DYYVVR is a tryptic peptide of Janus kinase 3 and has the two active tyrosine residues needed for the regulation of the protein's activity by phosphorylation. As a proof‐of‐concept experiment for localization of phosphorylation site, two‐dimensional ultraviolet‐mass spectrometry of singly protonated DYYVVR has been investigated at room temperature. Comparing the absorption spectrum with those of peptides with single phenylalanine substitution, DYFVVR and DFYVVR, it is confirmed that the ultraviolet (UV) absorption below 35 100 cm −1 is due to the first tyrosine residue. Combined with the fact that the absorption of a phosphorylated tyrosine is shifted above 37 500 cm −1 , two‐dimensional analysis employing mass spectrometry and ultraviolet spectroscopy can be used as a tool for localization of phosphorylation site in a peptide.

CO2enriched air bubbling is commonly used in the conventional algal culture systems, but for offshore culture systems, supplyg CO2 is costly.The effect of NaHCO3 concentration on the biomass productivity of Tetraselmis sp. KCTC12432BP, was investigated. The microalgae were cultured using modified f/2-Si medium based on natural seawater in Floating Cube photobioreactors (FC-PBRs) in the Youngheung island,Incheon, Korea. The biomass productivities were 0.19, 0.28, and 0.38g/L/day in control, 4.0 and 8.0 g/L NaHCO3 supplemented groups, respectively.The results indicate that NaHCO3 could be used as a carbon source alternative to CO2 gas for production of Tetraselmis sp. biomass in FC-PBRs.

The ocean provide great benefits for microalgal mass cultures with maintaining stable temperature due to high specific heat, mixing by wave energy, and providing large area for large-scale microalgae cultures. In this study, we cultivated a marine green microalga, Tetraselmis sp. KCTC12432BP, using marine photobioreactors on the ocean for investigating the effect of $NaHCO_3$ concentration on the biomass productivities and evaluating the potential of ocean microalgae culture. The culture medium consist of three fold concentrated f/2-Si with 4 g/L of $NaHCO_3$, which is dissolved in natural seawater. After 11 days of cultivation, the cultures reached stationary phase at biomass concentration of 1.6 g/L. At that time, $NaHCO_3$ concentration of 0, 2, and 4 g/L were fed to the cultures. The daily productivities of 0.11, 0.19, 0.30 g/L/day were attained with feeding rate of 0, 2, and 4 g/L $NaHCO_3$, respectively. Biomass productivity of Tetraselmis sp. KCTC12432BP was a function of the $NaHCO_3$ feeding rate as expected. This research shows that the microalgae can grow with $NaHCO_3$ as carbon source in marine photobioreactors on the ocean while exploiting various benefits of ocean cultivation.
영흥도에서 분리된 Phaeodactylum tricornutum의 증식 및 Monounsaturated fatty acid 관련 지방산 조성 분석

Oxidation stability and cold fuid property are considered as the most important factors for determining biodiesel quality. Among the fatty acids, monounsaturated fatty acid satisfy both oxidation stability and cold flow property of biodiesel quality standards. Microalgae with high monounsaturated fatty acid contents is have some benefit for producing to produce biodiesels with satisfying quality standards. In this study, monounsaturated fatty acid contents of a isolated microalga from Youngheung island was analyzed. Phaeodactylum tricornutum was isolated by streaking, and growth rate and fatty acid composition of the algae were investigated. Total FAME contents were consisted of 26% of saturated fatty acids, 43% of monounsaturated fatty acids, and 18% of polyunsaturated fatty acids. The contents of monounsaturated fatty acid were especially high in the Phaeodactylum species. This result implies that the FAMEs from P. tricornutum may contribute to improve the oxidation stability and cold flow property of biodiesel.

Oxidation stability and cold fluid property are important factors in the quality of biodiesel. Monounsaturated fatty acids (MUFAs) are suitable fatty acid for biodiesel when considering the low temperature fluidity and oxidative stability in regards to the biodiesel fuel standards. In this study, MUFAs productivity of a Phaeodactylum tricornutum MBEyh07L (KCTC18366P), isolated from coastal seawater of Youngheung Island, Incheon, Korea, was investigated. The biomass was produced using bubble column photobioreactors in modified f/2-Si medium prepared with natural seawater. The highest MUFAs productivity obtained was 28.5 mg/L/d. MUFAs accounted for 35.8% of the total fatty acids produced whereas saturated fatty acids and polyunsaturated fatty acids accounted for 36.5% and 14.6% of the total fatty acids, respectively. The result implies that the lipids from P. tricornutum MBEyh07L may contribute to improving the oxidation stability and cold flow property of biodiesel.

The microalgae were cultured using modified f/2-Si medium based on natural seawater in Floating Cube Photobioreactors (FC-PBRs) in the Youngheung island, Incheon, Korea. Initially, 4.0 g/L NaHCO3 was added to the cultures, and after 11 days of cultivation, when the cultures entered stationary phase, 0.0, 2.0, 4.0 g/L NaHCO3 were supplemented additionally. The biomass productivities were 0.19, 0.28, and 0.38 g/L/day in 0.0, 2.0 and 4.0 g/L NaHCO3 supplemented groups, respectively. The results indicate that NaHCO3 could be used as a carbon source alternative to CO2 gas for production of Tetraselmis sp. biomass in FC-PBRs.
