        문화적 재생을 위한 장소만들기 연구

        · Research topics: A study on cultural regeneration limits and solutions · Research background: Cultural regeneration is also biased toward the physical environment, revealing problems, and a strategic approach to solving the problem is required. · Difference from prior research: Until now, cultural regeneration only emphasizes the necessity of resource utilization and space creation, and overlooks the limitation of surface regeneration. · Research method: We will try to find a strategic approach to cultural regeneration by analyzing theoretical approaches to place making and cultural regeneration and specific field cases. · Research result: The cultural approach can also be a means to sustain the regeneration performance, and the usefulness of the place-making approach is suggested as a method for this. · Contribution points and expected effects: While revealing the usefulness of the approach to cultural regeneration, he suggested a place-making approach as a practical application method rather than the existing method that focused only on resources or space.         기니픽, 마우스 그리고 랫드에서 EPO (Erythropoietin)의 항원성에 관한 고찰

        The antigenicity of EPO (erythropoietin) was investigated in guinea pig, mice and rats. Antigenicity tests-active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA) of this materials were performed according to the established Regulation of Korean National Institute of Safety Research (1996, 4, 16). The results were followed: 1. After sensitizaion with EPO emulsified with complete Freund's adjuvant (CFA), guinea pigs didn't show any anaphylatic shock symptom in the ASA test 2. After sensitization with antisera of EPO sensitized mice, blue spots were observed on the hypodermis of back of rats in the PCA test, but diameter of each spot was smaller than 5 mm. From the results of this investigation, the antigenicity of EPO was negative under the present experimental condition.

        Mechanism of Humoral and Cellular Immune Modulation Provided by Porcine Sertoli Cells

        The understanding of main mechanisms that determine the ability of immune privilege related to Sertoli cells (SCs) will provide clues for promoting a local tolerogenic environment. In this study, we evaluated the property of humoral and cellular immune response modulation provided by porcine SCs. Porcine SCs were resistant to human antibody and complement-mediated formation of the membrane attack complex (38.41±2.77% vs. 55.02±5.44%, p=0.027) and cell lysis (42.95±1.75% vs. 87.99 ±2.25%, p<0.001) compared to immortalized aortic endothelial cells, suggesting that porcine SCs are able to escape cellular lysis associated with complement activation by producing one or more immunoprotective factors that may be capable of inhibiting membrane attack complex formation. On the other hand, porcine SCs and their culture supernatant suppressed the up-regulation of CD40 expression (p<0.05) on DCs in the presence of LPS stimulation. These novel findings, as we know, suggest that immune modulatory effects of porcine SCs in the presence of other antigen can be obtained from the first step of antigen presentation. These might open optimistic perspectives for the use of porcine SCs in tolerance induction eliminating the need for chronic immunosuppressive drugs.

        Role of Complement Regulatory Proteins in the Survival of Murine Allo-transplanted Sertoli Cells

        Sertoli cells (SC) are known to contain immunoprotective properties, which allow them to survive as allografts without the use of immunosuppressive drugs. Experiments were designed to determine which factors are related to prolonged survival of allogeneic SC. Balb/c derived Sertoli (TM4) and colon cancer (CT-26) cell lines were implanted beneath the kidney capsule of non-immunosuppressed C57BL/6 mice and compared their survival as allografts. Compared to TM4 graft, which survived more than 7 days after transplantation, CT-26 showed massive infiltration of polymorphonuclear cells, necrosis and enlargement of draining lymph nodes. Cultured cell lines showed no differences in their expression patterns of FasL, TGF 1, clusterin and two complement regulatory proteins (CRP, i.e., membrane cofactor protein, MCP; decay accelerating factor, DAF), but protectin (CD59), another member of CRP was expressed only on TM4. These results suggest that CD59 and unknown factors may contribute to the prolonged survival of SC in non-immunoprivileged sites.

        Effect of Hyperkalemia and Hemolysis Caused by Hyperacute Rejection on Cardiac Function in Pig to Human Ex Vivo Xenogeneic Cardiac Perfusion Model

        Background and Objectives: Hyperacute rejection (HAR) is a major obstacle to successful xenotransplantation of vascularized organs. This study was conducted to observe the effect of hemolysis of perfused human whole blood on pig heart function, and determine the major risk factors for preservation of xenoperfused cardiac function using ex-vivo pig to human xenogeneic cardiac perfusion model. Materials and Methods: Harvested pig hearts were perfused with normal human whole blood (group 1), two different types of pre-treated human whole blood (group 2: immunoglobulins were depleted by plasmapheresis, group 3: pre-treated with plasmapheresis, GAS914, cobra venom factor (CVF) and steroid), and normal porcine wh-ole blood as control (group 4) for 3 hours. Results: Duration of heart beat was significantly prolonged in group 2 and group 3. Histological examination showed widespread HAR features but was gradually delayed in groups 2 and 3 compared to group 1. The absolute levels of serum creatine kinase-MB and Troponin I increased gradually, and was lower in group 3. Serum hemo-globin levels were rapidly increased in groups 3 and 4, compared to group 1. Extracellular potassium level increased sharply from the beginning of blood perfusion in groups 1, 2 and 3, compared to group 4. Conclusion: Pretreatment of human whole blood, including immunoglobulin depletion, CVF and steroid reduced and delayed the destruction of pig myocardium by HAR. However, the increased extracellular potassium levels in groups 1, 2 and 3 reflected that these treatments could not prohibit myocardial injury by HAR.         신규 개인제독키트, SDK시제품(가칭)의 면역독성평가에 관한 고찰

        To evaluate immunotoxicity of skin decontamination kit(SDK) newly-developed in Agency for Defense Development(ADD), delayed contact hypersensitivity (maximization) test and passive cutaneous anaphylaxis(PCA) test of SDK were performed and the results were compared with those of M 291. In maximization test, sensitization reaction was induced by id injection (2.5 mg / 0.1 $\textrm{m}{\ell}$/ guinea pig or 2.5 mg+CFA/0.1 $\textrm{m}{\ell}$/guinea pig) and topical application (2.5 mg/$\textrm{m}{\ell}$/guinea pig) with SDK or M291 at an interval of 1 week, and 2 weeks later, challenged by topical application with 25 mg/$\textrm{m}{\ell}$/guinea pig. SDK and M291 did not induce any reactions, showing 0 point of sensitization score and 0% of sensitization rate. In conclusion, it is suggested that SDK and M291 do not induce delayed contact hypersensitivity. In PCA test, rats were administered id with mouse anti-SDK serum and challenged iv with a mixture of antigen SDK and Evan's blue. SDK did not induce blue spots at the injection sites of both high (2.5 mg/mouse) and low (1.25 mg/mouse) dose-induced antisera. In contrast, BSA, positive control produced spots larger than 5 mm in diameter at the injection sites of BSA-induced antiserum up to $2^2$ ~ $2^4$dilution. In conclusion, it is suggested that SDK do not induce IgE production and is not a PCA-reaction inducer.

        Kinetics of IFN-γ and IL-17 Production by CD4 and CD8 T Cells during Acute Graft-versus-Host Disease

        Graft-versus-host disease (GVHD) is a fatal complication that occurs after allogeneic hematopoietic stem cell transplantation. To understand the dynamics of CD4 and CD8 T cell production of IFN-γ and IL-17 during GVHD progression, we established a GVHD model by transplanting T cell-depleted bone marrow (TCD-BM) and purified T cells from B6 mice into irradiated BALB.B, creating an MHC-matched but minor histocompatibility (H) antigen-mismatched transplantation (B6 → BALB.B GVHD). Transplantation-induced GVHD was confirmed by the presence of the appropriate compositional changes in the T cell compartments and innate immune cells in the blood and the systemic secretion of inflammatory cytokines. Using this B6 → BALB.B GVHD model, we showed that the production of IFN-γ and IL-17 by CD4 T cells preceded that by CD8 T cells in the spleen, mesenteric lymph node, liver, and lung in the BALB.B GVHD host, and Th1 differentiation predated Th17 differentiation in all organs during GVHD progression. Such changes in cytokine production were based on changes in cytokine gene expression by the T cells at different time points during GVHD development. These results demonstrate that both IFN-γ and IL-17 are produced by CD4 and CD8 T cells but with different kinetics during GVHD progression.
