      • 鷄卵의 受精時期 및 受精部位에 關한 硏究 (第一報)

        I incised a hen’s left side abdominal region and injected a cock’s semen to experiment the time and region of fertilization, also I injucted the cock’s semen without incicion to experiment to get the above result. The eggs which I have through these follwing methods, I incubated with incubator within three days. with the above experiments I have known the following results. 1)Injection to Ovarian Follicle. I incised a hen’s left side abdominal region and injected with tuberculin iniector a cock’s semen, 0.01CC(number of sperm; adt, 7000,000)to 10 hens 2-4 ovarian Follicle one after another begining the biggest one. Through the above experiment, I have got 25 eggs and incubated those, but all these eggs were not fertilized. 2)The Injection to Ovarian Root. I incised a hen’s left side abdominal region and injected a cock’s semen, 0.1CC(number of sperm; abt, 21,000,000)to 12 hens Ovarian through the membrane. with the above experiment I have got 85 eggs and incubated, but 13 egigs were fertilized by 5 hens and other except 13 eggs were not fertilized, and all hen’s unripe Follices were not fertillized but large Follicles were fertilized. 3)Center of magnum region. I incised a hen’s left side-abdominal region within two houres after laying an egg and injected a cock’s semen 0.1CC(number of sperm; abt, 21,000,000) to 10 hens the front region of descenting Ovum. when injecting the descenting ovum Was not fertilized, but some eggs layed after that time all hens were 2-6 eggs fertilized. 4)Non Incision Injection to Abdominal-Cavity. I selected 15 hens which lay comparatively many eggs and injected with 10cm Tuberculin injector a cock’s semen, 0.1CC(number of sperm: abt, 100,000,000) to around the Ovum in the abdominal-Cavity. With this experiment five hens were fertilized and seven hens not. “Conculution” When I have examined these experiments as a Whole the Following come to the conculution: When I injected to the Follicle ti was not fertilized. but When injected to the Ovarian-Root a few fertilized eggs were laid after few days. as a result of this experiment I have known that the fertilization happens after ovulation. When I injected the semen to Oviduct the Ovum which is located at the center of Magnum region is not fertilized but the next Eggs were fertilized. As the result of this experiment I it seems to me that the fertilization is not done in the Oviduct. and five hens only were fertilized among 12 hens when I injected abdominal-Cavity through injection method. As a result of the above experiments I have got the conclution as followes: The fertilization region is around the infundebulum and the fertilizing time is during 15 Minutes Which is the time of coming into infundebulum, and is during 20 minutes which is time of coming down from infundebulum.

        P.M.S.에 의(依)한 가토(家兎)의 과잉배란(過剩排卵)에 있어서 Estrogen의 병용효과(倂用效果)

        가토(家兎)의 과배란유기(過排卵誘起)에 있어 P.M.S. 처리(處理)에 의(依)한 Estrogen의 병용효과(倂用效果)를 시험(試驗)하기 위(爲)해, P.M.S. 40I.U를 5일간(日間) 처리(處理)한것과, P.M.S.G. 40IU를 5일간(日間) 처리(處理)하고 최종일(最終日)에 Estrogen 0.1~0.5mg을 병용주사(倂用注射)한 것의 난소소견(卵巢所見)을 비교(比校)하며 배란수(排卵數)를 계측(計測)한 결과(結果), 다음과 같이 그 결과(結果)를 요약(要約)하였다. 1. P.M.S. 40IU를 5일간(日間) 피하주사(皮下注射)하여 24시간(時間) 및 48시간후(時間後)에 교미자극(交尾刺戟)을 준 대조구(對照區)의 평균(平均) 배란수(排卵數)는 21.3~24.1개(個), 과배란(過排卵) 양성률(陽性率) 50~63%에 비(比)하여 P.M.S.G 40, IU를 주사(注射)하고 최종일(最終日)에 Estrogen 0.1~0.5mg을 병용처리(倂用處理)한 구(區)의 평균(平均) 배란수(排卵數)는 32.5~37.8개과배란(個過排卵) 양성률(陽性率)은 87.5~100%로서 Estrogen을 병용처리(倂用處理)한 것이 배란수(排卵數)가 높아지는 경향(傾向)을 나타냈으며 미피열배란수(未破裂排卵數)는 대조구(對照區)가 많은 반면(反面), Estrogen 병용구(倂用區)는 적은 경향(傾向)을 나타냈다. 2. 대조구(對照區)와 Estrogen 병용구(倂用區)에 대(對)하여 난포(卵胞)를 1.0~1.4mm, 1.5~2.4mm, 2.5mm 이상(以上)의 3종(種)으로 분류(分類)한 후(後) 경시적(經時的) 관찰(觀察)에 의(依)한 발육정도(發育程度)를 비교(比較)한 결과(結果) 3종(種)에 대(對)한 배란수(排卵數)의 추이(推移)는 양구(兩區)가 서로 틀려서 총배란수면(總排卵數面)은 Estrogen 병용구(倂用區)가 우수(優秀)하였다 3. 이런 결과(結果)로 미루어 볼때 P.M.S. 주사(注射)에 대(對)한 Estrogen의 병용처리(倂用處理)는 외면적(外面的)으로는 난포발육(卵胞發育)에 대(對)하여 직접(直接) 영향(影響)을 주지 않으나 미피열난포(未破裂卵胞)를 줄여 배란수(排卵數)를 증가(增加)시키는 경향(傾向)을 나타냈다. This study was conducted to induce superovulation by P.M.S. injection and P.M.S. associate with Estrogen comparatively in the rabbit. The results obtained in this study are as follows. At 23-48 hours following mating, there were 21.3-24.1 ova (Ovulation rate was increased 50.5-63.6%) in average in the group of 40 I.U P.M.S. injected per day per herd for 5 days (total 200 I.U. of P.M.S.) and 32.5-37.8 ova (Ovulation rate was increased 87.5-100%) in the group of P.M.S. associats with 0.1mg at 5th day. She number of unovulated-follicle were fewer in P.M.S.-Estrogen group than PMS group. As the result that dia meter of ova were classified in 3 group as 1-1. 4mm, 1.5-2.4mm and 2.5mm and observed, the ova in P.M.S. Estrogen group were slightly larger in size than P.M.S. group. It was conclued that P.M.S. associate with Estrogen injection was not so much effect on growth of ova but effect on ovulation of ova. so the number of ovulated ova were increased than P.M.S. group.

        과배란처리(過排卵處理)에 대(對)한 Mouse의 배란반응(排卵反應)의 계통차(系統差)

        본(本) 연구(硏究)는 5계통(系統)(CF#1, C57BL/6, ICR, dd, CHN)의 mouse에 있어서 과배란처리(過排卵處理)에 대한 반응차(反應差)를 조사(調査)하기 위하여 수행(遂行)되었다. 과배란처리(過排卵處理)는 5IU의 PMS를 피하(皮下)에 주사(注射)한 다음 58시간(時間) 후(後)에 5IU의 HCG를 피하주사(皮下注射)하였으며, 배란수(排卵數)의 조사(調査)는 HCG 주사(注射) 24시간후(時間後)에 도살(屠殺)하여 난관내(卵管內)의 난자수(卵子數)를 현미경하(顯微鏡下)에서 계정(計定)하였다. 평균배란수(平均排卵數)는 CF#1, C57BL/6, ICR, dd 및 CHN계통(系統)에서 각각 21.3, 17.9, 20.7, 21.0 및 20.5개였다.         The Present study was examined the response of mice of 5 strains to superovulation treatment. Female mice of CF #1, C57BL/6, ICR, dd and CHN strains, aged 60 days were used. Superovulation was induced by the subcutaneous injection of 5 IU PMS and followed 48 hours later by the subcutaneous injection of 5 IU HCG. All females were killed 24 hours after HCG and then ova in the oviduct were counted under a microscope. The average number of ova ovulated in CF #1, C57BL/6, ICR, dd and CHN were 21.3, 17.9, 20.7, 21.0 and 20.5, respectively.

        조직배양액(組織培養液) 199배지(培地)에 의(依)한 가토난자(家兎卵子)의 저온(低溫)(5℃) 보존(保存)

        TC199 Medium를 Bas에 여러 양(量)의 가토혈청(家兎血淸)을 첨가(添加)하여 가토난자(家兎卵子)의 저온보존(低溫保存)를 시험(試驗)한 결과(結果) TC199단독(單獨)으로 $5^{\circ}C$에 48시간(時間) 보존(保存)한 16세포난자(細胞卵子)의 배양후(培養後)의 발생율(發生率) 31%에 대하여 TC199에 혈청(血淸)(20~80%)를 첨가(添加)했을때의 발생율(發生率) 80~100%라는 좋은 성적(成績)을 얻었다. 더욱이 비교혈청(比較血淸)만으로 생리적식염수(生理的食鹽水) 혈청(血淸)과 생리적(生理的) 식염수(食鹽水) 1:1의 Medium로 보존(保存)했을 때 이 발생율(發生率)은 100, 75, 및 83% 이였다. 이런 점(點)에서 TC199를 보존액(保存液)으로 사용(使用)했을 때에는 가토혈청(家兎血淸)의 첨가(添加)가 필요(必要)하다는 것이 판명(判明)되었다. Fertilized rabbit ova at 16-cell stage kept in TC 199, TC $199_+{^+}$ rabbitserum(4:1, 3:2, 2:3 and 1:4), rabbit serum, saline and serum+saline(1:1) were stored at $5^{\circ}C$ for 48 hours. They were than cultured at 37 C for 48 hours to determine their viability. The percentage of ova survived in each storage media was 31, 87, 100, 80, 100, 75 and 83%, respectively.

      • 過排卵處理 마우스에 있어서의 排卵數의 左右分布에 관한 硏究

        The distribution of number of ovulation between right and left ovary in superovulated mice were examined. 50 adult female mice of CF#1 strain were used in this study. Superovulation was induced by the subcutaneous injection of 5 IU PMS and followed 48 hours later by the subcutaneous injection of 5 IU HCG. Females a were killed at 24 hours after injection of HCG, and ova in the oviducts were counted under a microscope. The number of ova ovulated from the left and the right ovary were 603 and 594. The number of ova in both sides were found equal frequency by X^2 test method. Ovulation occurred more frequently in the left ovary than in the right in 24 females and with equal frequency in both ovaries in 3 females, while more frequently in the right ovary than in the left in 23 females. The possitive correlation was found between right and left ovary.

        계란의 수정시기및 수정부위에 관한 연구(제1보)

        本實驗을 綜合해서 要約해보면 濾胞에 直接注入한 것은 受精되지 안했고 卵巢莖部에 注入한 것은 注入後 數日만에 受精卵이 産卵되였으므로 排卵된 後에라야 受精되는 것이라고 보며 卵管內에 注入한 것은 注入當時 卵百分泌中間部에 있던 卵子가 受精되지 않고 그 以後에 낳은 알은 受精되였다. 그러한 것을 보면 卵管에서도 受精되지 않는다. 또한 注射法으로 腹腔內에 注入한 것은 12首中에서 5首만이 受精된 것을 보면 受精되는 場所는 漏斗狀部 附近이며 時期는 排卵되여 漏斗狀部로 드러오는 15分과 漏斗狀部로부터 내려오는 20分 동안이라고 본다.

        가토의 과배란유기에 있어서 Estrogen 병용효과에 관한 연구

        This experiment was conducted to induce the superovulation of the rabbits. Sixteen mature rabbits weighing 2 ㎏ on the average were fed preliminarily for a month. The treatments were done as below; 1. Estrogen (4000IU)+P.M.S. (250IU)×5 days+H.C.G.(125IU) and mating. 2. P.M.S. (50IU)×5 days+Estrogen(2000IU)+H.C.G. (250IU) and mating. 3. P.M.S. (50IU)×5 days+H.C.G. (250IU) and mating. The results obtained are as follows: 1. The group injected intramusculary 250 IU of P.M.S. showed no superovulation, and superovulation was occurred in the group injected subcutaneously 50 IU of P.M.S., the ovulation rate bring 4 to 5 times than the normal rabbits. 2. Estrogen treatment was not significant for the superovulation of the rabbits, but increased the mating capacity and enlarged the follicles and blood follicle.
