        한국산 팽나무버섯에서 선발된 두 계통에 대한 자실체배양상의 (子實體培養上) 특성

        The cultural characteristics of fruit bodies formation in the two selected strains(brown and yellow strain) of winter mushroom collected in Korea were investigated. The effects of various environmental conditions and physicochemical pretreatments were evaluated by the character of fruit bodies. There was no differences between two strains in non-selectivity of media, optimum temperature(11℃∼15℃), relative humidity(90%), and illumination(60 lux). In the application of alternated temperature (the best condition : 5℃∼10℃ and 5℃∼15℃), ultraviolet ray(30∼50 seconds), electricity(5 seconds), thiamine hydrochloride(0.05∼0.1%), urea, and IAA as a pretreatments, there was no differences between two strains. The brown strain had larger pileus and Longer stipes than these of the yellow strain, while the yellow strain yielded more weight and number of the fruit body formation than the brown strain.

        흉고직경별 약제 수간주입구멍의 크기에 따른 주입구멍의 치유도 - 포스팜 수간주입구멍에 대한 반응 -

        Responses of Pospam-injected chestnut and pine trees to drill wound were examined. Pospam was injected into the stem by just filling up the drill wound in September 10, 1993. The diameter of the injection wound were 0.5 and 1㎝. DBH of chestnut trees and pine trees injected were 10, 15, 20 and 10, 20, 30㎝, respectively. The results of tree response were examined in June, 1994. None of them showed any symptom of decay by the time. However, sapwood under the bark was remarkably discolored. The discoloration was more severe in 1㎝-injection wound than in 0.5㎝ one. The severity of discoloration was not differed between species and among DBH applied. The sapwood split was longer in 1㎝-injection wounded trees than in 0.5㎝ trees and longer in chestnut trees than in pine trees. From the split, callus grew out and almost closed the splits. In the trunk injection tested, the damage was mare severe in 1㎝ injection wound of chestnut trees than in any other combination.

        Desferoxamine 이 시험관내 인체 간암세포 성장에 미치는 영향

        N/A Objectives: It has been known that excessive iron promote the growth of cancer cells aad suggested that iron oversupply in cancer patients may enhance tumor growth and adversely affect cancer therapy and that desferoxamine (DFO) as a iron chelating agent may have a place in anticancer agents, which acts as a potent inhibitor of DNA synthesis via inhibition of the ribonucleotide reductase. However there have been no reports on the combined effect of DFO with other chemotherapeutics in vitro, This study was done to answer the questions: Does DFO inhibit the growth of cultured hepatoma cells? and Does the combination of DFO and chemotherapeutic agents have a synergistic effect on inhibition of tumor growth? Methods: Using PLC/PRF/5 and Hep G2 cells as hepatoma cells and normal diploid cells of WI-38 as control, the MTT assay was performed for evaluation of the cytotoxicity and an isobologram method to analyze the combined effects of DFO and other chemotherapeutic agents. Results: 1) The hepatoma cells grew faster in an iron-enriched medium than in the medium without additional iron supplementation but the normal diploid cells of WI-38 did not. 2) The 50% inhibitory concentrations (IC50) on PLC/ PRF/5 and Hep 62 cells were 75.1 and 29.1 ㎍/ml, respectively. But the growth of WI-38 was not inhibited below 74.4% with even 200 ㎍/ml of DFO. 3) On both hepatoma cells, the growth inhibitory effects of DFO were reversed with simultaneous administration of stoichiometric doses of ferric citrate. 4) Adriamycin (ADR), 5-fluorouracil (5-FU) and cisplatin (DDP) inhibited the growth of all 3 cell lines, but larger doses were required to inhibit the WI-38 to obtain the same effect. 5) When DFO was combined with chemotherapeutic agents, a marked synergistic effect was observed with the combination of DFO and 5-FU on PLC/PRF/5 cells. The synergistic effect was also noted in Hep G2 cells with all three combinations of DFO-ADR, DFO-5-FU and DFO-DDP. Conclusion: Iron oversupply may enhance the hepatoma growth and this should be taken into consideration treating patients with hepatoma. And the antitumor effect of DFO and its synergistic effect with other chemotherapeutic agents may suggest that iron chelator like the DFO may be a useful treatment for patients with hepatoma in conjuction with other anticancer drugs.

        폐포 대식세포 및 단핵구가 Interleukin-2 Enhanced Natural Killer 및 LAK Activity에 미치는 영향

        저자들은 폐포 대식세포 및 말초혈액내의 단핵구가 NK 활성도 및 LAK 활성도에 미치는 영향을 보기위하여, 임파구에 여러 가지 농도(0, 100 : 1, 10 : 1, 1 : 1)의 폐포 대식세포와 단핵구를 넣어 IL-2 enhanced NK 활성도 및 LAK 활성도를 비교하여 다음과 같은 결과를 얻었다. 1) 여러 가지 농도의 단해구는 IL-2 enchanced NK 활성도 및 LAK 활성도에 영향을 미치지 않았다. 2) 동량의 페포대식세포(임파구 : 폐포 대식세포= 1 : 1)는 IL-2 enhanced NK 활성도를 의의있게 억제하였으나(p<0.05), 소량의 폐포대식세포(임파구 : 폐포 대식세포-10 : 1과 100 : 1)는 IL-2 enhanced NK 활성도를 억제하지 못하였다. 3) 임팍와 폐포 대식세포의 비율이 1 : 1과 10 : 1에서는 LAK 활성도를 의의있게 억제하였으나, 소량의 폐포대식세포(임파구 : 폐포 대식세포=100 : 1)는 LAK 활성도를 억제하지 못하였다(p<0.05). 이상의 결과로 IL-2 enhanced NK 활성도 및 LAK 활성도는 폐포 대식세포의 양에 비례하여 억제되었으나, 말초혈액내의 단핵구에 의해서는 영향받지 않는 것을 알 수 있었다. Alveolar macrophages (AM) are thought to function as primary effector cells against tumors growing in the lung. Systemic administration of lymphokine activated killer (LAK) cells and IL-2 resulted in partial antitumor response in patients with advanced cancer. LAK activity is influenced by various factors. We studied the effects of AM and blood monocytes from healthy donors on IL-2 enhanced NK activity against K-562 cells and LAK activity against Raji cells utilizing a 4h ^(51)Cr release assay. The following results were obtained: 1) The addition of different doses of human blood monocytes showed no suppression or enhancement of IL-2 enhanced NK and LAK activity. 2) The addition of high dose of AM (Lymphocyte: AM=1:1) significantly suppressed IL-2 enhanced NK activity. Smaller doses of AM (Lymphocyte: AM= 10:1and 100:1) did not suppress IL-2 enhanced NK activity. 3) The addition of high dose of AM (Lymphocyte: AM = 1:1 and 10:1) significantly suppressed LAK activity. The smallest dose of AM (Lymphocyte: AM= 100:1) did not suppress LAK activity. In conclusion, IL-2 enhanced NK and LAK activity were dose-dependently suppressed by human alveolar macrophages. However IL-2 enhanced NK and LAK activity were not suppressed by blood monocytes.

      • 팽나무버섯의 子實體形成에 미치는 培地의 톱밥 크기와 低溫處理의 영향

        To investigate the effects of the sawdust size and the low temperature treatments on the mycelia in media in the fruit bodies formation of the winter mushroom (Flammulina velutipes), two yellow types (YA1 and YA0) were cultvated. The results were as follows. 1.In the cultivation of mushroom, the media prepared with coarse sawdust (size of 0.85-2.00mm) were more effective in mycelia growth, while less effective in the length of stipes, the number and weight of the fruit bodies formation than the media prepared with fine sawdust (size of 0.05-0.84mm). 2.the media placed at 11˚-15℃ after low temperature treatment of 3˚-4℃ in 7 days to promote the rudiments formation of fruit bodies were more effective in the number of fruit bodies formation but did not effect in another cultural characteristies. 3.YA1 compared with YA0 was reduced in the weight of fruit bodies formation since YA1 had small pileus, thin stipes and short stipes. However, there was no significant difference in the number of fruit bodies formation, the number of days required for mycelial growth and rudiments formation of fruit bodies. 4.In the preparation of sawdust media, it was considered that mixture of different size sawdust were more suitable than uniform size.

      • 팽나무버섯 選拔系統 各 世代의 培養的 特性

        To investigate purity, 6 isolates selected with 2 characteristics in 3 strains of the winter mushroom were conducted subculture for 5 generations. The cultural characteristics were as follows : 1.According to the characteristics such as fruit bodies formation weight, fruit bodies formation number, each fruit body weight, stipes length and pileus diameter, the winter mushroom can be classified into the brown and yellow types. 2.Compare the brown type with the yellow type, the former was smaller fruit bodies formation number, longer stipes, bigger pileus, and more fruit bodies formation weight. 3.There was no significant difference among each generation and among each isolate belong to the each type in the 8 characterstics, Therefore, it is considered that both brown and yellow types were spearated into pure lines. 4.In the correlation among 8 characteristics, there was no significant difference between days of primordia formation and days of fruit bodies production, between days of primordia formation and fruit bodies formation number, however, there was significant difference among the other two characteristics. 5.To estimate fruit bodies formation weight, two or more characteristics were more effective than one characteristic. 6.According to the simple and multiple regression analysis, linear regression equations based on various combination of characteristics to estimate fruit bodies formation weight were suggested for the practical use.

      • 韓國産 팽나무버섯(選拔한 두 栽培品種) 菌絲의 培養的 特性

        In order to investigate cultural characteristics of mycelia in two cultivars of winter mushroom (Flammulina velutipes), the author investigated the adaptability of cultivars to the temperature and H-ion concentration, the utility of carbon soures and nitrogen sources, and the wood- deca-yedness of cultivars in the broad- leaved woods. The results obtained were as follows : 1. In the adaptability of two cultivars to the tempeature, the brown species had the nature of growing in the optimum temperature of 20℃ to 25℃, and yellow species had the optimum tempe-rature of 25℃. And mycelial growth of the yellow species was discontinued at 35℃. 2. In the adaptability of two cultivars to the H- ion concentration, the brown species had optimum range of PH 5.5 to 6.5, and yellow species had optimum range of PH 5.0 to 6.0. 3. In the utility of carbon sources, the brown species utilized favorably starch, and yellow species utilized favorably maltose. 4. In the utility of nitrogen sources, the brown species utilized favorably ammonium phosphate dibasic, and the yellow species utilized favorably asparagine. 5. In the wood - decayedness of cultivars in the broad-leaved woods, remarkably powerful wood-decayedness of the brown species could be found in the diospyros kaki Th□nb. var. domestica, but remarkably powerful wood-decayedness of yellow species could be found in the Zelkowa serata and Celtis koraiensis Nakai var. arguta. 6. As above results, author can now conclude that there are differences in cultural characteristics of mycelia between brown species and yellow species.
