        Polyhydroxyalkanoate Chip for the Specific Immobilization of Recombinant Proteins and Its Applications in Immunodiagnostics

        In this study, a novel strategy was developed for the highly selective immobilization of proteins, using the polyhydroxyalkanoate (PHA) depolymerase substrate binding domain (SBD) as an active binding domain. In order to determine the appropriacy of this method for immunodiagnostic assays, the single-chain antibody (ScFv) against the hepatitis B virus (HBV) preS2 surface protein and the severe acute respiratory syndrome coronavirus (SARS-CoV) envelope protein (SCVe) were fused to the SBD, then directly immobilized on PHA-coated slides via microspotting. The fluorescence-labeled HBV antigen and the antibody against SCVe were then utilized to examine specific interactions on the PHA-coated surfaces. Fluorescence signals were detected only at the spotted positions, thereby indicating a high degree of affinity and selectivity for their corresponding antigens/antibodies. Furthermore, we detected small amounts of ScFv-SBD (2.7 ng/mL) and SCVe-SBD fusion proteins (0.6 ng/mL). Therefore, this mi-croarray platform technology, using PHA and SBD, appears generally appropriate for immunodiagnosis, with no special requirements with regard to synthetic or chemical modification of the biomolecules or the solid surface.

      • Performance of Dual Polarized MIMO System using Six-Port Receiver for Cognitive Radio

        Cognitive radio is a paradigm for wireless communication in which either network of wireless node itself changes particular transmission or reception parameters to execute its tasks efficiently without interfering with the licensed users. This paper represents a performance of the cognitive radio technology on dual polarized MIMO system using six-port receiver. Six-port technology is well known direct conversion receiver. In this paper, a six-port phase discriminator based polarization signal separation is shown. That is, the SER(Symbol Error Rate) performance is improved using polarization separator and simple receiver architecture is proposed applying six-port receiver. The six-port technology has priority to adapt changeable frequency system and variable environments for cognitive radio. In general, dual polarized MIMO system has good capacity and quality using polarization separator [1].

        토픽모델링을 활용한 교통경찰 민원 분석

        This study aims to investigate the security demand about the traffic policing by analyzing civil complaints. Latent Dirichlet Allocation(LDA) was applied to extract key topics for 2,062 civil complaints data related to traffic policing from e-People. And additional analysis was made of reports of violations, which accounted for a high proportion. In this process, the consistency and convergence of keywords and representative documents were considered together. As a result of the analysis, complaints related to traffic police could be classified into 41 topics, including traffic safety facilities, passing through intersections(signals), provisional impoundment of vehicle plate, and personal mobility. It is necessary to strengthen crackdowns on violations at intersections and violations of motorcycles and take preemptive measures for the installation and operation of unmanned traffic control equipments, crosswalks, and traffic lights. In addition, it is necessary to publicize the recently amended laws a implemented policies, e-fine, procedure after crackdown. 본 연구는 민원데이터를 분석함으로써 교통경찰에 대한 국민의 치안 수요를 탐색하고자 하였다. 이를 위해 교통경찰 관련 국민신문고 민원데이터 2,062건을 대상으로, 토픽모델링 방법 중 하나인 잠재 디리클레 할당(Latent Dirichlet Allocation)을 통해 주요 토픽을 추출하고 높은 비중을 차지한 위반신고에 대해 추가분석을 시도하였다. 이 과정에서 키워드와 대표문서의 일관성과 합치성을 함께 고려하였다. 분석 결과 교통경찰 관련 민원은 시설개선, 신호에 따른 교차로통행방법, 번호판 영치, 개인형 이동장치 등 41개의 토픽으로 분류할 수 있었다. 교차로내 위반과 이륜자동차의 위반에 대한 단속을 강화하고 무인교통단속장비, 횡단보도, 신호등의 설치 및 운영에 대한 선제적인 조치, 최근 개정된 법령과 시행된 정책, 경찰교통민원 사이트, 단속 사후 절차에 대한 더욱 활발한 홍보가 필요한 것으로 판단된다.

        단안상전장애에 동반된 눈꺼풀처짐에서 눈꺼풀올림근을 이용한 이마근걸이술 1예

        Purpose: For the correction of ptosis in monocular elevation deficiency (MED) patients, levator resection or frontalis sling operation with various materials has been used. The authors present a case of a monocular elevation deficiency patient who was treated for ptosis using the levator muscle as a frontalis sling. Case summary: A six-year-old male patient with left monocular elevation deficiency underwent a frontalis sling operation using a levator muscle. The patient had received left inferior rectus recession, left medial rectus resection and left lateral rectus recession with transposition. An upper eyelid skin crease incision was made, and the anterior surface of the levator muscle was exposed as far as the Whitnall ligament. A levator muscle flap was then formed from aponeurosis by incising the full width of the levator horizontally, just below the Whitnall ligament and anchored to the frontalis muscle. The surgical result was satisfactory as the preoperative MRD1 was 0.5 mm and the postoperative MRD1 was 2.5 mm. There were no complications such as keratitis or wound problems. Conclusions: The levator muscle flap can be an effective frontalis suspension material to correct ptosis in an MED patient with poor levator function. J Korean Ophthalmol Soc 2010;51(2):282-285

        두개골 봉합부의 초기 형태발생과정에서 Msx 유전자들의 발현양상

        두개골의 성장은 뇌의 발달과 밀접하게 연관되어 있으며, 두개봉합부 주위 여러 조직들사이의 조화로운 상호작용을 필요로 한다. 두개골의 조기융합으로 알려진 craniosynostosis는 이러한 상호작용의 불균형으로 인해 야기될 수 있으며, craniosynostosis의 원인 유전자의 하나로 Msx2가 알려져 있다. Msx1, Msx2, Msx3로 구성되어 있는 Msx 유전자는 homeobox를 포함하고 있는 전사조절인자로, 초파리의 muscle segment homeobox 유전자의 homologue이다. Msx1과 Msx2 유전자들은 척추동물의 발생과정에서 조직 상호작용하는 여러 부위에 서로 겹치는 양상으로 발현되며 기관형성동안 상피-간엽조직 상호작용과 연관되어 나타나고, BMP와 FGF signaling의 표적 유전자들이다. Mouse 두개골의 시상봉합부와 관상봉합부의 초기 형태발생과정에서의 Msx 유전자들의 기능을 알아보기위해, 태생 15일에서 태생 18일까지의 mouse의 두개골을 이용하여 in vivo 실험을, 태생 15.5일 mouse 두개골을 이용하여 in vitro 실험을 시행하였다. 시상두개봉합부에서는, Msx1은 봉합부의 간엽조직과 dura mater에 발현하였으며 Msx2는 두개봉합부와 dura mater에 강하게 발현되었다. 관상두개봉합부에서는 Msx1, Msx2 유전자 모두 강하게 발현되었으며, 골막에서도 관찰할 수 있었고, Msx1이 Msx2보다 좀더 광범위하게 발현되었다. In vitro 실험결과, BMP2 bead 주위로 Msx1 mRNA와 Msx2 mRNA 모두 발현되었으나, FGF2 bead 주위로는 Msx1만 발현되고 Msx2는 관찰되지 않았다. 이 결과들을 종합해 볼 때, Msx1, Msx2 유전자들은 태생기 두개봉합부의 형태발생 및 유지를 조절하는데 중요하며, Msx 유전자들은 거의 유사한 발현양상을 나타내고 있으나 이들 유전자들을 조절하는 상위 siganling에 의해 서로 보완적으로 혹은 독립적으로 기능을 나타내고 있음을 시사해 주고 있다. The development of calvarial bones is tighly co-ordinated with the growth of the brain and needs of harmonious interactions between different tissues within the calvarial sutures. Premature fusion of cranial sutures, known as craniosynostosis, presumably involves disturbance of these interactions. Mutations in the homeobox-containg gene Msx2 cause human craniosynostosis syndrome. Msx genes, which are consist of Msx1, Msx2 and Msx3, are homeobox-containg transcripton factors, and were originally identified as homologue of Drosophila msh(muscle segment homeobox) gene. Msx1 and Msx2 genes, expressed mostly in overlapping patterns at multiple site of tissue interactions during vertebrate development, are associated with epithelial-mesenchymal interactions during organogenesis, targets of BMP and FGF signaling. To elucidate the function of Msx genes in the early morphogenesis of mouse cranial suture, we analyzed the expression of them by in situ hybridization during embryonic(E15-E18) stage, and did vivo experiments in E15.5 mouse using rhBMP-2, rhFGF-2 protein soaked bead. In the sagittal suture, Msx1 was expressed in the mesenchyme of suture and the dura mater, Msx2 was intensely expressed in the sutural mesenchyme and the dura mater. In the coronal suture both of Msx genes were expressed intensely in the sutural mesenchyme and expressed in the periosteum also. Msx1 had a broader expression pattern than Msx2. BMP2 beads induced expression of both Msx1 and Msx2, FGF2 beads induced expression of Msx1, but not Msx2. Taken together, these data suggest that Msx1 and Msx2 genes have important role in regulating the morphogenesis and maintenance of embryonic cranial suture. Both of Msx genes are expressed similarly but because of their upstream signaling, they function dependently or cooperatively according to change of signaling molecule.
