        We conducted experiments on mask filters. We measured filtering efficiencies of several new mask filters which were manufactured by disassembling and reassembling of one type of mask filter. New filter (A+C: combination of the first layer and the third layer of the tested mask filter) showed the highest efficiency (97.7%) with the respiratory resistance of 98 pa.

        HIV-1 유래 렌티바이러스 벡터의 복제가능 바이러스 검출과 역가측정 분석방법 비교

        Human Immunodeficiency Virus type 1 (HIV-1) based lentivirus vector has demonstrated great potential as gene therapy vectors mediating efficient gene delivery and long-term transgene expression in both dividing and nondividing cells. However, for clinical studies it must be confirmed that vector preparations are safe and not contaminated by replication competent lentivirus (RCL) related to the parental pathogenic virus, HIV-1. In this study, we would like to establish the method for titration and RCL detection of lentivirus vector. The titration was determined by vector expression containing the green fluorescent protein, GFP in transduced cells. The titer was 1×107 Transducing Unit/ml in the GFP expression assay and 8.9×107 molecules/ml in the real-time PCR. Also, for the detection of RCL, we have used a combination method of PCR and p24 antigen detection. First, PBS/psi and VSV-G region in the genomic DNA of transduced cells was detected by PCR assay. Second, transfer and expression of the HIV-1 gag gene was detected by p24 ELISA. In an attempt to amplify any RCL, the transduced cells were cultured for 3 weeks (amplification phase) and the supernatant of amplified transduced cell was used for the second transduction to determine whether a true RCL was present (indicator phase). Analysis of cells and supernatant at day 6 in indicator phase were negative for PBS/psi, VSV-G, and p24 antigen. These results suggest that they are not mobilized and therefore there are no RCL in amplification phase. Thus, real-time PCR is a reliable and sensitive method for titration and RCL detection of lentivirus vector.

        외래성 Minute Virus of Mice 검출을 위한 시험법 개발에 관한 연구

        In manufacturing of biotechnological products, numerous adventitious agents might have been detected in cell substrates. Recently a biotech-pharmaceutical company has had experienced with contamination of large-scale cell cultures by minute virus of mice (MVM) and this resulted in both the loss of product and the need for major cleaning validation procedures to be put in place. We have developed a simple PCR assay to detect the presense of MVM in cell culture supernatant. The specificity of MVM was determined by testing viral preparations of other autonomous parvovirus and MVM specific PCR assay detects only in the presence MVM viral DNA. This assay has proven to be very sensitive and it can detect 10 or fewer genome equivalents (copies) of MVM. These results implicate that we may use this assay for routine screening test for MVM. The assay also can detect MVM following extraction procedure of DNA in cell lines at a concentration of 1 TCID₅₀/㎖ and establishes a sensitive, specific assay that can detect the presense of MVM sequences in a cell culture system.

        어린이 기호식품 중 트랜스지방 및 포화지방 실태조사

        어린이 기호식품 중 트랜스지방 및 포화지방 함량을 조사하고 그 지방산 조성을 확인하기 위하여 과자류 134건, 제빵류 55건, 도넛류 74건, 햄버거 60건(20종)을 서울 및 경기지역을 중심으로 수거하여 분석하였다. 조지방 함량은 과자류가 24.2±6.9(범위: 4.6~41.1)g/100 g으로 가장 높았고, 도넛 23.9±5.8(범위: 14.1~39.5), 제빵류 15.7±7.9(1.4~30.0), 햄버거 9.5±3.4(4.5~18.5)의 순이었다. 트랜스지방 최고값은 제빵류에서 1.3 g/100 g으로 업계의 저감화 노력을 확인할 수 있었다. 포화지방은 과자류가 11.6±4.8(범위: 2.0~22.7) g/100 g, 도넛 11.2±4.0(범위: 4.8~23.2) g/100 g, 제빵류 6.9±4.1(범위: 0.6~15.4) g/100 g, 햄버거 3.0±1.0(1.0~5.8) g/100 g의 순이었으며 조지방 함량에 의존성이 있는 것으로 확인하였다. 지방산 중 포화지방산 및 불포화지방산의 비율은 제품의 특성에 따라 매우 다양하게 나타났으며 식물성 유지를 사용하여 유탕하는 제조공정의 제품군이 다른 제품과 비교 시 포화지방산이 낮은 경향을 나타내었다. We studied the amounts of trans fatty acids and saturated fatty acids in child-favored foods. Confectioneries, breads, donuts and hamburgers were included in this study. The total fatty acid content and the amount of each type of fatty acid were determined by gas chromatography with a flame-ionized detector. Confectioneries were shown to have the highest content of crude fat. The mean content was 24.2±6.9 (range: 4.6~41.1) g/100 g food. The mean content of crude fat in donuts, bread and hamburgers was 23.9±5.8 (range: 14.1~39.5) g/100 g food, 15.7±7.9 (range: 1.4~30.0) g/100 g food, and 9.5±3.4 (range: 4.5~18.5) g/100 g food, respectively. Bread had the most trans fatty acids at 1.3 g/100 g food. This result inferred that manufacturers have tried to reduce the trans fat content. The mean content of saturated fatty acids in confectioneries, donuts, bread, and hamburgers was 11.6±4.8 (range: 2.0~22.7) g/100 g food, 11.2±4.0 (range: 4.8~23.2) g/100 g food, 6.9±4.1 (range: 0.6~15.4) g/100 g food, 3.0±1.0 (range: 1.0~5.8) g/100 g food, respectively. This content depended on crude fat. The composition of fatty acids varied according to the oil and fat used in the manufacturing process of each food, and the natural content. Foods that were fried in vegetable oil tended to be especially low in saturated fat.

        중기 다장기 발암성시험법을 이용한 플라스틱 가소제의 전암단계 병변에 미치는 영향

        This study was conducted to assess the modifying effects of diisodecyl phthalate (DIDP) and diethylhexyl phthalate (DEHP) in the rat medium-term multi-organ bioassay using five different carcinogens. Seventy five, 6-week-old, male F344 rats were divided into four groups. Animals were sacrificed at week 12. To evaluate modifying effects of preneoplastic lesions of DIDP and DEHP, we measured glutathione S transferase placental form (GST-P) and 5-bromodeoxyuridine (BrdU) expression level in liver, numbers of aberrant crypt foci (ACF) in colon and performed histopathological observation. The body weights of rats given 12,000 ppm DEHP and 8,000 ppm DIDP were significantly decreased compared to those of control group. Relative liver and kidney weights of rats given DIDP were significantly increased compared to those of control group in a dose dependent manner. There was no significance of GST-P positive foci and BrdU labeling indices in liver and numbers of ACF in colon among each group. Although hyperplasia of urinary bladder of rat given DEHP were significantly increased compared to that of control group, there were no significant changes of preneoplastic lesions on all organs of rats given DIDP. These results indicate that DIDP has no modifying effect on preneoplastic lesions in all organs.
