        Lactobacillus casei KCTC 3109에 의한 감국 추출물의 항산화능의 변화

        유산균에 의한 감국 추출물(CIL)의 항산화능의 변화를 알아보기 위해 64%, 80% ethanol 로 추출한 농도별 CIL를 발효시켜 total phenolic contents (TPC), flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power (RP), linoleic acid 자동산화 저해활성의 변화를 확인해 보았다. TPC의 경우 3109, 3237에서 증가하였으며, flavonoid에서는 모든 균주에서 감소를 나타냈다. DPPH 측정에서는 64% CIL 발효 후 3074, 3109에서 증가하였으며, 80% CIL 발효에서는 모든 균주가 증가를 나타냈다. RP측정에서는 64%, 80% CIL에서 모든 균주에서 증가를 보였으며, linoleic acid 자동산화 저해활성에서는 모든 균주에서 감소를 나타냈다. 특히 3109 항산화능을 평가하는 DPPH와 RP 측정에서 다른 균주에 비해 전반적으로 높게 측정이 되었다. 결과적으로 4종의 유산균 중에서 3109가 발효과정을 통해서 CIL의 항산화 효능을 효과적으로 증대하는 것을 확인할 수 있었다. The antioxidant activity of Chrysanthemum indicum L. extract (CIL) was investigated by fermenting lactic acid bacteria with the CIL from 64% and 80% ethanol extraction and measuring the total phenolic contents (TPC), flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power (RP), and linoleic acid auto-oxidation inhibitory activity. CIL was confirmed to inhibit bacterial auto-oxidation. TPC was increased in strains 3109 and 3237, while flavonoid decreased in all strains. DPPH was increased in strains 3074 and 3109 fermented with 64% CIL and all the strains with 80% CIL. RP was increased and linoleic acid auto-oxidation inhibitory activity decreased in all the strains fermented with 64% or 80% CIL. Among the 4 strains, strain 3109 had the highest DPPH and RP; thus, it was most effective in increasing CIL’s antioxidant efficacy through the fermentation process.

        Objective : This study aims to explain the effect of Buja-tang on Postherpetic Neuralgia with Chronic Fatigue Syndrome which a Soeum-byeong patient has. Methods : This is a case report on a female patient who has been suffering from Postherpetic Neuralgia with Chronic Fatigue Syndrome. According to Diagnostic system based on Shanghanlun provisions, Buja-tang was provided for one month. Results : After a series of Buja-tang treatments, the patient's symptoms and result improved. Conclusions : This case study show an effectiveness of using Buja-tang according to Diagnostic system based on Shanghanlun provisions on Postherpetic Neuralgia with Chronic Fatigue Syndrome.

        활성산소 (ROS) 조절에 의한 만병초의 c-Jun/MMP-1 저해효과

        This paper reports on the results obtained the 80% ethanol extract and fractions from Rhododendron brachycarpum (R. brachycarpum) had anti-oxidant induced collagen-degration inhibit effect. We tested R. brachycarpum extract and fractions for DPPH radical scavenging activity and reducing power, ROS induced NRF2 inhibit activity, HaCaT cell viability and collagen degration inhibit activity. As a result, ethyl acetate fraction showed DPPH radical scavenging activity (IC50=17.7±0 μg/mL, p<0.001), reducing power (OD700=2.58, p<0.001), ROS-NRF2 inhibit activity (16987±136 index, p<0.05) and R. brachycarpum extract showed reducing power (OD700=2.29, p<0.01), ROS-NRF2 inhibit activity (21183±1611 index, p<0.01) were the high anti-oxidant capacity. The cytotoxicity of HaCaT treated the R. brachycarpum extract and fractions was low activiy except for n-hexan fraction (78.6±1.5%) at 10 μg/well concentration. Inhibition of collagen degradation was confirmed by the decrease in the expression of MMP-1 at 1 μg/well concentration in extract and ethyl acetate fraction treated cells (p<0.001), however, BHA treatment cell did not inhibit effect of MMP-1. These results indicate that the antioxidants present in the R. brachycarpum extract and fractions can reduce the cellular ROS and control the expression for p-c-Jun and MMP-1. The possibility of the R. brachycarpum may be natural cosmetics material of skin elasticity and wrinkle improvement. 본 연구는 만병초의 80% 에탄올 추출물과 분획물을 이용하 여 화장품 소재로의 가능성을 예측하기 위해 실시하였다. 결 과는 DPPH 라디칼 소거능 ethyl acetate 분획물 (IC50=17.7±0 μg/mL), 환원력 측정 ethyl acetate 분획물 (OD700=2.58)과 만병 초 추출물 (OD700=2.29)에서 높은 항산화를 보였다 (p<0.01). NRF2 활성 감소는 ethyl acetate 분획물 (16987±136 index)과 만 병초 추출물 (21183±1611 index)에서 세포 내 활성산소 감소를보였다 (p<0.05). 이 같은 결과는 만병초 추출물 및 분획물에 존재하는 quercetin, quercitrin, ursolic acid 및 triterpenoids에 의 한 항산화 작용으로 사료된다. 또한, HaCaT 세포의 세포독성이 없었으며, 세포 내 H2O2에 의해 유도된 활성산소의 감소가 c-Jun의 인산화를 억제되었고, 하위 신호인 MMP-1의 발현을 감소시켰다. 이는 만병초에 존재하는 quercetin의 영향으로 보 여진다. 만병초 추출물 및 ethyl acetate 분획물에 함유된 항산 화 물질이 세포 내 활성산소를 억제시키고, c-Jun의 인산화 및 MMP-1의 발현을 감소를 통해 콜라겐 분해 억제 작용이 우수 하여 산화적 스트레스에 의해 발생된 주름개선 화장품 소재로 활용할 수 있음을 시사한다.

        락토바실러스 람노서스(Lactobacillus rhamnosus) KCTC 5033의 Probiotics와 Paprobiotics 섭취에 의한 얼굴미용효과

        Lactic acid bacteria are often taken in the form of yogurt-which is becoming a highly stable food type often consumed daily-or medicine. Lactic acid bacteria have a significant industrial value because of the various reports of health improvement effects that they may convey (eg, anti-microbial, improved immunity). This study examined changes in facial skin when middle-aged women (ie, 40~50s) took probiotic and para-probiotic medicines derived from Lactobacillus rhamnosus KCTC 5033. The research subjects were 30 middle-age women who were randomly assigned (10 per group) to consume either: i) probiotics of L. rhamnosus KCTC 5033, ii) bacteria-killing paraprobiotics, or, iii) a control. Each group consumed 1 capsule (1×1010 CFU or more) of assigned treatment on an empty stomach every morning for 12 weeks, and changes in their facial and neck skin before and after consumption were observed. Their skin change, melanin index, erythema index, moisture content, TEWL, skin roughness, and skin surface temperature were measured to identify any potential differences. A survey was also used to investigate respondent’s satisfaction with their skin before and after lactic acid bacteria intake on a 5-point Likert scale. Before and after results demonstrated that melanin index significantly increased in only the control group while moisture content increased significantly in the probiotics and paraprobiotics groups. Also in the probiotics and parabiotics groups, moisture content increased, while skin roughness decreased. Also in the probiotics and parabiotics groups: i) moisture content increased, ii) skin roughness decreased, and iii) no changes (before to after) in erythema index, TEWL or skin surface temperature were observed. The results of probiotics and paraprobiotics on neck skin indicated that the control group experienced a significant increase in melanin index, skin roughness, and TEWL; moisture content increased significantly in the probiotics and paraprobiotics groups, and skin surface temperature increased significantly in only the probiotics group.

        락토바실러스 람노서스(Lactobacillus rhamnosus) KCTC 5033제제 섭취가 40-50대 중년여성의 목피부에 미치는 영향

        This study investigated changes in neck skin appearance middle-aged women in their 40s and 50s after ingesting probiotic or paraprobiotic Lactobacillus rhamnosus KCTC 5033. Women in each group consumed one capsule (1 × 10<sup>10</sup>CFU) for 3 months and underwent measurements of changes in neck skin appearance. Melanin content was significantly increased only in the placebo control group, whereas the moisture content was not altered in either the probiotic or paraprobiotic group. in the probiotic group. The erythema index was not changed, but oil content and roughness tended to increased oil content and decreased roughness in the probiotic and paraprobiotic groups compared with the findings prior to the experiment. Moisture was increased in both the probiotic and paraprobiotic groups, whereas moisture evaporation was significantly increased only in the control group. The neck skin surface temperature was only changed in the probiotic group. Therefore, the probiotic and paraprobiotic treatments inhibited melanin production by altering the intestinal condition, in addition to increasing moisture by increasing the oil content and decreasing skin roughness. In fact, subjects in these groups expressed greater satisfaction after the experiment, especially those in the paraprobiotic group. Thus, L. rhamnosushas been identified asa potential therapeutic for improving the appearance of skin for the first time.

        락토바실러스 루테리 섭취가 여성의 목 피부 보습과 주름개선에 미치는 영향

        This study aimed to investigate the beneficial effects of Lactobacillus reuteri KCTC 3594 probiotics and paraprobiotics on the moisturization and wrinkle improvement of neck skin in women in their 30s–40s. For this investigation, a total of 30 participants were divided into the following three groups (10 participants each): control group (Cont), L. reuteri probiotic group (Pro), and L. reuteri paraprobiotic group (Para). L. reuteri was administered to each group (1×1010 CFU / capsule) on an empty stomach in the morning once a day for 12 weeks. After the 12-week intake of L. reuteri, no significant change in skin moisture content was observed in the Cont group, while a statistically significant increase was detected in the Pro and Para groups (p<0.001). Regarding wrinkle improvement, no statistically significant change was noted in the Cont group, while the Pro and Para groups revealed statistically significant declines (p<0.01 and p< 0.05, respectively). The obtained results revealed the following: moisture level and wrinkles improved in the Pro and Para groups. In the Pro group in particular, positive effects on wrinkles were identified . This study confirmed that the oral intake of L. reuteri is effective at slowing down the aging of neck skin and has a positive effect on the skin. It appears that such intake produced intrinsic changes in the skin by enhancing immunity in the intestines and skin. The results of this study confirm the usefulness of L. reuteri probiotics and paraprobiotics as potential cosmetics raw materials.

        다양한 농도의 에탄올로 추출한 불등풀가사리 추출물의 항산화, 항염 특성에 관한 연구

        Purpose: This study aimed to explore the differences in the antioxidant activity and anti-inflammatory effects of Gloiopeltis furcata based on its extraction conditions. Methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays were performed to determine the antioxidant activity of Gloiopeltis furcata extracts obtained using different concentrations of ethanol. Concomitantly, the concentrations of polyphenols and flavonoids, which are antioxidants present in Gloiopeltis furcata extract, were measured. Additionally, in cell-based experiments, the cytotoxicity and anti-inflammatory activity of a water extract, which exhibited the most potent antioxidative activity among all extracts, was measured. Results: In the DPPH assay, the Gloiopeltis furcata extract exhibited an antioxidant activity of 3.576 mg of ascorbic acid/g, whereas in the ABTS assay, the antioxidant activity was 4.892 mg of ascorbic acid/g. The polyphenol concentration was found to be 35.52±2.32 mg/g, whereas the flavonoid concentration was 0.88±0.34 mg/g. Furthermore, the extract had a cytotoxicity of ≤20% at all concentrations used in the experiment, thus indicating the absence of cytotoxicity and confirming that the extract was effective in reducing inflammation proportionally to its concentration. Moreover, the extract exhibited an anti-inflammatory activity of 47.66%±1.53% at a concentration of 100 μg/mL. Conclusion: Gloiopeltis furcata exhibited the most potent antioxidant activity in its water extract, and it was confirmed that the extract of Gloiopeltis furcata that was obtained through water had low cytotoxicity, an anti-inflammatory effect, and a commercial value. 목적: 본 연구에서는 불등풀가사리 추출 조건에 따른 항산화능 차이 및 불등풀가사리 추출물의 항염효과에 대해 연구하고자 하였다. 방법: 다양한 농도의 에탄올로 추출한 불등풀가사리 추출물의 항산화능 측정을 위해서 DPPH assay와 ABTS assay를 실시하였다. 동시에 불등풀가사리 추출물에 대해 항산화 물질인 폴리페놀과 플라보노이드의 농도를 측정하였다. 세포실험 역시 가장 항산화력이 우수했던 수성 추출물의 세포 독성과 항염활성을 측정하였다. 결과: DPPH 실험에서는 3.576 mg ascorbic acid/g의 항산화능을 나타내었으며, ABTS 실험에서는 4.892 mg ascorbic acid/g의 항산화능을 나타내었으며, 폴리페놀 농도는 35.52±2.32 mg/g으로 나타났다. 플라보노이드 농도는 0.88±0.34 mg/g으로 나타났다. 세포독성의 경우 실험에 사용된 모든 농도에서 20% 이하의 세포 독성을 보여 세포독성이 없음을 확인하였으며, 농도 비례적으로 염증을 감소시키는 효과를 확인함과 동시에 100 μg/mL 농도에서 47.66±1.53%의 염증 억제능을 보였다. 결론: 불등풀가사리의 경우 수성 추출에서 가장 높은 항산화능을 나타내었으며, 불등풀가사리의 수성추출물의 경우 낮은 세포 독성과 항염효과가 있음을 확인하였으며, 상업적 이용가치가 있음을 확인하였다

        비피도박테리움 아니말리스 락티스(Bifidobacterium animalis subsp. lactis KTCT 3594)의 섭취가 중년여성의 피부미용에 미치는 영향

        The purpose of this study is to investigate the skin changes of the face after ingesting Bifidobacterium animalis subsp. lactis probiotics and paraprobiotics in 24 person during 12 weeks, who were 40-60 middle-aged women. There were two experimental groups; first, a Cont-group was served an excipient. Second, the experimental group was divided into a ‘Pro-group’ with the B. animalis probiotics and a ‘Para-group’ with the B. animalis paraprobiotics. Then, these groups were served a 1 × 1010 CFU capsule containing the appointed substance per day each person. As a result, the rate of sebum was highly increased (**p<.01, *p<.05) 459% in the ‘Pro-group’ and 268% in the ‘Para-group’ as compared with the Cont-group. For the rate of moisture, it also has a significant increase (**p<.01) 602% and 973% each group. In case of texture, the Cont-group showed a significant increase (***p<.001), but the ‘Pro-group’ and ‘Para-group’ showed a significant decrease (**p<.01) 141% and 141% each. For the wrinkle, the control group showed a significant increase (***p<.001). In contrast, the two experimental groups had a significant decrease (**p<.01) 140% and 136% each. Therefore, the intake of B. animalis probiotics and paraprobiotics in 40-60 mid-aged women had a positive effect on the skin improvement and prevention of aging skin, so that it can be used as a great material of the inner beauty product.

        Objective : This study aims to explain the effect of Injinho-tang on Eczema which a Yangmyung-byeong patient has. Methods : This is a case report on a female patient who has been suffering from Eczema. According to Diagnostic system based on Shanghanlun provisions, Injinho-tang was provided for six months. Results : After a series of Injinho-tang treatments, the patient's symptoms and result improved. Conclusions : This case study showed an effectiveness of using Injinho-tang according to Diagnostic system based on Shanghanlun provisions on Eczema.
