사랑의 유전자 library로 부터 serine tRNA 유전자를 갖는 서로 다른 5개의 $\lambda$-phage 클론들을 분리하였다. 그 중 한개의 클론 ($\lambda$ St 1B)을 선택하여 유전자의 염기배열을 분석하였다. 이 클론은 그 insert내에 1.1 kb Eco RI 절편을 갖고 있었으며, 이 절편에 tRNA 유전자가 포함되어 있음을 southern blot hybridization과 dot blot hybridization에 의하여 알았다. 이 절편의 염기배열을 결정한 결과, 여기에 포함되어 있는 serine tRNA 유전자는 82개의 nucleotide들로 구성되어 있었고, anticodon 서열은 5'-AGA-3' 이었으며, intervening sequence는 갖고 있지 않았음을 알게되었다. 또한 유전자 내에는 promoter로 작용하는 두 군데의 보존된 염기배열들을 갖고 있었으며, 3' 인접부위에서는 RNA polymerase III에 의한 전사의 종결부위 (TTTTT)가 발견되였다. 한편, 이 유전자는 쥐의 간세포로 부터 분리한 serine $tRNA_{IGA}$와 그 염기배열이 같았고, 이미 보고된 한 개의 사람 serine $tRNA_{UGA}$유전자와는 anticodon의 5' 말단염기만이 달랐다. 이 결과는 포유동물의 serine tRNA 유전자들의 염기배열이 진화적으로 잘 보전되어 있음을 암시한다. Five different $\lambda$-phage clones which carry human serine tRNA genes were isolated from a human gene library. One of the clones ($\lambda$ St 1B) was selected for sequence analysis. Among the Eco RI digests of the phage DNA, a 1.1 kb fragment was shown to carry a tRNA gene by southern blot and dot blot hybridizations. Nucleotide sequence analysis of the fragment revealed the presence of a 82 bp serine $tRNA_{AGA}$ gene. The tRNA gene does not have intervening sequences nor does it encode the 3'-terminal CCA sequence found in mature tRNAs. As in other eucaryotic tRNA genes, the structural gene contains a characteristic internal split promoter sequence, and he 3'-flanking region as a typical RNA polymerase III termination site of five consecutive T residues. The sequence of this gene is homologous to that of a rat liver serine $tRNA_{IGA}$. In addition, it differs only in one position at 5' terminal nucleotide of anticodon from a previously sequenced human serine tRNA gene. This suggests that mammalian serine tRNA genes are highly conserved.

Kinetics and Reaction Mechanism for Alkaline Hydrolysis of Y-Substituted-Phenyl Diphenylphosphinates

The second-order rate constants (kOH−) for the reactions of Y-substituted-phenyl diphenylphosphinates (4a-4i) with OH– in H2O at 25.0 ± 0.1 oC have been measured spectrophotometrically. Comparison of kOH− with kEtO− (the second-order rate constants for the corresponding reactions with EtO– in ethanol) has revealed that EtO– is less reactive than OH– although the former is ca. 3.4 pKa units more basic than the latter, indicating that the reactivity of these nucleophiles is not governed by their basicity alone. The Brønsted-type plot for the reactions of 4a-4i with OH– is linear with βlg = –0.36. The Hammett plot correlated with σ– constants results in a slightly better correlation than that correlated with σo constants but exhibits many scattered points. In contrast, the Yukawa-Tsuno plot for the same reactions exhibits an excellent linear correlation with ρ = 0.95 and r = 0.55. The r value of 0.55 implies that a negative charge develops partially on the O atom of the leaving group. Thus, the reactions of 4a-4i with OH– have been concluded to proceed through a concerted mechanism.

Second-order rate constants (kHOO−) for the nucleophilic substitution reactions of Y-substituted-phenyl diphenylphosphinates (4a-4i) with HOO− in H2O have been measured spectrophotometrically. The α-nucleophile HOO− is 10-70 times more reactive than the reference nucleophile OH− although the former is ca. 4 pKa units less basic than the latter, indicating the α-effect is operative. The Brønsted-type plot for the reactions of 4a-4i with HOO– is linear with βlg = –0.51, a typical βlg value for reactions which were reported to proceed through a concerted mechanism. The Yukawa-Tsuno plot is also linear with ρ = 1.40 and r = 0.47, indicating that a negative charge develops partially on the O atom of the leaving group, which can be delocalized to the substituent Y through resonance interactions. Thus, the reactions have been proposed to proceed through a concerted mechanism. The magnitude of the α-effect (i.e., the kHOO−/kHO− ratio) decreases linearly as the leaving-group basicity increases. It has been concluded that solvation effect is not solely responsible for the α- effect found in this study but the transition-state stabilization through an intramolecular H-bonding interaction is also responsible for the α-effect.

쥐의 단일클론항체는 기초연구에 뿐만 아니라 질병의 진단시약으로서도 광범위하게 유용되어 왔다. 반면, 쥐의 단일클론항체를 인체에 임상적으로 사용하는 것은 인체에 면역반응을 유발시킨다는 근본적인 문제점 때문에 어려운 일이라 인식되어 왔다. 그러나, 최근 2-3년간 유전자 기술과 단백질공학적인 방법에 의하여 쥐의 단일클론항체를 인간화시키거나 항체절편의 생산을 시도하게 되어, 단일클론항체의 임상적인 이용에 대한 가능성이 긍정적으로 진단되고 있다. 본 난에서는 항체의 구조와 기능을 간략히 살펴보고, 최근 항체의 단백질공학에 대한 경향과 방법 및 engineered 항체의 특성들에 관하여 논의하고자 한다.
