을 항산화 및 멜라닌화에 대하여 분석함으로서 미용소재로서의 효능을 알아보고자 하였다. To clarify the effect of Chelidonium majus (CM) extract on mercury (Hg)-induced cytotoxicity and melanogenesis, cell viability was done by XTT assay after human skin melanoma cells (SK-MEL-3) were cultured in media containing various concentrations of mercuric chloride (HgCl2). And also, the effect of vitamin E on HgCl2-induced cytotoxicity was assessed. For the protective effect of CM extract on HgCl2-induced cytotoxicity and melanogenesis, SK-MEL-3 cells were pretreated with 50 or 80 μg/mL of CM extract for 2 hours, and also, the lactate dehydrogenase (LDH) activity for antioxidative effect as well as the melanin synthesis and tyrosinase activity for the melanogenesis on CM extract were analysed. In this study, HgCl2 significantly decreased cell viability in dose-dependent way compared with control, and the XTT50 value was calculated at 29.1 μM of HgCl2. In the protective effect of vitamin E, it effectively prevented HgCl2- induced cytotoxicity by a significant increase of cell viability. In the protective effect of CM extract on HgCl2-induced cytotoxicity, CM extract significantly increased cell viability which was decreased by HgCl2-induced cytotoxicity, and also it showed the inhibitory effect of LDH activity. While, in the melanogenesis of CM extract, CM extract remarkably decreased melanin synthesis and tyrosinase activity compared with HgCl2-treated group. From these results, it suggested that HgCl2-induced cytotoxicity was involved in oxidative stress, and also, CM extract effectively prevented HgCl2- induced cytotoxicity and melanogenesis. Conclusively, plant extract such as CM may be a putative beauty resources for the prevention or diminution of the toxicity or melanogenesis induced by heavy metals correlated with oxidative stress.
