        Objectives: Dementia is a disease in which a person maintains a normal intellectual level during the growth period, but has acquired cognitive impairment and personality change. In this study, we tried to check whether the network pharmacology analysis method is useful in the search for herbal medicine resources for the treatment of dementia. Methods: The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) database is a database frequently used in Chinese medicine research. We used the TCMSP to identify herbal medicines and their molecular targets that can be used for dementia by using network pharmacology research methods. Results: It was possible to select 28 types of components that are expected to be active by applying them to the living body, and 75 types of targets that these components act on were secured. In addition, 16 kinds of drugs were identified by checking the drugs containing 28 kinds of ingredients, and it was found that Radix Salviae contained 2 kinds of the selected 28 kinds of ingredients. Conclusions: Through this study, we were able to identify ingredients, drugs, and targets that can be used for basic and clinical research on dementia.

      • 생쥐의 하악 치아 절단으로 인한 부정교합이 뇌경색에 미치는 영향

        Objectives: Although intraoral balancing appliance therapy has been used effective to several diseases, verification studies through cerebral diseases are poorly reported so far. Thus we investigated the effect of tooth-cut induced dental malocclusion against mouse model of ischemic stroke. Methods: Tooth-cut and 90 min middle cerebral artery occlusion (MCAO) were loaded to C57BL/6 male mice, and total infarct area, neurological deficit scores (NDS), histological change of hippocampal region were observed. Production levels of reactive oxygen species (ROS) and inducible nitric oxide synthase (iNOS) in cerebral tissue were also measured. Results: The longer the tooth-cut period, the greater the area of cerebral infarction caused by MCAO, and NDS began to increase as the tooth was cut, and the results were more negative when MCAO was loaded. Histological change of hippocampal cells was significant when tooth-cut was maintained for 7 days. Those damages were thought to depend on the generation of ROS and iNOS in brain tissue. Conclusions: Since tooth-cut increased total area of cerebral infarction due to MCAO in mice, it is able to be confirmed that anomaly of the temporomandibular occlusion can affect neurological diseases.

        인삼(人蔘)과 홍삼(紅蔘)의 네트워크 약리학적 분석 결과 비교

        Objectives : Network pharmacology analysis is commonly used to investigate the synergies and potential mechanisms of multiple compounds by analyzing complex, multi-layered networks. We used TCMSP and BATMAN-TCM databases to compare results of network pharmacological analysis between White Ginseng(WG) and Red Ginseng(RG). Methods : WG and RG were compared with components and their target molecules using TCMSP database, and compound-target-pathway/disease networks were compared using BATMAN-TCM database. Results : Through TCMSP, 104 kinds of target molecules were derived from WG and 38 kinds were derived from RG. Using the BATMAN-TCM database, target pathways and diseases were screened, and more target pathways and diseases were screened compared to RG due to the high composition of WG ingredients. Analysis of component-target-pathway/disease network using network analysis tools provided by BATMAN-TCM showed that WG formed more networks than RG. Conclusions : Network pharmacology analysis can be effectively performed using various databases used in system biology research, and although the materials that have been reported in the past can be used efficiently for research on diseases related to targets, the results are unreliable if prior studies are focused on limited or narrow research areas.

        Recently, clinical efficacies of the intraoral balancing appliance therapy have been reported by several researchers, and it has been found that there are various kinds of diseases that can be effectively applied. However, studies on cerebrovascular disease, one of the main diseases with a high mortality rate, are still poorly reported, and studies of temporomandibular joint disorder (TMD)-induced changes in brain function suggest that cerebrovascular disease is more appropriate as an adaptive disorder of the temporomandibular joint (TMJ) balancing device. In the developed countries, the importance of research on the structure and function of the brain has been recognized and spurred on the related research. In Korea, the research on brain function and cognitive disorders should have promoted more massively. In order to regain its former reputation in the Korean medicine in the field of cerebrovascular disease, it should be spurred on basic research and clinical case studies. In addition, extensive and in-depth studies including animal studies are needed to establish the basis of underlying mechanisms of the TMJ balancing therapies.

      • 네트워크 약리학 분석을 통한 뚜렛 증후군에 유용할 것으로 예측되는 한약 자원 탐색

        Objectives: Tourette syndrome (TS) is a disease that occurs evenly in many social classes. Despite the long experience of drug treatment, the preference is low due to various side effects. The aim of this study was to discover herbal medicine resources through network pharmacology analysis predicted to be useful for Tourette syndrome. Methods: We used Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) to identify herbal medicines that can be used for TS by using network pharmacology research methods and to predict the mechanism of action. After evaluating compounds of each identified herb, molecular target proteins and mechanisms of action were analyzed, focusing on compounds that are likely to exhibit clinical activity in consideration of the pharmacokinetic parameters of these individual compounds. Results: Fifty nine ingredients such as atropine, veraguensin, and nuciferin among the compounds contained in 48 types of medicinal herbs such as Daturae Flos (洋金花), Salviae Radix (丹参), and Nelumbinis Plumula (蓮子心) act on the D(2) dopamine receptor, which is a protein involved in the development of TS. It has been found that atropine, veraguensin, and nuciferin are highly likely to exhibit activity by acting on the G protein-coupled receptor signaling pathway. Conclusions: It can be used in conjunction with non-invasive treatment means such as FCST Yinyang Balancing Appliance with herbal therapy to bring about a significant therapeutic effect, and it will be possible to develop a treatment that can replace drug therapy used in Western medicine.

        코로나19 유행 상황에서 온라인 학습을 경험한 고학년 간호대학생의 임상수행능력에 영향을 미치는 요인

        Purpose: This study identifies factors affecting the clinical competence of senior nursing students who experience online learning during the coronavirus disease 2019 (COVID-19) pandemic. Methods: In this study, 180 nursing students from third and fourth grades in South Korea participated from June 25 to July 3, 2021. Study data were collected through an online survey using self-report questionnaires that contained items on online learning satisfaction, online learning participation, learning immersion, learning transfer, and the clinical competence scale. Data analysis was performed using the SPSS 26.0 program; descriptive statistics, Pearson s correlation coefficients, and stepwise multiple regression were applied, as well. Results: Participants’ clinical competence averaged 3.80±0.55 on the 5-point scale, The factors with the most influence on the clinical practicum were learning transfer (=.398, p <.001) and online learning participation (=.316, p <.001), and the model’s explanatory power was 25.5% (F=16.30, p <.001). Conclusion: The study results provided a foundation to develop an efficient online intervention program to improve senior nursing students’ clinical competence during the COVID-19 pandemic.

        Object : As though the roots of Stephania tetrandra and Cocculus trilobus are used for medicinal purposes, climbing stems of these plants which do not have medicianl value are often adulterated and distributed in markets. Therefore, distinguishing the medicinal parts of these plants from the non-medicinal parts is necessary. Method : Samples of Stephania tetrandra and Cocculus trilobus were collected and authenticated. They were separated into root(StR, CtR) and climbing stem(StS, CtS), and then examined using stereoscope and microscope. Result : 1. Morphological characteristics: StR contained abundant powder and had a lotus flower-patterned section whereas StS had less powder and a wheel-patterned section. CtR had an irregular longitudinally wrinkled surface and a superb pink flower-patterned section whereas CtS had a regular furrowed surface and wheel-patterned section. 2. Anatomical characteristics: StR was abundant in starch grains and crystals (calcium oxalate), whereas StS lacked these features. In contrast to CtS, the width of the CTR medullary rays was equal or greater than that of the wood rays. Additionally, CtS contained high number of crystals but CtR did not. Conclusion : Morphological and anatomical characteristics are useful for identifying non-medicinal climbing stems of S. tetrandra and C. trilobus.

        Dracocephalum palmatum Stephan 잎 헥산 분획 추출물의 인간 유래 전립선 암세포 사멸에 대한 작용

        Objectives : Dracocephalum palmatum Stephan (Lamiaceae) is a medicinal plant used by the East-Russian nomads but there were few studies on this plant. This study was to evaluate anti-cancer effects of D. palmatum Stephan leaf hexane fraction on human derived prostate cancer cell death. Methods : The dried leaves of D. palmatum were dissolved in methanol, and hexane fraction (DpLH) was again obtained from lyophilized methanol extract (DpLM). DpLH was investigated by measuring by MTT assay and annexin V/PI staining to evaluate its effects on the cell viability and apoptosis of PC-3 cells. The ROS generations were detected by DCF-DA dye. The protein expressions were confirmed by p-AKT, Bcl-2, Bax, procaspase-3 activities. Results : After treatment of DpLH to PC-3 cells, the cell proliferation was significantly inhibited, and in addition, DpLH treatment also accelerated apoptosis of PC-3 cells. When DpLH was treated to the PC-3 cells, its ROS production significantly decreased. The proportion of all proteins (p-AKT/actin, Bcl-2/Bax and procaspase-3/actin ratios) showed decreasing tendency of expression compared with the control group. Conclusions : As shown in the above results, the extract from D. palmatum inhibits ROS production and promotes cell death, which is considered to be a relatively safe induction of cell death when administered to a living body. In conclusion, these results suggested that DpLH may have anti-cancer effect in human prostate cancer cell.

        상백피(桑白皮) 메탄올 추출물 전처치가 일과성 허혈에 의한 생쥐의 뇌 손상에 미치는 영향

        Objectives : Mori Radicis Cortex (MRC), the root epidermis of Morus alba L., has been traditionally used to treat lung-related diseases in Korean Medicine. The common of MRC is Mulberry bark Morus bark, and it`s pharmaceutical properties and taste are known as sweet and cold, and it promotes urination and reduce edema by reducing heat from the lungs and soothe asthma. In the present study, anti-apoptotic mechanism of MRC in middle cerebral artery occlusion (MCAO) model in mice. Methods : Two-hundred grams of MRC was extracted with methanol at room temperature for 5 days, and this was repeated one time. After filtration, the methanol was removed using vacuum evaporator, then stored at -20℃ until use. C57BL/6 male mice were housed in an environment with controlled humidity, temperature, and light cycle. In order to determine beneficial effects of MRC on ischemia induced brain damage, infarct volume, neurological deficit scores, activities of several apoptosis-related proteins such as caspase-8, -9, Bcl-xL in MCAO-induced brains of mice were analyzed. Mice in MRC-treated groups were orally administered 30, 100, or 300 mg/kg of body weight for three consecutive days before commencing the MCAO procedure. Results : Pre-treatment of MRC significantly reduced infarct volume in MCAO subjected mice applied with 300 ㎎/ ㎏ of MRC methanol extract, and MRC effectively inhibited Bcl-xL reduction and caspase-9 activation caused by MCAO-induced brain damage. Conclusions : MRC showed neuro-protective effects by regulating apoptosis-related protein signals, and it can be a potential candidate for the therapy of ischemia-induced brain damage.
