Graphene have been attracted lots of interests for bio-sensing devices because their ultra-thin structure enables strong electrostatic coupling with target molecules and their excellent electrical mobility promise for ultra-fast sensing speed. However, thickly stacked receptors on graphene surface interrupted the electrostatics coupling between graphene and charged biomolecules, which can reduce the sensitivity in graphene biosensor. Here, we report highly sensitive graphene biosensor by monomolecular self–assembly of newly designed peptide protein receptors. The graphene channel was non-covalently functionalized with peptide protein through the π–π interaction along graphene’s Bravais lattice, allowing ultra-thin monomolecular self-assembly through graphene lattice. In the thickness dependent characterization, graphene sensor with monomolecular receptor (thickness less than 1 nm) shown 5 times higher sensitivity and 3 times higher voltage shift than graphene sensor with thick receptor stack (thickness over 20 nm), which is attributed to excellent gate coupling between graphene and streptavidin through ultrathin receptor insulator. Along with fast inherent response time (less than 0.6 s) by extremely high mobility in graphene, our graphene biosensor is promising as a new platform for highly sensitive real-time monitoring of biomolecules with high spatiotemporal resolution.
